In this study,intelligent,pH-responsive colorimetric films were prepared by encapsulating anthocyanins in nanocomplexes prepared from glutenin and carboxymethyl *** nanocomplexes were added to a corn starch matrix and used in the freshness monitoring of chilled *** effects of anthocyanin-loaded nanocomplexes on the physical,structural,and functional characteristics of the films were *** addition of anthocyanin-loaded nanocomplexes increased the tensile strength,elongation at break,hydrophobicity,and light transmittance of the films while decreasing their water vapor *** is because new hydrogen bonds are formed between the film components,resulting in a more homogeneous and dense *** colorimetric film has a significant color response to pH *** films were used in experiments on the freshness of chilled pork,in which the pH changes with changing freshness *** results show that the colorimetric film can monitor changes in the freshness of chilled pork in real time,where orange,pink,and green represent the fresh,secondary fresh,and putrefied states of pork,***,the intelligent colorimetric film developed in this study has good application potential in the food industry.

Gluconobacter oxydans is an important Gram-negative industrial microorganism that produces vitamin C and other products due to its efficient membrane-bound dehydrogenase *** incomplete oxidation system has many crucial industrial ***,it also leads to slow growth and low biomass,requiring further metabolic modification for balancing the cell growth and incomplete oxidation *** a non-model strain,*** lacks efficient genome editing tools and cannot perform rapid multi-gene editing and complex metabolic network *** the last 15 years,our laboratory attempted to deploy multiple CRISPR/Cas systems in different *** strains and found none of them as *** this study,Cpf1-based or dCpf1-based CRISPRi was constructed to explore the targeted binding ability of Cpf1,while Cpf1–FokI was deployed to study its nuclease activity.A study on Cpf1 found that the CRISPR/Cpf1 system could locate the target genes in *** but lacked the nuclease cleavage ***,the CRISPR/Cpf1–FokI system based on FokI nuclease was ***-gene knockout with efficiency up to 100%and double-gene iterative editing were achieved in *** this system,AcrVA6,the anti-CRISPR protein of *** was discovered for the first time,and efficient genome editing was realized.

Tripartite motif 21(TRIM21)is an E3 ubiquitin ligase that shows great promise for protein degradation through ***,ultrasmall chiral gold nanoparticles(D-or L-NPs)modified by D-or L-glutathione ligands were *** conjugated with an NLR family pyrin domain-containing protein 3(NLRP3)antibody(D-NP-a NLRP3 or L-NP-a NLRP3),DNP-a NLRP3 showed effective delivery efficiency of the antibody into microglia and prevented Aβ-mediated microglia senescence,and p16^(ink4a),a marker of senescence,in the microglia was reduced by 90.3%±7.8% while L-NP-a NLRP3 decreased by 48.01%±3.1%.Mechanistic investigations revealed that the D-NP-a NLRP3((1.5±0.3)×10^(7)M^(-1))exhibited sixteen-fold larger binding affinity to transmembrane glycoprotein SLC3A2 than L-type((9.5±2.7)×10^(5)M^(-1)),which led to a high efficiency of antibody delivery and TRIM21-dependent NLRP3 ***,the blood-brain barrier(BBB)-crossing ability of chiral NP-a NLRP3 as well as NLRP3 degradation was demonstrated in *** APP/PS1 Alzheimer's disease(AD)model mice experiments exhibited a reduction of 89.7%±6.8% for the NLRP3 protein and 84.2±7.5% for p16^(ink4a)following the intravenous administration of D-NP-a NLRP3 once a week for 60 ***,the levels of the AD markers Aβ and phosphorylatedTau in the brains were reduced by 86.2%±8.2% and 81.6%±9.1%;these were 2.1-fold and 1.9-fold higher than those treated with L-NP-a NLRP3,*** studies provide a method to rescue AD-like pathologies and prevent senescence.

During the development of diet-induced obesity， the change of energy matebolism is closely related to the function of the circadian clock in mammals. Luteolin (LU)， one of the most common natural flavonoids riched in many edible plants， can ameliorate obesity by activating adipose tissue browning， but its effect on circadian clock in this process remains poorly understood. Here we found that dietary LU improved circadian misalignment of energy expenditure in high-fat diet (HFD)-fed wild-type (WT) mice. Moreover， dietary LU efficiently elevated UCP1 levels in adipose tissue during the dark period， which was similar to the LU-increased hepatic PER2 expressions. Hepatic PPARα/RXRα/FGF21 pathway was rhythmically elevated by dietary LU in HFD-fed WT mice， whereas the promotion was inhibited in Per2-/-mice. Meanwhile， Per2 deletion abolished the effects of dietary LU on adipose tissue browning in HFD-fed mice. Further， LU treatment directly activated PPARα/RXRα/FGF21 signaling in primary cultured hepatocytes from WT mice rather than Per2-/-mice. Taken together， the deletion of the core clock component Per2 impedes LU-induced adipose tissue browning through weakening PPARα/RXRα/FGF21 pathway in mice， providing a new insight into the interplay of energy metabolism and circadian clock for the anti-obesity activity of LU.

Twenty-four rice genotypes were examined to assess genetic variability,heritability,and correlations for seven-grain quality traits,eight nutritional elements,and *** revealed signiﬁcant differences for the quality traits *** every trait under study,the phenotypic coefﬁcient of variation was higher than the correspon-dence genotypic coefﬁcient of *** in a broad sense varied from 29.75%for grain length to 98.31%for the elongation *** percentage recovery had a highly signiﬁcant positive correlation with milling and head rice ***,milling percentage had a highly positive correlation with head rice *** amylose percentage,all the genotypes belonged to low amylose except the Hassawi-1 variety,which had intermediate amylose *** nutrition contents of magnesium(Mg),sodium(Na),potas-sium(K),calcium(Ca),copper(Cu),manganese(Mn),zinc(Zn),iron(Fe),or protein percentage gave different variations for 24 rice genotypes under all the nutritional *** the 24 genotypes,ten rice genotypes–HighNutrient-1,HighNutrient-2,HighNutrient-9,HighNutrient-8,HighNutrient-3,Hassawi-2,HighNutrient-7,HighNutrient-6,Hassawi-1,and HighNutrient-4–had the highest heist value for all nutritional and protein con-tents,and could be used as a donor to improving new *** was a positive and signiﬁcant correlation between magnesium Mg,K,Zn and ***,K had a positive correlation with zinc Zn,Fe,and protein *** analysis was divided into two groups:theﬁrst group included all genotypes rich in nutri-ents,while the remaining genotypes with low nutritional content were included in the second group.

The association between the exposure of organochlorine pesticides(OCPs)and serum uric acid(UA)levels remained *** this study,to investigate the combined effects of OCP mixtures on hyperuricemia,we analyzed serum OCPs and UA levels in adults from the National health and Nutrition Examination Survey(2005–2016).Four statistical models including weighted logistic regression,weighted quantile sum(WQS),quantile g-computation(QGC),and bayesian kernel machine regression(BKMR)were used to assess the relationship between mixed chemical exposures and *** analyses were conducted to explore potential *** 6,529 participants,the prevalence of hyperuricemia was 21.15%.Logistic regression revealed a significant association between both hexachlorobenzene(HCB)and trans-nonachlor and hyperuricemia in the fifth quintile(OR:1.54,95%CI:1.08–2.19;OR:1.58,95%CI:1.05–2.39,respectively),utilizing the first quintile as a *** and QGC analyses showed significant overall effects of OCPs on hyperuricemia,with an OR of 1.25(95%CI:1.09–1.44)and 1.20(95%CI:1.06–1.37),*** indicated a positive trend between mixed OCPs and hyperuricemia,with HCB having the largest weight in all three mixture *** analyses revealed that females,individuals aged 50 years and above,and those with a low income were more vulnerable to mixed OCP *** results highlight the urgent need to protect vulnerable populations from OCPs and to properly evaluate the health effects of multiple exposures on hyperuricemia using mutual validation approaches.

Stroke is one of the leading causes of death and disability ***,information on stroke-related tongue coating microbiome(TCM)is limited,and whether TCM modulation could benefit for stroke prevention and rehabilitation is ***,TCM from stroke patients(SP)was characterized using molecular *** occurrence of stroke resulted in TCM dysbiosis with significantly reduced species richness and *** abundance of Prevotella,Leptotrichia,Actinomyces,Alloprevotella,Haemophilus,and TM7_[G-1]were greatly reduced,but common infection Streptococcus and Pseudomonas were remarkably ***,an antioxidative probiotic Lactiplantibacillus plantarum AR113 was used for TCM intervention in stroke rats with cerebral ischemia/reperfusion(I/R).AR113 partly restored I/R induced change of TCM and gut microbiota with significantly improved neurological deficit,relieved histopathologic change,increased activities of antioxidant enzymes,and decreased contents of oxidative stress ***,the gene expression of antioxidant-related proteins and apoptosis-related factors heme oxygenase-1(HO-1),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),nuclear factor erythroid 2-related factor 2(Nrf2),NAD(P)H:quinone oxidoreductase-1(NQO-1),and Bcl-2 was significantly increased,but cytochrome C,cleaved caspase-3,and Bax were markedly decreased in the brain by AR113 *** results suggested that AR113 could ameliorate cerebral I/R injury through antioxidation and anti-apoptosis pathways,and AR113 intervention of TCM may have the application potential for stroke prevention and control.
