Effect of different twirling and rotating acupuncture manipulation techniques on the blood flow perfusion at acupoints

OBJECTIVE: To analyze the effect of different twirling and rotating acupuncture manipulation techniques on the blood flow perfusion at acupoints to provide a reference for the study of acupoint specificity and the quantification and effectiveness of acupuncture methods. METHODS: Twenty healthy male or female college students each received four different acupuncture manipulation techniques, including simple acupuncture, uniform reinforcing-reducing needling, twirling reinforcing needling, and twisting reducing needling. The self-control method was applied. acupuncture was performed by an acupuncture manipulation simulator at Neiguan (PC 6) and Zusanli (ST 36). The process of twirling and rotating was divided into seven timepoints. The PeriCam Perfusion Speckle Imager (PSI) System was used to collect the blood flow perfusion data at each acupoint. The specificity of twirling and rotating acupuncture manipulation was analyzed based on changes in the curve, video, and numerical blood flow perfusion data at each timepoint. RESULTS: There were two peaks in the blood flow perfusion curve of twirling and rotating acupuncture manipulation;one appeared after 3 min of needle retention, and the other appeared 3 min after needle removal. The blood flow perfusion parameters showed that the greatest differences between the four manipulation techniques occurred after 5 and 10 min of needle retention. The specificity of various manipulation techniques was most obvious at these two timepoints. There were significant differences between the four manipulation groups in the blood flow perfusion and the relative change rates of blood flow perfusion at each timepoint. CONCLUSION: Laser speckle imaging enables the real-time, non-invasive, rapid, and accurate collection of blood flow perfusion data during acupuncture. This imaging technique enables the easy attainment of various parameters such as visual images, two-dimensional curves, and data tables. At various timepoints, the four groups

Objective: To observe the influence of acupuncture on microcirculation perfusion of the pericardium meridian and heart in acute myocardial ischemia(AMI) rats and evaluate whether acupuncture can simultaneously affect the meridians and corresponding viscera. Additionally, acupoints at different meridians were compared and whether they exert the same effects was discussed. Methods: Totally 32 Sprague-Dawley rats were subjected to left anterior descending(LAD) ligation to develop an AMI *** were divided into 4 groups, including AMI, acupuncture Neiguan(PC 6), Lieque(LU 7) and Qiansanli(LI 10) groups(n=8). Eight rats received only thoracotomy(sham-operated group). The rats in the acupuncture groups received manual acupuncture at PC 6, LU 7 and LI 10 acupoints for 15 min, respectively. The microcirculation perfusion of pericardium meridian and heart was monitored by laser speckle perfusion imager(LSPI) before, during and after acupuncture manipulation for 15 min. Subsequently, the perfusion unit(PU)was calculated and analyzed by PSI System. Results: After LAD, compared to pre-acupuncture stage, the heart microcirculation perfusion(HMP) in the AMI group decreased continuously at during-acupuncture(P>0.05) and post-acupuncture stages(P0.05). Compared to pre-acupuncture stage, the PMP and HMP in PC 6 group significantly increased during acupuncture manipulation(both Pacupuncture(P0.05);however, they were significantly reduced after acupuncture manipulation(both Pacupuncture, especially compared to pre-acupuncture stage(Pacupuncture at PC 6 obviously increased the PMP and HMP in AMI rats, and the effects were superior to at LU 7 and LI 10 acup

Objective: To evaluate the therapeutic effects of needle-rolling therapy for chronic insomnia. Methods: In the present multi-central randomly controlled clinical study, 180 cases of chronic insomnia were randomly divided into the following two groups, a treatment group (90 cases) treated by the needle-rolling therapy and a control group (90 cases) treated with clonopin. The treatment course for both the two groups was 4 weeks. The therapeutic effects were evaluated based on improvement of the TCM symptoms and the Pittsburgs's sleep-quality index (PSQI). Results: After treatment, there were significant differences between the two groups in the effective rate (P<0.05), and in the total score of PSQI and in the scores of the 4 sub-items, i.e. sleep-quality, sleep-efficiency, hypnotic and daytime function (P<0.05). Although there was no significant difference between the two groups in the effective rate after a 3-month follow-up period, significant differences still existed in the 3 sub-items of sleep-efficiency, hypnotic, and daytime function of the PSQI (P<0.05). Conclusion: As compared with hypnotics of the second generation, the needle-rolling therapy may show better therapeutic effects for chronic insomnia patients.

OBJECTIVE: To examine the effect of electroacupuncture(EA) at bilateral Neiguan(PC 6) on voltage-gated Na+currents(INa) and channels(Nav) in ischemic ventricular ***: EA serum was prepared from six male adult Sprague-Dawley rats that had received EA at bilateral Neiguan(PC 6). Eighteen ventricular myocytes were prepared from six SD rats using an enzymolysis approach. Myocardial ischemia was mimicked by perfusion of ischemic solution. Whole-cell patch-clamping was used to record three currents evoked from isolated cells. The first current was the control, and recorded in absence of ischemic solution current. The second was the ischemic current,and recorded after perfusion of ischemic solution for 5 min, while the EA current was last, and recorded after perfusion of EA serum for 5 min. Navkinetic curves were fitted using related ***: Compared with those in controls, in the presence of ischemic solution, peak amplitudes of INasignificantly increased from ﹣40 m V to +30 m V,and half-maximal inactivation potentials of Navincreased significantly, while half-maximal activation potentials, slope factors and the recovery time from inactivation to activation of Navwere unchanged. Compared with those in the ischemic solution, in the presence of EA serum, peak ischemic current amplitudes significantly reduced from ﹣40m V to + 40 m V, and half-maximal inactivation potentials were restored, while half-maximal activation potentials, slope factors and the recovery time from inactivation to activation of Navwere ***: EA at bilateral Neiguan(PC 6) can reduce enhanced INavia restoration of delayed Navinactivation in ischemic ventricular myocytes.

Objective:To observe the effect of acupuncture on local skin tissue metabolites of Tianshu(ST25)in rats with Crohn's disease(CD)using metabolomics and to explore the mechanism of acupuncture treatment metabolites and pathway on local skin tissue of Tianshu(ST25).Materials and Methods:Thirty-eight Sprague-Dawley rats were randomly assigned to a normal group(n=14)and a CD modeling group(n=24).Rats in the CD modeling group were administered with 2,4,6-trinitrobenzene sulfonic acid and ethanol enema,in addition to the normal *** normal rats and four CD modeling rats were selected at random after model identification was ***,CD modeling group rats were randomly assigned to two groups of 10 rats:the model group and acupuncture *** in the acupuncture group underwent acupuncture of Tianshu(ST25)and Shangjuxu(ST37)for 7 *** the intervention,the colon tissue was collected from each group of rats and the pathological changes were observed through hematoxylin and eosin(HE)*** local skin tissues at Tianshu(ST25)of the rats in each group were taken,and the proton nuclear magnetic resonance technique was used to detect the metabonomics of the local skin tissues of Tianshu(ST25)in ***:Compared with the normal group,the following were observed in the model group after HE staining:irregular colon morphology and other pathological changes such as intestinal mucosal hyperemia,edema,ulcers,polyps,and inflammatory cell *** the acupuncture group,colon tissue structure was relatively complete and layered,the colon gland structure was restored,and inflammatory cell infiltration was significantly *** choline,glycerin,glycine,guanidoacetic acid,and proline levels were significantly higher in the model group than in the normal group(Pacupuncture group had significantly increased contents of alanine,leucine,L-phenylalanine,and tyrosine in the skin(P<0.05 or P<0.01).
