Metabolites isolated from Penicillium HDS-Z-1E, an endophytic fungal strain isolated from Taxus cuspidata and their activation effect of catalase

Objective:To study the compounds isolated from Penicillium HDS-Z-1E,an endophytic fungal strain isolated from Taxus cuspidata and their activation effect of catalase(CAT).Methods:The chemical constituents were isolated from Penicillium HDS-Z-1E,by using silica gel,Sephadex LH-20 and HPLC.The structural elucidations of five metabolites were elucidated on the basis of spectroscopic including 1 H-NMR,13C-NMR,HMBC and HSQC.Their activation sites of catalase have been investigated by molecular docking.Results:Five metabolites,compounds(1–5)were isolated from Penicillium HDS-Z-1E and identified as 4-hydroxy-4-methyltetrahydro-2H-pyran-2-one(1),4-hydroxymethyl-5,6-dihydro-pyran-2-one(2),5,6-dihydro-2-oxo-2H-pyran-4-carboxylic(3),N-acetyl-hydrazinobenzoic acid(4),and methyl 2-(2,5-dihydroxyphenyl)acetate(5).Conclusion:Compound 3 is a new compound.Compounds 3 and 4 may have potential activators of catalase,providing a theoretical basis for the development of CAT activators.

Selective immobilization of Pb(Ⅱ)by biogenic whewellite and its mechanism

The development of bio-adsorbents with highly selective immobilization properties for specific heavy metals is a great challenge,but has important application value.Biogenic whewellite(BW)with high selectivity for Pb(Ⅱ)was synthesized by mineral microbial transformation.The selective immobilization properties and mechanism of BW for Pb(Ⅱ)were analyzed by combining mineral characterization technology and batch adsorption research methods.The results indicated that BW can efficiently and selectively immobilize Pb(Ⅱ)in single or composite heavy metal adsorption solutions,and the immobilized Pb(Ⅱ)is difficult to desorb.BW undergoes monolayer adsorption on Pb(Ⅱ),Qmax≈1073.17 mg/g.The immobilization of Pb(Ⅱ)by BW is a physico-chemical adsorption process with spontaneous heat absorption and an accompanying increase in entropy.In addition,the sequestration of Pb(Ⅱ)by BW remains around 756.99 mg/g even at p H=1.The excellent selective immobilization properties of BW for Pb(Ⅱ)are closely related to its smaller Ksp,electrostatic repulsion effect,organic-inorganic composite structure,acid resistance and the formation of Pb(Ⅱ)oxalate.This study provides beneficial information about the recycling of lead in acidic lead-containing wastewater and composite heavy metal contaminated water bodies.

Microtubules play a central role in cytoskeletal changes during neuronal development and maintenance.Microtubule dynamics is essential to polarity and shape transitions underlying neural cell division,differentiation,motility,and maturation.Kinesin superfamily protein 2A is a member of human kinesin 13 gene family of proteins that depolymerize and destabilize microtubules.In dividing cells,kinesin superfamily protein 2A is involved in mitotic progression,spindle assembly,and chromosome segregation.In postmitotic neurons,it is required for axon/dendrite specification and extension,neuronal migration,connectivity,and survival.Humans with kinesin superfamily protein 2A mutations suffer from a variety of malformations of cortical development,epilepsy,autism spectrum disorder,and neurodegeneration.In this review,we discuss how kinesin superfamily protein 2A regulates neuronal development and function,and how its deregulation causes neurodevelopmental and neurological disorders.

What is already known about this topic?Mushroom poisoning poses a significant food safety concern in China,with a total of 196 species identified in poisoning incidents by the end of 2022.What is added by this report?In 2023,the China CDC conducted an investigation into 505 cases of mushroom poisoning spanning 24 provincial-level administrative divisions.This investigation resulted in 1,303 patients and 16 deaths,yielding a case fatality rate of 1.23%.A total of 97 mushrooms were identified as the cause of 6 distinct clinical disease types,with 12 species newly documented as poisonous mushrooms in China.What are the implications for public health practice?Close collaboration among CDC staff,physicians,and mycologists remains crucial for the control and prevention of mushroom poisoning in the future.

Dear Editor,Influenza A viruses(IAVs)are responsible for significant respiratory illnesses in humans and a broad range of animal species.Repeated outbreaks and the rapid spread of genetically and antigenically distinct IAVs represent a considerable challenge for swine production.Pigs are susceptible to avian and human IAV infections and are considered“mixing vessels”where human,avian,and swine IAVs can recombine and generate novel reassortant influenza viruses with pandemic potential(Vincent et al.,2014).

Cadmium(Cd)is a non-essential toxic heavy metal,seriously posing high environmental risks to human health.Digging genetic resources relevant to functional genes is important for understanding the metal absorption and accumulation in crops and bioremediation of Cd-polluted environments.This study investigated a functionally uncharacterized ATP binding cassette transporter G family(ABCG)gene encoding a Pleiotropic Drug Resistance 20(PDR20)type metal transporter which is localized to the plasma membrane of rice.OsPDR20 was transcriptionally expressed in almost all tissues and organs in lifespan and was strongly induced in roots and shoots of young rice under Cd stress.Ectopic expression of OsPDR20 in a yeast mutant ycf1 sensitive to Cd conferred cellular tolerance with less Cd accumulation.Knockdown of OsPDR20 by RNA interference(RNAi)moderately attenuated root/shoot elongation and biomass,with reduced chlorophylls in rice grown under hydroponic medium with 2 and 10μmol/L Cd,but led to more Cd accumulation.A field trial of rice grown in a realistic Cd-contaminated soil(0.40 mg/kg)showed that RNAi plants growth and development were also compromised compared to wild-type(WT),with smaller panicles and lower spikelet fertility but little effect on yield of grains.However,OsPDR20 suppression resulted in unexpectedly higher levels of Cd accumulation in rice straw including lower leaves and culm and grain.These results suggest that OsPDR20 is actively involved in Cd accumulation and homeostasis in rice crops.The increased Cd accumulation in the RNAi plants has the potential application in phytoremediation of Cd-polluted wetland soils.

Recently,the substance P(SP)/neurokinin-1 receptor(NK-1R)system has been found to be involved in various human pathophysiological disorders including the symptoms of coronavirus disease 2019(COVID-19).Besides,studies in the oncological field have demonstrated an intricate correlation between the upregulation of NK-1R and the activation of SP/NK-1R system with the progression of multiple carcinoma types and poor clinical prognosis.These findings indicate that the modulation of SP/NK-1R system with NK-1R antagonists can be a potential broad-spectrum antitumor strategy.This review updates the latest potential and applications of NK-1R antagonists in the treatment of human diseases and cancers,as well as the underlying mechanisms.Furthermore,the strategies to improve the bioavailability and efficacy of NK-1R antagonist drugs are summarized,such as solid dispersion systems,nanonization,and nanoencapsulation.As a radiopharmaceutical therapeutic,the NK-1R antagonist aprepitant was originally developed as radioligand receptor to target NK-1R-overexpressing tumors.However,combining NK-1R antagonists with other drugs can produce a synergistic effect,thereby enhancing the therapeutic effect,alleviating the symptoms,and improving patients’quality of life in several diseases and cancers.

Meeting the continuous glucose monitoring requirements of individuals necessitates the research and development of sensors with high sensitivity and stability.In this study,a straightforward strategy was proposed for synthesizing ultra-thin oxygen-rich graphitized carbon nanosheets（denoted as GCS-O）.These nanosheets are obtained by calcining a topologically two-dimensional indium-based coordination polymer.Subsequently,the growth of FeNi Prussian blue analogue（PBA） on GCS-O effectively introduces active sites and increases the nitrogen content within the carbonaceous matrix.The resulting FeNi-PBA/GCS-O composite exhibits excellent glucose sensing performance with a broad linear range of 1 to 1300μmol·L-1.Meanwhile,it also achieves a high sensitivity of 2496 μA·mmol-1·L·cm-2,a limit of detection of 100nmol·L-1（S/N=3）,and commendable long-term durability.The relatively simple synthesis process,exceptional sensitivity,and satisfactory electrochemical sensing performance of FeNi-PBA/GCS-O open up new directions for biosensor applications.
