Downregulation of Astrocyte Elevated Gene-1 Expression Combined with All-Trans Retinoic Acid Inhibits Development of Vasculogenic Mimicry and Angiogenesis in Glioma

Objective This study aimed to investigate the effects of downregulating astrocyte elevated gene-1(AEG-1)expression combined with all-trans retinoic acid(ATRA)on vasculogenic mimicry(VM)formation and angiogenesis in *** U87 glioma cells were transfected with AEG-1 shRNA lentiviral vectors(U87-siAEG-1)and incubated in a medium containing 20µmol/L ***-based tube formation assay was performed to evaluate VM formation,and the cell counting kit-8(CCK-8)assay was used to analyze the proliferation of glioma cells in *** transcription-quantitative polymerase chain reaction and Western blot analysis were used to investigate the mRNA and protein expression of related genes,*** xenograft models were generated via subcutaneous implantation of glioma cells in nude ***-bearing mice received an intraperitoneal injection of ATRA(10 mg/kg per day).Immunohistochemistry was used to evaluate the expression of related genes and the microvessel density(MVD)in glioma xenograft ***34/periodic acid-Schiff double staining was performed to detect VM channels in *** volume and weight of tumors were measured,and a tumor growth curve was drawn to evaluate tumor *** A combination of ATRA intervention and downregulation of AEG-1 expression significantly inhibited the proliferation of glioma cells in vitro and glioma VM formation in vitro and in *** also significantly decreased MVD and inhibited tumor ***,the expression levels of matrix metalloproteinase(MMP)-2,MMP-9,vascular endothelial-cadherin(VE-cadherin),and vascular endothelial growth factor(VEGF)in glioma significantly decreased in vivo and in *** Hence,a combinatorial approach might be effective in treating glioma through regulating MMP-2,MMP-9,VEGF,and VE-cadherin expression.

Pediatric femoral shaft fractures treated by flexible intrameduUary nailing

Background： Nowadays pediatric femoral fractures are more commonly managed with operative treatment rather than conservative treatment because of more rapid recovery and avoidance of prolonged immobilization. Children between the ages of 5-13 years are treated either by traction plus hip spica and flexible/elastic stable retrograde intramedullary nail, or external fixators in the case of open fractures. The aim of this study is to evaluate the outcome of pediatric femoral shaft fractures treated by stainless steel flexible intramedullary nail in children between 5 and 13 years of age. Methods： There were 32 cases of femoral shaft fractures which were all fixed with stainless steel flexible intramedullary nail under fluoroscopy. Long leg cast was applied at the time of fixation. Partial weight bearing was started 2 weeks after surgery. Patients were evaluated in follow-up study to observe the alignment of fracture, infection, delayed union, nonunion, limb length discrepancy, motion of knee joint, and time to unite the fracture. Results： We were able to follow up 28 out of 32 patients. The patients were 8.14 years of age on average. The mean hospital stay after operation was 4 days and fracture union time was 9.57 weeks. There were 3 cases of varus angulation, 2 cases of anterior angulation, and 4 cases of limb lengthening. Conclusion： Patients aged between 5 and 13 years treated with flexible intramedullary nail for closed femoral shaft fracture have rapid union and recovery, short rehabilitation period, less immobilization and psychological impact, and cost-effective.

Aim: To investigate depression, anxiety, PTSD, and insomnia levels among Sudanese citizens after a year has passed since the start of Armed Forces conflict in Sudan. Methods: An online survey was distributed. It is composed of five parts, covering the following areas: 1) sociodemographic data;2) depression assessment;3) generalized anxiety disorder assessment;4) post-traumatic stress disorder assessment;and 5) insomnia assessment. Statistical Package for Social Sciences version 27 was used for data analysis;frequency and percentage were used to describe the qualitative variables. Spearman’s correlation analysis and Chi-square test were used for correlation and association analysis;a P-value equal to or less than 0.05 was considered statistically significant. Results: The study included 283 participants, primarily female (76.0%), with a mean age of 35.26 ± 6.96 years. The majority of participants were married (50.5%) and had a university level of education (50.2%). At the start of the war, 70.3% of participants were inside the war zones, and at the time of data collection, 73.5% were outside Sudan. The length of stay in the war zone ranged from 1 to 300 days, with a mean of 48.59 ± 70.284 days. Additionally, 63.3% of participants stated that they did not receive any form of mental health intervention, and the remaining did not provide an answer. There was a high prevalence of moderate-severe depression (50.5%), moderate anxiety (35.3%), PTSD (56.5%), and sub-threshold insomnia (53.4%) among the participants. Correlation analyses revealed associations between demographic factors such as sex, age, marital status, and education and mental health issues. Notably, female participants showed a higher prevalence of PTSD (53.0%) compared to male participants (67.6%). Conclusions: There is a substantial impact of war-related trauma on mental health, revealing a high prevalence of moderate-severe depression, moderate anxiety, PTSD, and sub-threshold insomnia among the particip

Contour changes in human alveolar bone following tooth extraction of the maxillary central incisor

The purpose of this study was to apply cone-beam computed tomography （CBCT） to observe contour changes in human alveolar bone after tooth extraction of the maxillary central incisor and to provide original mor- phological evidence for aesthetic implant treatment in the maxillary anterior area. Forty patients were recruited into the study. Each patient had two CBCT scans （CBCT I and CBCT II）, one taken before and one taken three months after tooth extraction of maxillary central incisor （test tooth T）. A fixed anatomic reference point was used to orient the starting axial slice of the two scans. On three CBCT I axial slices, which represented the deep, middle, and shallow layers of the socket, labial and palatal alveolar bone widths of T were measured. The number of sagittal slices from the start point to the pulp centre of T was recorded. On three CBCT II axial slices, the pulp centres of extracted T were oriented according to the number of moved sagittal slices recorded in CBCT I. Labial and palatal alveolar bone widths at the oriented sites were measured. On the CBCT I axial slice which represented the middle layer of the socket, sagittal slices were reconstructed. Relevant distances of T on the sagittal slice were measured, as were the alveolar bone width and tooth length of the opposite central incisor. On the CBCT II axial slice, which represented the middle layer of the socket, relevant distances recorded in CBCT I were transferred on the sagittal slice. The height reduction of alveolar bone on labial and palatal sides was measured, as were the alveolar bone width and tooth length of the opposite central incisor at the oriented site. Intraobserver reliability assessed by intraclass correlation coefficients （ICCs） was high. Paired sample t-tests were performed. The uniaxial compressive response of steel-polypropylene hybrid fiber reinforced concrete（HFRC） and steel fiber-reinforced concrete（SFRC） was analyzed under high strain rate loading with a 74 mm diameter split Hopkinson pressure bar（SHPB）.The experimental investigation focused on recorded data and resulted in distinguishing the strain rate that mobilized different ductility of steel-polypropylene hybrid fiber reinforced concrete（HFRC） and steel fiber-reinforced concrete（SFRC）.64 specimens of HFRC and SFRC with higher static compressive strength were tested at the strain rates changing from 20 to 120 *** static compressive strength and dynamic stress-strain curves of the two materials were obtained at 4 different strain rates and the failure stress,and peak strain and peak toughness were also *** results show that HFRC has quite good dynamic mechanical property and clear strain-rate effect,and the failure mechanism of HFRC and SFRC was also compared based on the specimens＇ failure modes in static and dynamic compressive tests.

Identification of Serum Exosomal MicroRNA Expression Profiling in Menopausal Females with Osteoporosis by High-throughput Sequencing

Estrogen deficiency,which mainly occurs in postmenopausal women,is a primary reason for osteoporosis in clinical ***,the molecular regulation of osteoporosis in menopausal females is still not adequately explained in the literature,with the diagnosis and treatment for osteoporosis being ***,exosomal microRNAs(miRNAs)were used to evaluate their diagnosis and prediction effects in menopausal females with *** this study,6 menopausal females without osteoporosis and 12 menopausal females with osteoporosis were *** serum exosomes were isolated,and the miRNA expression was detected by miRNA high-throughput *** miRNA effects were analyzed by Gene Ontology and Kyoto Encyclopedia of Genes and Genomes *** miRNA-targeted genes were evaluated by Targetscan 7.2 and the protein-protein interactions(PPI)by *** genes were analyzed by the CytoHubba app of *** results showed that 191 aberrant miRNAs were found in the group of menopausal females with osteoporosis,including 72 upregulated miRNAs and 121 downregulated *** miRNAs were involved in many signaling pathways,such as the Wnt,MAPK,and Hippo *** on PPI network analysis,FBXL3,FBXL13,COPS2,UBE2D3,DCUN1D1,DCUN1D4,CUL3,FBXO22,ASB6,and COMMD2 were the 10 most notable genes in the PPI *** conclusion,aberrant serum exosomal miRNAs were associated with an altered risk of osteoporosis in menopausal females and may act as potential biomarkers for the prediction of risk of osteoporosis in menopausal females.

Group Decision-Making Model of Renal Cancer Surgery Options Using Entropy Fuzzy Element Aczel-Alsina Weighted Aggregation Operators under the Environment of Fuzzy Multi-Sets

Since existing selection methods of surgical treatment schemes of renal cancer patients mainly depend on physicians’clinical experience and judgments,the surgical treatment options of renal cancer patients lack their scientifical and reasonable information expression and group decision-making model for renal cancer *** multi-sets(FMSs)have a number of properties,which make them suitable for expressing the uncertain information of medical diagnoses and treatments in group decision-making(GDM)*** choose the most appropriate surgical treatment scheme for a patient with localized renal cell carcinoma(RCC)(T1 stage kidney tumor),this article needs to develop an effective GDM model based on the fuzzy multivalued evaluation information of the renal cancer ***,we propose a conversionmethod of transforming FMSs into entropy fuzzy sets(EFSs)based on the mean and Shannon entropy of a fuzzy sequence in FMS to reasonably simplify the information expression and operations of FMSs and define the score function of an entropy fuzzy element(EFE)for ranking ***,we present the Aczel-Alsina t-norm and t-conorm operations of EFEs and the EFE Aczel-Alsina weighted arithmetic averaging(EFEAAWAA)and EFE Aczel-Alsina weighted geometric averaging(EFEAAWGA)***,we develop a multicriteria GDM model of renal cancer surgery options in the setting of ***,the proposed GDM model is applied to two clinical cases of renal cancer patients to choose the best surgical treatment scheme for a renal cancer patient in the setting of *** selected results of two clinical cases verify the efficiency and rationality of the proposed GDM model in the setting of FMSs.
