In order to explore the changes in the growth and protein contents of Spirulina and obtain a proper strain for the fixation of carbon dioxide(CO2 from flue gas,the strains isolated from the Spirulina farms and the strain 208 were cultured under different aeration conditions including no CO2,10%CO2 and coal power plant flue gas *** physiological indexes including filament length,biomass yield and chlorophyll a,soluble protein and phycocyanin contents were determined,*** cultured without CO2 supplement,the strain 4-5 exhibited the highest biomass yield(1.880 g L^(−1)and a specific growth rate(0.367 d−***,the specific growth rate of all strains decreased significantly when they were cultured under 10%CO2 and unfiltered coal power plant flue gas *** differences were noted in the performance of the experimental microalgal strains under different contemporaneous *** strain 7-8 achieved the highest biomass yield(1.603 g L^(−1)and relatively high phycocyanin content(7.1%)under 10%CO2 *** noted that strain 4-5 had the highest specific growth rate(0.182 d−1 and biomass yield(0.43 g L^(−1)under coal power plant flue gas *** 6-10 displayed the highest soluble protein content(66.02%),and strain 7-8 showed the highest phycocyanin content(9.28%)under coal power plant flue gas supplement.

The dried shellfish products with rich nutrients and low-calorie content are favorite food in china,especially in coastal ***,the species of dried shellfish products in the market are usually unknown,as the taxonomic features were removed during the production *** study described the application of DNA barcoding technique to the identification of 100 dried shellfish(scallop,squid,octopus and cuttlefish)products in *** were authenticated by comparing mitochondrial cytochrome oxidase subunit I(COI)gene and 16S ribosomal RNA(16S rRNA)gene sequences with public reference taxonomic *** results showed that all the 100 products can be identified at species *** four scallop adductor products were processed using the bay scallop,Argopecten irradians,and one was from Portuguese oyster,Crassostrea *** the 27 squid,2 cuttlefish and 6 octopus products were produced by the Jumbo flying squid,Dosidicus *** neighbour-joining tree is in agreement with the results of DNA barcoding *** 64 scallop samples formed one *** cluster,leaving Sca65 clustered with the reference oyster sequence ***(MH997922).All the 35 cephalopod(squid,octopus and cuttlefish)samples formed a *** *** investigation revealed a low variety of dried shellfish products sold on the market,and highlighted the high rate of mislabeling and species *** present work provides a practical method for tracing and authenticating shellfish products.

Sea cucumber(Apostichopus japonicus)is an excellent model for investigating effects of bottom-dwellers on carbon mig-ration and ***,the molecular mechanism of respiratory metabolism process variation caused by feeding rations is poorly *** this study,treatment groups set as 1%(about 0.63g),3%,and 7%of total body weight(named F1,F3 and F7 groups respectively).The potential molecular mechanisms behind the functions of respiratory tree and body wall were investigated by RNA-Seq.A total of 52411 expressed genes were identified from 89342 expressed *** results showed 759,254 and 334 genes were up-regulated,and 334,445 and 992 genes were down-regulated in respiratory tree of F1 vs.F3,F1 vs.F7 and F3 vs.F7,***,2070,1601 and 896 genes were up-regulated,and 1303,1337 and 1144 genes were down-regulated in body wall between F1 vs.F3,F1 vs.F7 and F3 vs.F7,*** expressed genes were enriched in salivary secretion and ECM-receptor interaction pathways in respiratory tree,and in various types of N-glycan biosynthesis,ribosome and sphingolipid metabolism pathways in body *** results suggested respiratory tree and body wall were involved in activation of respiratory metabolisms in response to different feeding *** research provided valuable knowledge for physiological differences in res-piratory metabolism.

There is an increasing demand for salmonid authentication due to the globalization of the salmonid *** barcoding and mini-DNA barcoding are widely used for identifying fish species based on a fragment of the mitochondrial cytochrome c oxidase subunit I(COI)*** this study,rainbow trout(Oncorhynchus mykiss),steelhead trout(***),and Atlantic salmon(Salmo salar)collected from two salmonid aquaculture bases in china were authenticated by DNA barcoding(about 650 bp)and mini-DNA barcoding(127 bp)to evaluate the accuracy of the two methods in the identification of different salmonid *** results revealed that both methods could effectively distinguish *** and *** with 100%***,the two methods failed to separate rainbow trout(***)and steelhead trout(***),which are the same species but cultured in different water ***,salmonid samples from three main distribution channels in the Qingdao area(traditional supermarkets,online supermarkets,and sushi bars)were identified by the two *** of *** with *** was discovered,and the 27.78%overall substitution rate of salmonids in the Qingdao area was higher than those in other regions reported in previous *** addition,the mislabeling rates of salmonids from traditional supermarkets,online supermarkets,and sushi bars were compared in this *** mislabeling rate was significantly greater in sushi bars(50%)than in the other two channels(16.67%),suggesting that stronger monitoring and enforcement measures are necessary for the aquatic food catering industry.

Identification,expression analysis,and functional characterization of ghrelin and its receptors in spotted sea bass(Lateolabrax maculatus)

Ghrelin(Ghrl),an appetite-inducing peptide hormone secreted by the stomach,is the endogenous ligand for the growth hormone secretagogue receptor(Ghs-r).In this study,we identified the preproghrelin gene and its receptors in spotted sea bass(Lateolabrax maculatus).The ghrl gene consisted of an open reading frame(ORF)of 324 nucleotides encoding 107 amino acids,and the premature protein contained a 20-amino-acid mature *** a syntenic analysis,we also validated the annotation of growth hormone secretagogue receptor la(ghs-rla)and growth hormone secretagogue receptor la-1ike(ghs-rla-like),which contained seven-transmembrane structures,in spotted sea *** ORF of ghs-rla consisted of 1152 bp that encoded a 383-amino-acid protein,and(ghs-rla-like) contained an ORF of 2631 bp and produced a protein with 876 amino acids.A phylogenetic analysis showed that spotted sea bass ghrl and its receptors clustered with those of other fishes and were more distantly related to those of other *** situ hybridization revealed that ghrl was highly expressed in the stomach and localized in the mucosa and *** expression of these genes varied during short-term starvation in a time-dependent *** vitro studies showed that after incubation with Ghrl for 3 h enhanced the expression of motilin(mln),gastrin(gas)and cholecystokinin(cck)、but this effect vanished after 6 h of *** summary,Ghrl and its receptors might play important roles in the regulation of food intake in spotted sea bass.

There is an increasing requirement for traceability of aquaculture products, both for consumer protection and for food safety. There are high error rates in the conventional traceability systems depending on physical labels. Genetic traceability technique depending on DNA-based tracking system can overcome this problem. Genealogy information is essential for genetic traceability, and microsatellite DNA marker is a good choice for pedigree analysis. As increasing genotyping throughput of microsatellites, microsatellite multiplex PCR has become a fast and cost-effective technique. As a commercially important cultured aquatic species, Pacific oyster Crassostrea gigas has the highest global production. The objective of this study was to develop microsatellite multiplex PCR panels with dye-labeled universal primer for pedigree analysis in C. gigas, and these multiplex PCRs were validated using 12 full-sib families with known pedigrees. Here we developed six informative multiplex PCRs using 18 genomic microsatellites in C. gigas. Each multiplex panel contained a single universal primer M13(-21) used as a tail on each locus-specific forward primer and a single universal primer M13(-21) labeled with fluorophores. The polymorphisms of the markers were moderate, with an average of 10.3 alleles per locus and average polymorphic information content of 0.740. The observed heterozygosity per locus ranged from 0.492 to 0.822. Cervus simulations revealed that the six panels would still be of great value when massive families were analysed. Pedigree analysis of real offspring demonstrated that 100% of the offspring were unambiguously allocated to their parents when two multiplex PCRs were used. The six sets of multiplex PCRs can be an important tool for tracing cultured individuals, population genetic analysis, and selective breeding program in C. gigas.

Transcriptome profiles of embryos before and after cleavage in Eriocheir sinensis:identification of developmental genes at the earliest stages

In crab, embryogenesis is a complicated developmental program marked by a series of critical events. RNA-Sequencing technology offers developmental biologists a way to identify many more developmental genes than ever before. Here, we present a comprehensive analysis of the transcriptomes of Eriocheir sinensis oosperms （Os） and embryos at the 2-4, cell stage （Cs）, which are separated by a cleavage event. Atotal of 18 923 unigenes were identified, and 403 genes matched with gene ontology （GO） terms related to developmental processes. In total, 432 differentially expressed genes （DEGs） were detected between the two stages. Nine DEGs were specifically expressed at only one stage. These DEGs may be relevant to stage-specific molecular events during development. A number of DEGs related to ＇hedgehog signaling pathway＇, ＇Wnt signaling pathway＇＇germplasm＇, ＇nervous system＇, ＇sensory perception＇ and ＇segment polarity＇ were identified as being up-regulated at the Cs stage. The results suggest that these embryonic developmental events begin before the early cleavage event in crabs, and that many of the genes expressed in the two transeriptomes might be maternal genes. Our study provides ample information for further research on the molecular mechanisms underlying crab development.

Effects of the Amplitude and Frequency of Salinity Fluctuation on the Body Composition and Energy Budget of Juvenile Tongue Sole(Cynoglossus semilaevis)

Effects of the amplitude(± 2, ± 4, ± 6, and ± 8) and frequency(2, 4, and 8 d) of salinity fluctuation on the body composition and energy budget of juvenile tongue sole(Cynoglossus semilaevis) were investigated in a 64-d experiment. Results showed that the amplitude and frequency of salinity fluctuation had significant interaction and both substantially affected the final weight and specific growth rate of juvenile tongue sole. The tongue sole exhibited better growth in treatments with moderate amplitude and frequency of salinity fluctuation(amplitude ± 4–6; frequency 4–8 d) than in other treatments and the control. In terms of energy budget, salinity fluctuation strongly affected the proportions of energy components, including those deposited for growth and lost in respiration, feces, and excretion. Moderately amplitude and frequency of salinity fluctuationg that favored the growth of tongue sole partitioned more energy for growth and less energy for metabolism than the constant and other amplitude and frequency of salinity fluctuation. Average energy budget for tongue sole at moderately fluctuating salinity was determined to be 100C(food) = 30.92G(growth) + 10.30F(feces) + 6.77U(excretion) + 52.01R(respiration). Energetic advantage at moderately fluctuating salinity, including increased energy intake, high assimilation efficiency, reduced metabolism expenditure, and more energy partitioned into growth, might account for the enhancement of tongue sole growth. Commercial farmers are recommended to rear juvenile tongue sole with moderate salinity fluctuations for better growth performance of this species.
