Electroacupuncture inhibits hippocampal oxidative stress and autophagy in sleep-deprived rats through the protein kinase B and mechanistic target of rapamycin signaling pathway

Objective: To investigate the effects of acupuncture on learning and memory impairment， oxidative stress and autophagy induced by sleep deprivation in rats， and to analyze the related mechanism. Methods: Thirty Wistar rats were randomly divided into a normal group， sleep deprivation group and acupuncture group. The rat model of sleep deprivation was established by a modified multiplatform sleep deprivation method. The Baihui (GV20)， Shenmen (HT7) and Sanyinjiao (SP6) acupoints of rats were located to give electroacupuncture (density wave， frequency 20 Hz， intensity 1 mA) to maintain the needle feeling， and to keep the needle for 15 min and continuous acupuncture for 7 d. The spatial learning and memory abilities of the rats were detected by the water maze test. The content of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GPX) in the brain were detected by an assay kit， and the autophagy related proteins light chain 3 alpha (LC3A)， light chain 3 beta (LC3B) and Beclin 1 and the activation of the protein kinase B (PKB/AKT) and mechanistic target of rapamycin (mTOR) signaling pathway in the rat’s brain were detected by Western blotting. Results: Compared with the normal group， the time spent in the target quadrant (P < 0.05) and the number of times entering the target quadrant (P < 0.05) in the rats of sleep deprivation group were significantly reduced， and the content of MDA was significantly increased (P < 0.01)， while the activities of SOD and GPX (P < 0.01) in the brain were significantly decreased， and LC3A II/I， LC3B II/I and Beclin 1 increased significantly (P < 0.01)， while p-AKT (ser473)/AKT， p-mTOR (ser2448)/mTOR and p-p70s6K (thr389)/p70S6 decreased significantly (P < 0.01). Enterogenic Stenotrophomonas maltophilia migrates to the mammary gland to induce mastitis by activating the calcium-ROS-AMPK-mTOR-autophagy pathway

Background Mastitis is an inflammatory disease of the mammary gland that has serious economic impacts on the dairy industry and endangers food *** previous study found that the body has a gut/rumen-mammary gland axis and that disturbance of the gut/rumen microbiota could result in‘gastroenterogenic mastitis'.However,the mechanism has not been fully ***,we found that long-term feeding of a high-concentrate diet induced mastitis in dairy cows,and the abundance of Stenotrophomonas maltophilia(***)was significantly increased in both the rumen and milk ***,we hypothesized that‘gastroenterogenic mastitis'can be induced by the migration of endogenous gut bacteria to the mammary ***,this study investigated the mechanism by which enterogenic *** induces *** First,*** was labelled with superfolder GFP and administered to mice via *** results showed that treatment with *** promoted the occurrence of mastitis and increased the permeability of the blood-milk barrier,leading to intestinal inflammation and intestinal ***,tracking of ingested *** revealed that *** could migrate from the gut to the mammary gland and induce ***,mammary gland transcriptome analysis showed that the calcium and AMPK signalling pathways were significantly upregulated in mice treated with ***,using mouse mammary epithelial cells(MMECs),we verified that *** induces mastitis through activation of the calcium-ROS-AMPK-mTOR-autophagy *** In conclusion,the results showed that enterogenic *** could migrate from the gut to the mammary gland via the gut-mammary axis and activate the calcium-ROS-AMPK-mTOR-autophagy pathway to induce *** the gut-mammary gland axis may also be an effective method to treat mastitis.

Evaluation Indicators of Traditional chinese medicine Syndromes for Gouty Arthritis with Damp Heat Accumulation and the Effect of Administering Tongfeng Qingxiao formula(痛风清消方)

OBJECTIVE: To evaluate the indicators of an animal model of gouty arthritis (GA) with dampness heat accumulation and the intervention effect of Tongfeng Qingxiao formula (痛风清消方， TFQXF).METHODS: Seventy-two healthy adult Sprague-Dawley male rats were selected and randomly divided into a normal group， model group， low-dose group， medium-dose group， high-dose group， and diclofenac group using a random number table method， with 12 rats in each group. After group intervention， the general condition of the rats in each group was monitored and recorded， and the swelling index was measured. After separating the serum， the changes in glutamic pyruvic transaminase (ALT)， glutamic oxaloacetic transaminase (AST)， carbamide (UREA)， creatinine (CREA)， triglyceride (TG)， total serum cholesterol (TC)， low density lipoprotein cholesterol (LDL-C)， and high density lipoprotein cholesterol (HDL-C) in the serum of the rats in each group were measured using an automatic biochemical analyzer. The levels of motilin (MTL)， gastrin (GAS)， endothelin (ET)， calcitonin gene-related peptide (CGRP)， heat shock protein 70 (HSP70)， interleukin (IL)-1β， and nuclear factor kappa-B (NF-κB) in the serum of the rats in each group were evaluated using enzyme-linked immunosorbent assay (ELISA) kits. Kidney tissues were used to evaluate the protein and mRNA expression of aquaporin (AQP)1 and AQP2. Colon tissue was used to evaluate the protein and mRNA expression of AQP3 and AQP4 by Western blotting (WB) assay and real-time quantitative polymerase chain reaction (RT-qPCR). The levels of ALT， AST， UREA， and CREA were used to evaluate the liver and kidney function of rats. The levels of MTL and GAS were used to evaluate the gastrointestinal function of rats. The levels of TG， TC， LDL-C， HDL-C， AQP1， AQP2， AQP3， and AQP4 were used to evaluate the "dampness" syndrome performance in rats. Isorhamnetin Downregulates MMP2 and MMP9 to Inhibit Development of Rheumatoid Arthritis through SRC/ERK/CREB Pathway
