Diterpenoids from the aerial parts of Leonurus macranthus

Four new diterpenoids including one cis clerodane-type(1) and three highly oxygenated labdane-type diterpenes(2–4) were isolated from the aerial parts of Leonurus macranthus. Their structures were elucidated on the basis of spectroscopic data(NMR, UV, IR, and MS). Compound 1 represents the first example of cis clerodane-type diterpene in the plants of Leonurus genus. Compounds 1 and 4 exhibited weak inhibition of nitric oxide production in lipopolysaccharide-stimulated BV-2 microglial cells with IC_(50) values of 35.8 ± 3.6 mmol/L and 48.6 ± 4.8 mmol/L,

In the intricate process of natural product biosynthesis, a metabolon can enhance metabolic flux by associating sequential enzymes. A fungal metabolon, comprising of flavin-dependent monooxygenase Spe F and P450 monooxygenase Spe G, is identified in the biosynthesis of spiro polycyclic alkaloids(+)-notoamide B and its diastereomer(+)-versicolamide B. Using notoamide E as a substance, Spe F/Spe G metabolon can control the stereoselectivity of its 2,3-epoxidation, followed by hydrogen atom ion at C-17 to generate reactive epoxide tau-m A with dienyl iminium unit. Subsequently,(+)-notoamide B and(+)-versicolamide B are produced via tandem nonenzymatic inverse-electron-demand Diels-Alder reaction and semipinacol rearrangement. This provides the first example of metabolon in the biosynthesis of spiro-prenylated indole alkaloids.

Behcet's disease is a condition with a complicated and unclear etiology that comprises multi-systemic, chronic, inflammatory vasculitis. Behcet's disease can affect every tissue and organ in the body,and is characterized by recurrent oral and genital ulcers, ocular inflammation, skin lesions, and other manifestations. The incidence of Behcet's disease has a distinct regional specificity, and is most prevalent along the Silk Road, a route that stretched between the Mediterranean, Middle East and Far *** article reviews the recent literature to evaluate the prevalence, clinical manifestations, pathogenesis and mechanism, and current treatments of Behcet's disease. Furthermore, the etiology of Behcet's disease will be evaluated from the aspect of traditional Chinese medicine(TCM) syndrome differentiation. As Behcet's disease is complex and intractable, its treatment warrants further ***, Behcet's disease is treated with Western medicine(WM) via medications that act locally and systemically; this WM treatment protocol usually has a good effect, but relapse can occur after reducing the dosage. Thus, it may be ideal to treat Behcet's disease via a combination of WM and *** studies have indicated that such a combination of Chinese and Western treatments has a better effect than either treatment alone. The aim of the present review is to describe the clinical features of Behcet's disease, and to outline its possible pathogenesis in terms of both TCM and WM. Based on these findings, the present review proposes a Behcet's disease treatment protocol composed of a combination of Chinese and WM that can effectively improve the occurrence of relapse caused by the reduction of the dosage of Western medication.

Brucellosis is an old, infectious and common zoonosis whose causative agents are Gramnegative bacteria from the Brucella genus. Brucellosis is transmitted through direct contactwith infected animals or using unpasteurized dairy products of goats, pigs, camels, sheep,buffalo and cows. Brucellosis is still the most common zoonosis in the world, with mostof cases occurring in developing countries. Today, an approach to traditional medicine andmedicinal plants, especially with regards to the repeated recommendations of the World HealthOrganization, is a necessity. One-third of chemical drugs are produced by using plants andthere is a high potential to produce more drugs from plants. medicinal plants are helpful inthe management of various conditions, especially bacterial diseases. Although there is notenough scientific evidence regarding the clinical effectiveness of herbal drugs for the treatmentof brucellosis, there is strong evidence on the antimicrobial effects of herbal drugs to preventinfection. Therefore, this article seeks to describe the antibacterial effects of some plantderived essential oils or extracts, so that they can serve as promising choices to develop newanti-Brucella medications, as suitable alternatives to conventional antibiotics for brucellosis, asmuch as possible, taking into account the benefits of these herbal drugs.

Programmed cell death ligand-1(PD-L1)is a T cell inhibitory immune checkpoint molecule that interacts with programmed cell death-1(PD-1)to promote immune escape of tumor *** with antibody therapies,small molecule drugs show better prospects due to their advantages such as higher bioavailability,better tissue penetration,and reduced risk of ***,we found that the small molecule demethylzeylasteral(Dem)can significantly downregulate the expression of PD-L1 in colorectal cancer cells and enhance the killing effect of T cells on tumor ***,Dem binds to the deubiquitinating enzyme USP22 and promotes its degradation,resulting in increased ubiquitination and degradation of PD-L1 through the proteasome *** addition,Dem increased the activity of cytotoxic T cells and reduced the number of myeloid-derived suppressor cells(MDSCs)and regulatory T cells(Tregs)in tumor-infiltrating lymphocytes(TILs),thereby activating the tumor immune microenvironment and inhibiting the growth of subcutaneous MC38 tumors in C57BL/6 ***,we also found that the combination of Dem and CTLA4 antibodies can further improve the efficacy of antitumor *** study reveals the mechanism by which Dem promotes PD-L1 degradation and suggests that the combination of Dem and CTLA4 antibodies may improve the efficacy of immunotherapy.

OBJECTIVE:To investigate the efficacy of water fraction from Dioscorea cirrhosa(WF)on oxidative damage and apoptosis of cardiomyocytes induced by H2O2,and to study its ***:Cell viability was measured by the MST assay *** content of malondialdehyde(MDA),release of lactate dehydrogenase(LDH)and activity of catalase(CAT)and superoxide dismutase(SOD)were detected by biochemical *** content of reactive oxygen species(ROS)was assessed by nonfluorescent probe 2′,7′-dichlorofluorescin diacetate(DCFH-DA).JC-1 was used to analyze the mitochondrial membrane potential(mtΔΨ)and Annexin-V-FITC/PI staining was applied to assess apoptosis of H9c2 by flow ***,the expression of B-cell lymphoma-2(Bcl-2),Bcl-2-associated X(Bax),caspase-3,caspase-9,cleaved-caspase-3 and cleaved-caspase-9 proteins was determined by western blot ***:WF increased cell viability and decreased LDH leakage in H9c2 cells exposed to *** treatment decreased ROS and MDA level,enhanced SOD and CAT activities,improved mtΔΨand inhibited *** blot analysis demonstrated that the ratio of Bcl-2/Bax was increased and the expression cleaved-caspase-3,caspase-3,cleaved-caspase-9 and caspase-9 were decreased in group treated with ***:WF protects H9c2 myocardial cells on H2O2-induced oxidative stress and apoptosis by scavenging ROS,improving antioxidant capacity,protecting mitochondrial and regulating the proteins expression related to apoptosis.

As a traditional Chinese medicine,the root of Astragalus membranaceus ***(AMM) or ***(AM) has been widely used in China and other Asian countries for thousands of *** now,the flavonoids,phenolic acids and saponins are considered as the main active components contributing to their therapeutic effect in these *** order to clarify the distribution and contents of these compounds in different organs of these plants,a rapid and sensitive analytical method for simultaneous determination of 25 active compounds including seven types(***,isoflavane,isoflavones,***,phenolic acid and saponins) within 10 min was established using ultra-pressure liquid chromatography coupled with tandem mass spectrometry(UPLC-MS/MS).Then,the established method was fully validated and successfully applied to the determination of the contents of these analytes in different parts(root,rhizome,stem,leaf and flower) of AMM and *** results indicated that the contents of the same type of compounds in two different species plants were significantly ***,the obvious differences were also found for the distribution and contents of different type of compounds in five organs of the same *** present study could provide necessary information for the rational development and utilization of AMM and AM resource.
