Parkinson’s disease is a common neurodegenerative disease with movement disorders associated with the intracytoplasmic deposition of aggregate proteins such asα-synuclein in *** one of the major intracellular degradation pathways,the autophagy-lysosome pathway plays an important role in eliminating these *** evidence has shown that upregulation of the autophagy-lysosome pathway may contribute to the clearance ofα-synuclein aggregates and protect against degeneration of dopaminergic neurons in Parkinson’s ***,multiple genes associated with the pathogenesis of Parkinson’s disease are intimately linked to alterations in the autophagy-lysosome ***,this pathway appears to be a promising therapeutic target for treatment of Parkinson’s *** this review,we briefly introduce the machinery of ***,we provide a description of the effects of Parkinson’s disease–related genes on the autophagy-lysosome ***,we highlight the potential chemical and genetic therapeutic strategies targeting the autophagy–lysosome pathway and their applications in Parkinson’s disease.

The aim of this paper was to investigate the prevalence of human papilloma virus(HPV)infection in women under 30 years old with cervical intraepithelial neoplasia(CIN)or cervical cancer,in order to provide a basis for cervical cancer prevention and *** recruited 2052 female cases from the education system in Futian District,Shenzhen city,from April 2006 to April 2008,with age ranging from 22 to 60 years ***-generation hybrid capture test was done for detection of female genital tract HPV and cervical colposcopy for screening CIN or cervical *** prevalence of HPV and cervical precancerous lesions or cervical cancer was evaluated in different age groups of *** HPV positive rate was 247/2052(12.00%),and 35/2052(1.71%)of the cases were≥CIN I by pathological *** the 22–29-year-old age group,there were 291 cases,including 39 HPV-positive cases(13.40%)and one case of pathological diagnosis≥CIN I(0.34%),and there were 1761 cases in the 30–60-year-old age group,including 208 HPV-positive cases(11.81%)and 34 cases of pathological diagnosis≥CIN I(1.93%).The HPV-positive rate of 22–29-year-old age group was higher than that of 30–60-year-old age group,but the difference was not signiﬁcant(χ2=0.5967,P=0.4398);the CIN and cervical cancer detection rate of the former group was lower than that of the latter one,and the difference was not signiﬁcant either(χ2=3.7519,P=0.0527).The cases in 30–60-year-old age group were divided intoﬁve age sub-groups,and the HPV-positive rate of the 22–29-year-old age group was compared with the remaining groups,and it was found that the HPV-positive rate of the former group(13.40%)was lower than that of the 40–44-year-old age group(14.70%)but higher than that of the other four ***,the difference in the HPV-positive rate between the 22–29-year age group and the 50–60-year-old age group(6.06%)was signiﬁcant(χ2=5.545,P=0.018),but the difference between the 22–29-year-old age group and each of the other f

Irisin is a myokine that is generated by cleavage of the membrane protein fibronectin type Ⅲ domain-containing protein 5(FNDC5) in response to physical exercise. Studies reveal that irisin/FNDC5 has neuroprotective functions against Alzheimer's disease, the most common form of dementia in the elderly, by improving cognitive function and reducing amyloid-β and tau pathologies as well as neuroinflammation in cell culture or animal models of Alzheimer's disease. Although current and ongoing studies on irisin/FNDC5 show promising results, further mechanistic studies are required to clarify its potential as a meaningful therapeutic target for alleviating Alzheimer's disease. We recently found that irisin treatment reduces amyloid-β pathology by increasing the activity/levels of amyloid-β-degrading enzyme neprilysin secreted from astrocytes. Herein, we present an overview of irisin/FNDC5's protective roles and mechanisms against Alzheimer's disease.

Viruses are important components of the human *** evidence suggests that they are engaged in the physiology and disease status of the *** though the vaginal microbiome is involved in human papillomavirus(HPV)infection and cervical cancer(CC)progression,little is known about the role of the vaginal *** this pilot exploratory study,using unbiased viral metagenomics,we aim to investigate the vaginal eukaryotic virome in women with different levels of cervical lesions,and examine their associations with different cervical disease *** altered eukaryotic virome was observed in women with different levels of lesions and Lactobacillus *** and papillomaviruses are the most commonly detected eukaryotic viruses of the vaginal *** abundance and richness of anelloviruses and papillomaviruses were associated with lowgrade squamous intraepithelial lesion(LSIL)and ***,higher anellovirus abundance was also associated with lactobacillus-depleted microbiome profiles and bacterial community state(CST)type ***,increased correlations between Anelloviridae and Papillomaviridae occurred in the women with increased cervical disease severity level from LSIL to *** data suggest underlying interactions between different microbes as well as the host *** abundance and diversity of both anelloviruses and papillomaviruses shared by LSIL and CC suggest that anellovirus may be used as a potential adjunct biomarker to predict the risk of HPV persistent infection and/or *** studies need to focus on the clinical relevance of anellovirus abundance with cervical disease status,and the evaluation of their potential as a new adjunct biomarker for the prediction and prognoses of CC.

Objective Antimicrobial resistance(AMR)has become a global concern and is especially severe in *** effectively and reliably provide AMR data,we developed a new high-throughput real-time PCR assay based on microfluidic dynamic technology,and screened multiple AMR genes in broiler fecal *** A high-throughput real-time PCR system with an new designed integrated fluidic circuit assay were performed AMR gene detection.A total of 273 broiler fecal samples collected from two geographically separated farms were screened AMR *** The new assay with limits of detection ranging from 40.9 to 8,000 copies/*** sensitivity rate,specificity rate,positive predictive value,negative predictive value and correct indices were 99.30%,98.08%,95.31%,99.79%,and 0.9755,*** this assay,we demonstrate that AMR genes are widely spread,with positive detection rates ranging from 0 to 97.07%in 273 broiler fecal *** CTX-M,bla TEM,mcr-1,fex A,cfr,optr A,and int I1 showed over 80%*** dissemination of AMR genes was distinct between the two *** We successfully established a new high-throughput real-time PCR assay applicable to AMR gene surveillance from fecal *** widespread existence of AMR genes detected in broiler farms highlights the current and severe problem of AMR.

AIM: To evaluate urine β2-microglobulin(β2-M), retinol-binding protein(RBP) excretion, and renal impairment with adefovir dipivoxil(ADV) for chronic hepatitis B. METHODS: We enrolled 165 patients with chronic hepatitis B infection who were treated with ADV monotherapy(n = 90) or ADV plus lamivudine combination therapy(n = 75). An additional 165 chronic hepatitis B patients treated with entecavir were recruited as controls. We detected serum creatinine, urine β2-M, and RBP levels, and estimated the glomerular filtration rate(e GFR) at the initiation of antiviral therapy and every 6 mo for a period of five years. RESULTS: Urine β2-M abnormalities were observed in patients during the first(n = 3), second(n = 7), third(n = 11), fourth(n = 16), and fifth(n = 21) year of ADV treatment. Urinary RBP abnormalities were observed in patients during the first(n = 2), second(n = 8), third(n = 12), fourth(n = 15), and fifth(n = 22) year of ADV treatment. e GFR decreased 20%-30% from baseline in 20 patients, 30%-50% in 12 patients, and > 50% in 3 patients during the five years of treatment. Further analysis indicated that decreases in e GFR of ≥ 30% relative to the baseline level correlated significantly with urine RBP and β2-M abnormalities. In contrast, both serum creatinine and e GFR remained stable in patients treated with entecavir, and only one of these patients developed a urine β2-M abnormality, and two developed urine RBP abnormalities during the five years of treatment. CONCLUSION: Urine RBP and β2-M are biomarkers of renal injury during long-term ADV treatment for chronic hepatitis B, and indicate when treatment should be switched to entecavir.

Because of the diversity of the dinical and laboratory manifestations, the diagnosis of autoimmune liver disease （AILD） remains a challenge in clinical practice. The value of metabolomics has been studied in the diagnosis of many diseases. The present study aimed to determine whether the metabolic profiles, based on ultraperformance liquid chromatography-mass spectrometry （UPLC-MS）, differed between autoimmune hepatitis （AIH） and primary biliary cir- rhosis （PBC）, to identify specific metabolomic markers, and to establish a model for the diagnosis of AIH and PBC. METHODS： Serum samples were collected from 20 patients with PBC, 19 patients with AIH, and 25 healthy individuals. UPLC-MS data of the samples were analyzed using principal component analysis, partial least squares discrimination analysis and or- thogonal partial least squares discrimination analysis. RESULTS： The partial least squares discrimination analysis model （R2y=0.991, Q2=0.943） was established between the AIH and PBC groups and exhibited both sensitivity and speci- ficity of 100%. Five groups of biomarkers were identified, in- eluding bile acids, free fatty acids, phosphatidylcholines, lyso- lecithins and sphingomyelin. Bile acids significantly increased in the AIH and PBC groups compared with the healthy con- trol group. The other biomarkers decreased in the AIH andPBC groups compared with those in the healthy control group. In addition, the biomarkers were downregulated in the AIH group compared with the PBC group. CONCLUSIONS： The biomarkers identified revealed the pathophysiological changes in AILD and helped to discrimi- nate between AIH and PBC. The predictability of this method suggests its potential application in the diagnosis of AILD.
