CRISPR/CasRx-mediated resistance to Soybean mosaic virus in soybean

Soybean mosaic virus(SMV),an RNA virus,is the most common and destructive pathogenic virus in soybean *** newly developed CRISPR/Cas immune system has provided a novel strategy for improving plant resistance to viruses;hence,this study aimed to engineer SMV resistance in soybean using this ***,multiple sgRNAs were designed to target positive-and/or negative-sense strands of the SMV HC-Pro ***,the corresponding CRISPR/CasRx vectors were constructed and transformed into *** inoculation with SMV,39.02%,35.77%,and 18.70%of T_(1)plants were confirmed to be highly resistant(HR),resistant(R),and mildly resistant(MR)to SMV,respectively,whereas only 6.50%were identified as susceptible(S).Additionally,qRT-PCR and DAS-ELISA showed that,both at 15 and 30 d post-inoculation(dpi),SMV accumulation significantly decreased or was even undetectable in HR and R plants,followed by MR and S ***,the expression level of the CasRx gene varied in almost all T_(1)plants with different resistance level,both at 15 and 30 ***,when SMV resistance was evaluated in the T_(2)generation,the results were similar to those recorded for the T_(1)*** findings provide new insights into the application of the CRISPR/CasRx system for soybean improvement and offer a promising alternative strategy for breeding for resistance to biotic stress that will contribute to the development of SMV-immune soybean germplasm to accelerate progress towards greater soybean crop productivity.

Anti-infective immune functions of type Ⅳ interferon in grass carp(Ctenopharyngodon idella):A novel antibacterial and antiviral interferon in lower vertebrates

Type Ⅳ interferon(IFN-υ)is a recently discovered cytokine crucial for host defense against viral ***,the role and mechanisms of IFN-υin bacterial infections remain *** study investigated the antibacterial and antiviral functions and mechanisms of grass carp(Ctenopharyngodon idella)IFN-υ(CiIFN-υ)both in vivo and in *** CiIFN-υgene was first identified and characterized in grass ***,the immune expression of CiIFN-υsignificantly increased following bacterial challenge,indicating its response to bacterial *** eukaryotic recombinant expression plasmid of CiIFN-υwas then constructed and transfected into fathead minnow(FHM)*** were collected and incubated with four bacterial strains,followed by plate spreading and colony *** indicated that CiIFN-υexhibited more potent antibacterial activity against gram-negative bacteria compared to gram-positive bacteria and aggregated gram-negative bacteria but not gram-positive *** vivo experiments further confirmed the antibacterial function,showing high survival rates,low tissue edema and damage,reduced tissue bacterial load,and elevated proinflammatory response at the early stages of bacterial *** addition,the antiviral function of CiIFN-υwas confirmed through in vitro and in vivo experiments,including crystal violet staining,survival rates,tissue viral burden,and reverse transcription-quantitative real-time polymerase chain reaction(RT-qPCR).This study highlights the antibacterial function and preliminary mechanism of IFN-υ,demonstrating that IFN-υpossesses dual functions against bacterial and viral infections.

Integrins and their potential roles in mammalian pregnancy

Integrins are a highly complex family of receptors that, when expressed on the surface of cells, can mediate reciprocal cell-to-cell and cell-to-extracellular matrix(ECM) interactions leading to assembly of integrin adhesion complexes(IACs) that initiate many signaling functions both at the membrane and deeper within the cytoplasm to coordinate processes including cell adhesion, migration, proliferation, survival, differentiation, and metabolism. All metazoan organisms possess integrins, and it is generally agreed that integrins were associated with the evolution of multicellularity, being essential for the association of cells with their neighbors and surroundings, during embryonic development and many aspects of cellular and molecular biology. Integrins have important roles in many aspects of embryonic development, normal physiology, and disease processes with a multitude of functions discovered and elucidated for integrins that directly influence many areas of biology and medicine, including mammalian pregnancy, in particular implantation of the blastocyst to the uterine wall, subsequent placentation and conceptus(embryo/fetus and associated placental membranes) development. This review provides a succinct overview of integrin structure, ligand binding, and signaling followed with a concise overview of embryonic development, implantation, and early placentation in pigs, sheep, humans, and mice as an example for rodents. A brief timeline of the initial localization of integrin subunits to the uterine luminal epithelium(LE) and conceptus trophoblast is then presented, followed by sequential summaries of integrin expression and function during gestation in pigs, sheep, humans, and rodents. As appropriate for this journal, summaries of integrin expression and function during gestation in pigs and sheep are in depth, whereas summaries for humans and rodents are brief. Modulation of gut microbiota and serum metabolome by Apostichopus japonicus derived oligopeptide in high-fructose diet-induced hyperuricemia in mice

Fructose consumption has risen dramatically in recent decades due to the use of sucrose and high fructose corn syrup in beverages and processed foods， contributing to rising rates of hyperuricemia. The purpose of this experiment was to explore the anti-hyperuricemia effects of an active oligopeptide (GPSGRP) derived from sea cucumber in fructose induced hyperuricemia mouse model， and to clarify the underlying mechanism in sight of gut microbiota and serum metabolites. Peptide GPSGRP treatment rebalanced uric acid metabolism and alleviated inflammatory response in mice. In addition， treatment with GPSGRP decreased the abundance of Bacteroides and Proteobacteria at the phylum level， Muribaculum， Prevotella and Bacteroides at the genus level， and inhibited the related pathways of purine metabolism and glycolysis/gluconeogenesis metabolism. Moreover， serum metabolites， including linoleic acid， indole and its derivatives， arachidonic acid and uridine， as well as related metabolic pathways， such as tricarboxylic acid cycle， ketone production and sugar production， were altered in response to GPSGRP treatment. This study provides a valuable reference for the application and development of marine biological peptides in uric acid management.

Mucosal immune responses to Ichthyophthirius multifiliis in the ocular mucosa of rainbow trout(Oncorhynchus mykiss,Walbaum),an ancient teleost fish

The eye,as a specialized visual organ,is directly exposed to the external environment,and,therefore,it faces constant chal-lenges from external pathogenic organisms and *** the ocular mucosa(OM)of mammals,mucosal-associated lymphoid tissues(MALTs)constitute the primary line of ***,the immune defense role of the OM remains unknown in aquatic *** gain insights into the immune processes within the OM of teleost fish,we developed an infection model of rainbow trout(Oncorhynchus mykiss)OM using a parasite,Ichthyophthirius multifiliis(Ich).Immunofluorescence,qPCR,and H&E staining revealed that Ich successfully infiltrates the OM of rainbow trout,leading to pathological struc-tural changes,as evidenced by A&B ***,the qPCR results indicate an up-regulation of immune-related genes following Ich infection in the ***,transcriptome analyses were conducted to detect immune responses and impairments in eye function within the OM of rainbow trout with Ich *** results of the transcriptome analysis that Ich infection can cause an extensive immune response in the OM,ultimately affecting ocular *** the best of our knowledge,our findings represent for the first time that the teleost OM could act as an invasion site for parasites and trigger a strong mucosal immune response to parasitic infection.

Optimization of inter-seasonal nitrogen allocation increases yield and resource-use efficiency in a water-limited wheat-maize cropping system in the North China Plain

Winter wheat–summer maize cropping system in the North China Plain often experiences droughtinduced yield reduction in the wheat season and rainwater and nitrogen(N)fertilizer losses in the maize *** study aimed to identify an optimal interseasonal water-and N-management strategy to alleviate these *** ratios of allocation of 360 kg N ha^(-1)between the wheat and maize seasons under one-time presowing root-zone irrigation(W0)and additional jointing and anthesis irrigation(W2)in wheat and one irrigation after maize sowing were set as follows:N1(120:240),N2(180:180),N3(240:120)and N4(300:60).The results showed that under W0,the N3 treatment produced the highest annual yield,crop water productivity(WPC),and nitrogen partial factor productivity(PFPN).Increased N allocation in wheat under W0 improved wheat yield without affecting maize yield,as surplus nitrate after wheat harvest was retained in the topsoil layers and available for the subsequent *** W2,annual yield was largest in the N2 *** risk of nitrate leaching increased in W2 when N application rate in wheat exceeded that of the N2 treatment,especially in the wet *** to W2N2,the W0N3 maintained 95.2%grain yield over two *** WPCwas higher in the W0 treatment than in the W2 ***,following limited total N rate,an appropriate fertilizer N transfer from maize to wheat season had the potential of a“triple win”for high annual yield,WPCand PFPN in a water-limited wheat–maize cropping system.

Thymus is the important immune organ,responsible for T cell development and *** lower circulating T counts have been observed in patients who died from COVID-19 compared with ***,also known as FNC,is a thymus-homing anti-SARS-CoV-2 drug in treating COVID-19 *** this study,single-cell transcriptome,proteomics,and parallel reaction monitoring(PRM)were applied to insight into the activation process of FNC in rat and SARS-CoV-2 rhesus monkey *** results indicated that thymic immune cells possess a robust metabolic capacity for cytidine-analogue drugs such as *** enzymes involved in the FNC phosphorylation process,such as Dck,Cmpk1,and Nme2,were highly expressed in CD4+ T cells,CD8+ T cells,and DP(CD4+ CD8+)***,FNC could upregulate multiple phosphorylated kinases in various cell types while downregulating the phosphatases,phosphoribosyl transferases,and deaminases,*** robust phosphorylation capacity of the thymus for cytidine analogue drug FNC,and the activation effect of FNC on the NAs metabolism system potentially contribute to its enrichment in the thymus and immune protection *** suggests that it is crucial to consider the expression level of phosphorylation kinases when evaluating NA drug properties,as an important factor during antiviral drug design.

Minimally invasive surgery(MIS)has significantly progressed and emerged as the preferred approach for various gastrointestinal procedures,including gastrectomy,cholecystectomy,and various colorectal surgeries(1).However,the widespread acceptance of robotic surgery in liver resection has encountered obstacles,attributed to factors such as the lack of appropriate surgical instruments,the intricate learning curve,and safety ***,the utilization of MIS in liver resection is linked to decreased blood loss,reduced post-operative pain,shorter hospital stays,and lower morbidity rates compared to both open and laparoscopic liver resection procedures(2).
