A total of 400 yeast strains from seawater, sediments, saltern mud, marine fish guts, and marine algae were obtained. The protease activity of the yeast cultures was estimated, after which four strains （HN3.11, Nllb, YF04C and HN4.9） capable of secreting extracellular alkaline protease were isolated. The isolated strains were identified as Aureobasidium pullulans, Yarrowia lipolytica, lssatchenkia orientalis and Cryptococcus cf. aureus. The optimal pH of the protease activity produced by strains HN3.11, YF04C, and HN4.9 was 9.0, while that of the protease produced by strain N1 lb was 10.0. The optimal temperature for protease activity was 45℃for strains HN3.11, N11b, and YF04C, and 50℃ for strain HN4.9. After digestion of shrimp （Penaeus vannamei） protein and spirulina （Arthospira platens＆） protein with the four crude alkaline proteases, the filtrate from spirulina （Arthrospira platensis） powder digested by the crude alkaline protease of strain HNYl 1 was found to have the highest antioxidant activity （61.4%） and the highest angiotensin I converting enzyme （ACE）-inhibitory activities （68.4%）. The other filtrates had much lower antioxidant activity and ACE-inhibitory activities.

Yeast is widely used for industrial production of various types of products,such as ethanol and ***,its fermentation efficiency is strongly reduced by harmful environmental ***,harmful environmental stresses damage important cellular components,such as cell wall,cell membrane,proteins,***,these damages cause cellular metabolic disorders or even *** the past decades,there has been a portfolio of studies on the environmental stress tolerance of yeasts,which mainly aimed at cell damages caused by different environmental stresses,different ways to improve yeast environmental stress tolerance or a tolerance mechanism for certain environmental ***,a comprehensive overview of how yeasts respond to environmental stresses is lacking,and the correlation of tolerance mechanism between different environmental stresses is *** this review,we summarized the general damages induced by most of environmental stresses,the existing major mechanisms of environmental stress tolerance from the perspective of key signalling pathways,and the common ways to improve the resistance to environmental stresses in yeast *** tolerance mechanisms of yeast cells to different environmental stresses are diverse,but sometimes they share the same signalling *** use sensors on the cell surface to recognize environmental stresses and transmit signals to the nucleus to cause changes in gene *** summarizing the main signalling pathways,including MAPK pathway,cAMP/PKA pathway,YAP1/SKN7 pathway,it will provide a powerful reference for future efforts to promote yeast environmental stress tolerance and study yeast tolerance mechanisms.

Pollution by various heavy metals as environmental stress factors might affect bacteria. It was established that iron（Fe（Ⅲ））, manganese（Mn（Ⅱ）） and copper（Cu（Ⅱ）） ion combinations caused effects on Enterococcus hirae that differed from the sum of the effects when the metals were added separately. It was shown that the Cu2＋–Fe3＋combination decreased the growth and ATPase activity of membrane vesicles of wild-type E. hirae ATCC9790 and atp D mutant（with defective FoF1-ATPase） MS116. Addition of Mn2＋–Fe3＋combinations within the same concentration range had no effects on growth compared to control（without heavy metals）. ATPase activity was increased in the presence of Mn2＋–Fe3＋, while together with0.2 mmol/L N,N′-dicyclohexylcarbodiimide（DCCD）, ATPase activity was decreased compared to control（when only 0.2 mmol/L DCCD was present）. These results indicate that heavy metals ion combinations probably affect the FOF1-ATPase, leading to conformational changes. Moreover the action may be direct or be mediated by environment redox *** effects observed when Fe3＋was added separately disappeared in both cases, which might be a result of competing processes between Fe3＋and other heavy metals. These findings are novel and improve the understanding of heavy metals ions effects on bacteria,and could be applied for regulation of stress response patterns in the environment.

Background: The homeobox containing transcription factor Nanog plays crucial roles in embryonic development/proliferation and/or maintenance of spermatogonial stem cells(SSCs) via interacting with transcription factors such as Oct4 and Sox2 in mammals. However, knowledge of its exact mechanistic pathways remains unexploited. Very little is known about teleost Nanog. Information on the Nanog gene of farmed rohu carp(Labeo rohita) is lacking. We cloned and characterized the Nanog gene of rohu carp to understand the expression pattern in early developmental stages and also deduced the genomic organization including promoter ***: Rohu Nanog(LrNanog) cDNA comprised an open reading frame of 1,161 nucleotides bearing a structural homeodomain; whereas, the genomic structure contained four exons and three introns suggesting that it is homologous to mammalian counterparts. Phylogenetical y, it was closely related to freshwater counterparts. Protein sequence(386 AA of42.65 kDa) comparison revealed its low similarity with other vertebrate counterparts except that of the conserved homeodomain. Tissue distribution analysis revealed the existence of LrNanog transcripts only in adult gonads. The heightened abundances in the ovary and proliferating spermatogonia suggested its participations in maternal inheritance and male germ cell development. The potentiating abundances from fertilized egg onwards peaking at blastula stage vis-à-vis decreasing levels from gastrula stage onwards demonstrated its role in embryonic stem cell development. We also provided evidence of its presence in SSCs by western blotting analysis. BCI induces apoptosis via generation of reactive oxygen species and activation of intrinsic mitochondrial pathway in H1299 lung cancer cells

The compound(E)-2-benzylidene-3-(cyclohexylamino)-2,3-dihydro-1 H-inden-1-one(BCI) is known as an inhibitor of dual specific phosphatase 1/6 and mitogen-activated protein kinase. However, its precise anti-lung cancer mechanism remains unknown. In this study, the effects of BCI on the viability of non-small cell lung cancer cell lines NCI-H1299, A549, and NCI-H460 were evaluated. We confirmed that BCI significantly inhibited the viability of p53(-) NCI-H1299 cells as compared to NCI-H460 and A549 cells, which express wild-type p53. Furthermore, BCI treatment increased the level of cellular reactive oxygen species and pre-treatment of cells with N-acetylcysteine markedly attenuated BCI-mediated apoptosis of NCI-H1299 cells. BCI induced cellular morphological changes, inhibited viability, and produced reactive oxygen species in NCI-H1299 cells in a dose-dependent manner. BCI induced processing of caspase-9, caspase-3, and poly ADP-ribose polymerase as well as the release of cytochrome c from the mitochondria into the cytosol. In addition, BCI downregulated Bcl-2 expression and enhanced Bax expression in a dose-dependent manner in NCI-H1299 cells. However, BCI failed to modulate the expression of the death receptor and extrinsic factor caspase-8 and Bid, a linker between the intrinsic and extrinsic apoptotic pathways in NCI-H1299 cells. Thus, BCI induces apoptosis via generation of reactive oxygen species and activation of the intrinsic pathway in NCI-H1299 cells.

Bacteriocins are a large group of chromosome or plasmid-encoded and ribosomally synthesized low-molecular-weight (2 to 6 kDa) antimicrobial and amphiphilous peptides produced by Gr+ or Gr- bacteria [1]. Their low toxicity as well as the absence of allergenicity and reactogenicity is confirmed by testing selected bacteriocins [2] [3]. Bacteriocins can be widely used as preservatives and antibiotic alternatives in medicine. Nisin, a Streptococcus lactis-derived bacteriocin, has been in practice in food industry for a long time. A relevant product contains about 2.5% of nisin. For medical use (e.g., when injected into the blood stream), highly purified drugs are required. However, the yield of bacteriocins accounts for no more than a few percents from the total activity in the culture liquid. In this paper, we propose methods (by example of two B. subtilis strains), allowing to increase the yield up to ~80%. It is believed that other bacteriocins may be purified by these methods and with the same yield.

Maize (Zea mays L.) is the world’s leading cereal crop, with production estimated at over 1 billion tonnes in 2022. In Côte d’Ivoire, maize is one of the most widely consumed foods, with national production exceeding 1 million tonnes in 2022. Despite its importance, this crop is subject to numerous biotic constraints, including Maize streak virus (MSV). The objective of this study was to characterize MSV isolates from Côte d’Ivoire using molecular biology techniques. To achieve this, maize leaf samples displaying characteristic symptoms of MSV were collected from different agro-ecological zones (AEZs) of Côte d’Ivoire. Total DNA was extracted from the collected samples using the CTAB method and quantified with a Nanodrop spectrophotometer. The C2 region of the MSV genome’s open reading frame (ORF) was amplified through polymerase chain reaction (PCR) using MSV-specific primers. The resulting PCR products were sequenced using the Sanger method. Bioinformatics analysis was performed using MSV sequences from other African countries (retrieved from NCBI) alongside sequences obtained in this study. The analysis was conducted using MEGA X version 10.05 software. The results showed that the main symptoms observed in the field included the presence of longitudinal light green streaks on leaves, stunted plant growth, and incomplete seed development in infected plants. Phylogenetic analysis of the sequences from Côte d’Ivoire revealed three monophyletic groups, with sequences of some isolates collected from the same plot belonging to different groups. The MSV sequences from Côte d’Ivoire are very close to those from Rwanda. This study underscores the need for further investigation into the genetic diversity of MSV strains to enhance the diagnosis and management of this viral disease, which is particularly prevalent in maize crops in Côte d’Ivoire.
