Dietary fatty acids and adipose tissue inflammation at the crossroad between obesity and colorectal cancer

Excess adiposity, a worldwide-growing pathological condition, is now recognized as a main risk factor for most chronic diseases including colorectal cancer (CRC). Obese subjects show an increased cancer incidence with obesity representing an important indicator of survival, prognosis, recurrence and response to therapy. A low-grade chronic inflammation of metabolically active tissues including the adipose tissue (AT), defined as meta-inflammation, is a main feature of obesity. Fatty acids (FA), the main AT components, are important modulators of inflammation, and the type of FA stored in AT critically affects tissue functions. Their profile within AT mirrors FA dietary intake but also depends on a metabolic control. Obesity, changes in the habitual diet, weight loss or pathological conditions like CRC influence FA profile of AT pointing to these molecules as important actors in AT dysfunction and meta-inflammation, that characterize metabolic diseases and may favor cancer development. Worth of note, diet is receiving growing attention as a main determinant in cancer prevention due to its capacity to modulate immune response and inflammation. Alterations in the balance between different families of FA may contribute to generate a pro-inflammatory profile with deleterious effects on metabolic and immune homeostasis at both local and systemic levels. This review focuses on FA as regulators of human AT inflammation discussing the role of obesity-, diet-, and weight loss-associated changes in FA profile in this process. The relevance of FA composition of AT in linking diet, obesity and CRC will be also reviewed.

Comparative study of behavior of inhaled samarium and cerium in mice

Distribution of Sm and Ce in mice was investigated after inhalation exposure to their oxide particles of 5 μm diameter and 15 mg/m3.Sm and Ce were mainly distributed in lung,their concentrations decreased with passing time,and their concentrations in lung were correlated with exposure periods.Concentrations of Ce were lower than that of Sm in each point.Sm was also distributed in the other organs and bone but Ce was scarcely detected.These results suggest that the behaviors of Sm and Ce inhaled were different,although their chemical properties are similar.

Interplay of neuropilin-1 and semaphorin 3A after partial hepatectomy in rats

AIM:To elucidate the role of neuropilin-1(Nrp-1) and semaphorin 3A(Sema3A) in sinusoidal remodeling during liver regeneration in rats.METHODS:Male Wistar/ST rats at 7 wk of age,weighing about 200 g,were used for all animal experiments.In vivo,at 24,48,72,96,144 and 192 h after twothirds partial hepatectomy(PHx),the remnant livers were removed.Liver tissues were immunohistochemically stained for Nrp-1,Sema3A and SE-1,a liver sinusoidal endothelial cell(SEC) marker.Total RNA of the liver tissue was extracted and reversely transcribed into cDNA.The mRNA expression of Sema3A was analyzed by quantitative real-time polymerase chain reaction and normalized to that of ribosomal protein S18.In vitro,SECs were isolated from rat liver and cultured in endothelial growth medium containing 20 ng/mL vascular endothelial cell growth factor.Migration of SECs in primary culture was assessed by cell transwell assay with or without recombinant Sema3A.Apoptotic cells were determined by a terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end labeling method.RESULTS:In vitro,immunohistochemistry study revealed that Sema3A and Nrp-1 were constitutively expressed in hepatocytes and SECs,respectively,in normal rat liver tissues.Nrp-1 expression in SECs was quantified by the percentage of immunostained area with antiNrp-1 antibody in relation to the area stained with SE-1.Between 24 h and 96 h following resection of liver,Nrp-1 expression in SECs was transiently increased.Compared with the baseline(5.2% ± 0.1%),Nrp-1 expression in SECs significantly increased at 24 h(17.3% ± 0.7%,P < 0.05),48 h(39.1% ± 0.6%,P < 0.01),72 h(46.9% ± 4.5%,P < 0.01) and 96 h(29.9% ± 3.8%,P < 0.01) after PHx,then returned to the basal level at termination of liver regeneration.Interestingly,the expression of Sema3A was inversely associated with that of Nrp-1 in liver after PHx.Sema3A mRNA expression was significantly reduced by about 75% over the period 24-144 h after PHx(P < 0.05),and returned to basal levels at 192 h after PHx.In vi

Nanocatalytic bacteria disintegration reverses immunosuppression of colorectal cancer

Tumor-associated bacteria(TAB) play a critically important role in regulating the microenvironment of a tumor, which consequently greatly deteriorates the therapeutic effects by chemo-and radiotherapy deactivation and, more considerably, leads to substantial immunosuppression. On the contrary, herein we propose a nanocatalytic tumor-immunotherapeutic modality based on the bacteria disintegration by bacteria-specific oxidative damage under magnetic hyperthermia for highly effective immune response activation-promoted tumor regression. A monodispersed and superparamagnetic nanocatalytic medicine modified by arginyl-glycyl-aspartic acid(RGD) and(3-carboxypropyl)triphenylphosphonium bromide(TPP), named as MNP-RGD-TPP herein, has been synthesized, which features selective accumulation at the TAB by the electrostatic affinity, enabling effective TAB disintegration by the nanocatalytic Fenton reaction producing abundant cytotoxic hydroxyl radicals in situ under alternating magnetic field-induced hyperthermia. More importantly, the lipopolysaccharide has been metabolically secreted from the destructed TAB as pathogen-associated molecular patterns(PAMPs) to M1-polarize tumor-associated macrophages(TAMs) and promote the maturation of dendritic cells(DCs) for innate immuno-response activation of TAMs, followed by cytotoxic T lymphocytes awakening under the PAMPs presentation by the mature DCs against tumor cells. The integrated innate and adaptive immunity activations based on this TAB-promoted nanocatalytic immunomedicine, instead of magnetic heating-induced hyperthermia or the released Fe2+/Fe3+Fenton agent, has been found to achieve excellent therapeutic efficacy in an orthotopic colorectal cancer model, demonstrating the great potential of such an integrated immunity strategy in clinical tumor immunotherapy.

Inhibition of rheumatoid arthritis by blocking connective tissue growth factor

The pathogenesis of rheumatoid arthritis(RA) remains to be completely elucidated so far; however, it is known that proinflammatory cytokines play a pivotal role in the induction of RA. Tumor necrosis factor(TNF-α), in particular, is considered to play a central role in bone destruction by mediating the abnormal activation of osteoclasts or the production of proteolytic enzymes through direct or indirect mechanisms. The use of TNF-α blocking agents has a significant impact on RA therapy. Anti-TNF-α blocking agents such as infliximab are very effective for treatment of RA, especially for the prevention of articular destruction. We have previously shown that several proteins exhibited extensive changes in their expression after amelioration of RA with infliximab treatment. Among the proteins, connective tissue growth factor(CTGF) has a significantrole for the development of RA. Herein, we review the function of CTGF in the pathogenesis of RA and discuss the possibility of a novel treatment for RA. We propose that CTGF is a potentially novel effector molecule in the pathogenesis of RA. Blocking the CTGF pathways by biological agents may have great beneficial effect in patients with RA.

Background:By the end of October 2019,there were 958 thousand people were reported living with HIV/AIDS in China.Unhealthy lifestyle factors,such as smoking,drinking alcohol,using illicit drugs and no physical activity have been found to mitigate the positive impact of antiretroviral therapy(ART)on viral load and HIV-related quality of life.Moreover,risky sexual behavior among HIV-positive persons places their partners at risk for HIV transmission and other sexually transmitted infections.The aim of the study is to determine the prevalence of unhealthy behavior of people living with HIV/AIDS and related influencing factors,particularly those that are closely connected with HIV infection and ART effects.Methods:An institutional based cross-sectional study design was used to collect data from people living with HIV/AIDS(PLWHA)in Beijing and Yunnan Province.The following information was included in the questionnaire survey:social-demographic characteristics,health behavior information,sexual risk behaviors.Binary logistic regression model was conducted to analyze the influencing factors of unhealthy general health behaviors and risky sexual behaviors.Results:In total,2575 PLWHA were included in the study and 78.3%(2017/2575)were male.For the general health behaviors,34.2%(987/2544)smoke;33.8%(870/2575)drank alcohol and 2.3%(49/2134)reported the use of illicit drugs in the previous 6 months.From the sexual behaviors perspective,59.0%(1519/2575)had sex in the previous 6 months.Among people who had sex,92.0%(1398/1519)had fixed sexual partners.Among those with no fixed sexual partner,38.0%(46/121)had more than three partners.Among men who had sex,34.7%(448/1292)reported having sex with men in the previous 6 months and 167%(75/448)of these had group sexual activity.Among participants,72.2%(1053/1458)used condoms every time they had sex while 6.4%(94/1458)of people never used condom.Male people living with HIV/AIDS were more likely to have sexual risk behaviors(adjusted odds ratio[OR]=2.208,95%confidence interval[CI]:1.1

Nation from Village: The Evolving Landscape for Rural Women in the First Half of the Twentieth Century

In the first half of the 20th century, during China's rapid transition from tradition to modernity, three major historical events occurred successively in Ding County, Hebei Province. These events illustrate the gradual broadening of the horizons of some rural women: shifting from the Confucian expectation that "women's rightful place is in the home" to a gradual extension into the public domain. The Zhaicheng village system took the first small step in breaking the Confucian gender norms by promoting female education in public spaces, yet it did not truly challenge the fundamental gender order of males outside the home and females within it. The Mass Education Movement saw the emergence of the first group of professional women in rural Ding County. However, they faced social exclusion as a consequence of entering the public domain. The Zhaicheng system and the gender-based teaching practices during the Mass Education Movement sowed the seeds of the impending tension and conflict experienced by women juggling dual roles within and outside the family. Amidst the bloodshed of the Chinese People's War of Resistance against Japanese Aggression and the War of Liberation(wars of Resistance and Liberation), female warriors, through sacrifice and devotion, ventured into the newly expanded public sphere of war, yet traditional gender norms continued to confine their broadened horizons to a transient phase in their lives. A collective examination of these three major occurrences shows that the gradual broadening of horizons for rural women in Ding County during the first half of the 20th century was inherent in the developmental trajectory of the village, embodying the idea that the modern nation was gradually emerging within the village in the concept of "nation from village."

Emergency department management of acute agitation in the reproductive age female and pregnancy

BACKGROUND:Agitation is a common presentation within emergent departments(EDs).Agitation during pregnancy should be treated as an obstetric emergency,as the distress may jeopardize both the patient and fetus.The safety of psychotropic medications in the reproductive age female has not been well established.This review aimed to explore a summary of general agitation recommendations with an emphasis on ED management of agitation during pregnancy.METHODS:A literature review was conducted to explore the pathophysiology of acute agitation and devise a preferred treatment plan for ED management of acute agitation in the reproductive age or pregnant female.RESULTS:While nonpharmacological management is preferred,ED visits for agitation often require medical management.Medication should be selected based on the etiology of agitation and the clinical setting to avoid major adverse effects.Adverse effects are common in pregnant females.For mild to moderate agitation in pregnancy,diphenhydramine is an effective sedating agent with minimal adverse effects.In moderate to severe agitation,high-potency typical psychotropics are preferred due to their neutral effects on hemodynamics.Haloperidol has become the most frequently utilized psychotropic for agitation during pregnancy.Second generation psychotropics are often utilized as second-line therapy,including risperidone.Benzodiazepines and ketamine have demonstrated adverse fetal outcomes.CONCLUSION:While randomized control studies cannot be ethically conducted on pregnant patients requiring sedation,animal models and epidemiologic studies have demonstrated the effects of psychotropic medication exposure in utero.As the fetal risk associated with multiple doses of psychotropic medications remains unknown,weighing the risks and benefits of each agent,while utilizing the lowest effective dose remains critical in the treatment of acute agitation within the EDs.

Ginger-indirect moxibustion plus acupuncture versus acupuncture alone for chronic fatigue syndrome:a randomized controlled trial

OBJECTIVE:To assess the efficacy and safety of gingerindirect moxibustion for chronic fatigue syndrome(CFS).METHODS:In this central randomized,controlled trial,290 CFS participants were recruited and randomly allocated to group A(ginger-indirect moxibustion plus acupuncture)or group B(acupuncture alone).The study consisted of a treatment period of 8 weeks with a total of 24 treatments(3 sessions per week,every other day),and a follow-up period of 12 weeks.The outcome was measured by Fatigue Severity Scale(FSS),Psychological Health Report(SPHERE),the Self-rating depression scale(SDS)and the Hamilton anxiety scale(HAMA)at baseline,2,4,6,8,12 and 20 weeks.RESULTS:With the treatment undergoing,the changes of FSS,SPHERE,SDS and HAMA scores in both groups increased gradually,and the effect maintained at the 12th week.Between groups,significantly higher score changes were seen in group A in FSS after 4 weeks treatment(11.94 vs 9.12,95%CI:0.94,4.7)and in SPHERE after 2 weeks treatment(3.7 vs 2.27,95%CI:0.56,2.31).But for SDS and HAMA,the improvement did not differ significantly between groups.No severe adverse events were reported.CONCLUSION:Ginger-indirect moxibustion is a safe and effective intervention to relieve fatigue and accompanying physical symptoms of CFS.
