Fulminant myocarditis induced by SARS-CoV-2 infection without severe lung involvement:insights into COvID-19 pathogenesis

Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection often leads to pulmonary *** sequelae,including myocarditis and heart failure,have also been ***,the study presents two fulminant myocarditis cases infected by SARS-CoV-2 exhibiting remarkable elevation of cardiac biomarkers without significant pulmonary injury,as determined by imaging *** staining reveals the viral antigen within cardiomyocytes,indicating that SARSCoV-2 could directly infect the *** full viral genomes from respiratory,anal,and myocardial specimens are obtained via next-generation *** analyses of the whole genome and spike gene indicate that viruses in the myocardium/pericardial effusion and anal swabs are closely related and cluster together yet diverge from those in the respiratory *** addition,unique mutations are found in the anal/myocardial strains compared to the respiratory strains,suggesting tissue-specific virus mutation and *** findings indicate genetically distinct SARS-CoV-2 variants have infiltrated and disseminated within myocardial tissues,independent of pulmonary injury,and point to different infection routes between the myocardium and respiratory tract,with myocardial infections potentially arising from intestinal *** findings highlight the potential for systemic SARS-CoV-2 infection and the importance of a thorough multi-organ assessment in patients for a comprehensive understanding of thepathogenesisof COVID-19.

Cretaceous-Neogene Exhumation of the Daqing Shan,North China Constrained by Apatite Fission Track Thermochronology

The Daqing Shan(DQS)located in the Yinshan-Yanshan Orogenic Belt plays an important role in the Mesozoic to Cenozoic evolution of the North China ***,the cooling and exhumation history since the Cretaceous is still *** the apatite fission track(AFT)data in both this study and previous works,a three-stage exhumation history from Cretaceous to Neogene of the DQS is proposed.(1)The first stage is composed of the early exhumation during early Cretaceous driven by the collision between the North China and Siberia cratons(ca.148-132 Ma)and the far-field effect of the subduction of the Pacific Plate(ca.132-114 Ma).(2)Due to the subsidence of the Hetao Basin and the subsequent compensation between the DQS and the Hetao Basin,the DQS experienced the second rapid exhumation from early Eocene to early Oligocene(ca.54-29 Ma).(3)Since the Late Miocene(ca.13.5 Ma),the third rapid cooling and exhumation of the DQS occurred due to the far-field effect of the uplift of the Tibetan Plateau and the subduction of the Pacific Plate.

NEDD8(Neural precursor cell expressed developmentally downregulated protein 8)is an ubiquitin-like protein that is covalently attached to a lysine residue of a protein substrate through a process known as neddylation,catalyzed by the enzyme cascade,namely NEDD8 activating enzyme(E1),NEDD8 conjugating enzyme(E2),and NEDD8 ligase(E3).The substrates of neddylation are categorized into cullins and non-cullin *** of cullins activates CRLs(cullin RING ligases),the largest family of E3 ligases,whereas neddylation of non-cullin substrates alters their stability and activity,as well as subcellular ***,the neddylation pathway and/or many neddylation substrates are abnormally activated or over-expressed in various human diseases,such as metabolic disorders,liver dysfunction,neurodegenerative disorders,and cancers,among ***,targeting neddylation becomes an attractive strategy for the treatment of these *** this review,we first provide a general introduction on the neddylation cascade,its biochemical process and regulation,and the crystal structures of neddylation enzymes in complex with cullin substrates;then discuss how neddylation governs various key biological processes via the modification of cullins and non-cullin *** further review the literature data on dysregulated neddylation in several human diseases,particularly cancer,followed by an outline of current efforts in the discovery of small molecule inhibitors of neddylation as a promising therapeutic ***,few perspectives were proposed for extensive future investigations.

Ferroptosis, a unique type of non-apoptotic cell death resulting from iron-dependent lipid peroxidation, has a potential physiological function in tumor suppression, but its underlying mechanisms have not been fully elucidated. Here, we report that the long non-coding RNA(lncRNA) LncFASA increases the susceptibility of triple-negative breast cancer(TNBC) to ferroptosis. As a tumor suppressor, LncFASA drives the formation of droplets containing peroxiredoxin1(PRDX1), a member of the peroxidase family, resulting in the accumulation of lipid peroxidation via the SLC7A11-GPX4 axis. Mechanistically, LncFASA directly binds to the Ahpc-TSA domain of PRDX1, inhibiting its peroxidase activity by driving liquid-liquid phase separation, which disrupts intracellular ROS homeostasis. Notably, high LncFASA expression indicates favorable overall survival in individuals with breast cancer, and LncFASA impairs the growth of breast xenograft tumors by modulating ferroptosis. Together, our findings illustrate the crucial role of this lncRNA in ferroptosis-mediated cancer development and provide new insights into therapeutic strategies for breast cancer.

Introduction:Respiratory infectious diseases,such as influenza and coronavirus disease 2019(COVID-19),present significant global public health *** emergence of artificial intelligence(AI)and big data offers opportunities to improve traditional disease surveillance and early warning ***:The study analyzed data from January 2020 to May 2023,comprising influenza-like illness(ILI)statistics,Baidu index,and clinical data from *** methodologies were evaluated:the adaptive dynamic threshold method(ADTM)for dynamic threshold adjustments,the machine learning supervised method(MLSM),and the machine learning unsupervised method(MLUM)utilizing anomaly *** comparison focused on sensitivity,specificity,timeliness,and warning ***:ADTM issued 37 warnings with a sensitivity of 71%and a specificity of 85%.MLSM generated 35 warnings,with a sensitivity of 82%and a specificity of 87%.MLUM produced 63 warnings with a sensitivity of 100%and specificity of 80%.The initial warnings from ADTM and MLUM preceded those from MLSM by five *** Kappa coefficient indicated moderate agreement between the methods,with values ranging from 0.52 to 0.62(Pearly warning *** emphasizes the validation of machine learning’s reliability and underscores the unique advantages of each ***,it stresses the significance of integrating machine learning models with various data sources to enhance public health preparedness and response,alongside acknowledging limitations and the need for broader validation.
