Plant viruses are a group of intracellular pathogens that persistently threaten global food *** advances in plant virology have been achieved by Chinese scientists over the last 20 years,including basic research and technologies for preventing and controlling plant viral ***,we review these milestones and advances,including the identification of new crop-infecting viruses,dissection of pathogenic mechanisms of multiple viruses,examination of multilayered interactions among viruses,their host plants,and virus-transmitting arthropod vectors,and in-depth interrogation of plantencoded resistance and susceptibility ***,various plant virus-based vectors have also been successfully developed for gene function studies and target gene expression in *** also recommend future plant virology studies in China.

Background:Acute suppurative terminal cholangitis(ASTC)is rarer than acute obstructive cholangitis and is not well *** explore this subtype of acute cholangitis,we described our clinical experience with ***:We performed a retrospective review of patients with ASTC admitted to our center from September 2014 to August *** analyzed their clinical characteristics,including etiology,clinical manifestations,imaging features,treatment and ***:A total of 32 ASTC patients were included in the *** majority of the patients had a history of biliary operations,and clinical manifestations were occult and *** positive rate of bacterial culture was 46.9%.All the patients had typical imaging features on computed tomography and magnetic resonance *** with effective antibiotics was provided as soon as diagnosis was *** treatment,most patients had a good *** levels of total bilirubin,aspartate aminotransferase,procalcitonin and gamma-glutamyltransferase were the characteristics of critically ill patients and were associated with relatively poor ***:Our results demonstrated that ASTC should be recognized as a new subtype of acute cholangitis,and that earlier diagnosis and more personalized treatments are needed.

Plant cell walls are a critical site where plants and pathogens continuously struggle for physiological *** we show that dynamic remodeling of pectin methylesterification of plant cell walls is a compo-nent of the physiological and co-evolutionary struggles between hosts and pathogens.A pectin methyles-terase(PsPME1)secreted by Phytophthora sojae decreases the degree of pectin methylesterification,thus synergizing with an endo-polygalacturonase(PsPG1)to weaken plant cell *** counter PsPME1-mediated susceptibility,a plant-derived pectin methylesterase inhibitor protein,GmPMl1,protects pectin to maintain a high methylesterification ***1 protects plant cell walls from enzymatic degrada-tion by inhibiting both soybean and *** pectin methylesterases during ***,constitutive expression of GmPMl1 disrupted the trade-off between host growth and defense *** therefore used AlphaFold structure tools to design a modified form of GmPMI1(GmPMI1R)that specifically targets and inhibits pectin methylesterases secreted from pathogens but notfrom *** expression of GmPMi1R enhanced plant resistance to oomycete and fungal *** summary,our work highlights the biochemical modification of the cell wall as an important focal point in the physiological and co-evolutionary conflict between hosts and microbes,providing an important proof of concept that Al-driven structure-based tools can accelerate the development of new strategies for plant protection.

Wheat germplasm is a fundamental resource for basic research,applied studies,and wheat breeding,which can be enriched normally by several paths,such as collecting natural lines,accumulating breeding lines,and introducing mutagenesis *** methane sulfonate(EMS)is an alkylating agent that can effectively introduce genetic variations in a wide variety of plant *** this study,we created a million-scale EMS population(MEP)that started with the Chinese wheat cultivars‘Luyan 128’,‘Jimai 38’,‘Jimai 44’,and‘Shannong 30’.In the M1 generation,the MEP had numerous phenotypical variations,such as>3,000 chlorophyll-deficient mutants,2,519 compact spikes,and 1,692 male sterile *** were also rare mutations,including 30 independent tillers each with double *** M1 variations of chlorophyll-deficiency and compact spikes were inheritable,appearing in the M2 or M3 *** advance the entire MEP to higher generations,we adopted a single-seed descendent(SSD)*** other seed composites of M2 were used to screen other agronomically important traits,such as the tolerance to herbicide *** MEP is available for collaborative projects,and provides a valuable toolbox for wheat genetics and breeding for sustainable agriculture.

An efficient and universal protoplast-based transient gene expression system for genome editing in Brassica crops

Protoplast-based transient gene expression system has been widely used in plant genome editing because of its simple operation and less *** order to establish a universal protoplast-based transient transfection system for verifying activities of genome editing vectors containing targets in Brassica,we systematically optimized factors affecting protoplast isolation and transient gene *** established an efficient protoplast-based transient gene expression system(PTGE)in Chinese cabbage,achieving high protoplast yield of 4.9×10^(5)·g^(-1)FW,viability over 95%,and transfection efficiency of 76%.We showed for the first time that pretreatment of protoplasts with a hypotonic MMG could significantly enhance the transfection ***,protoplasts incubated at 37℃ for 6 min improved the transfection efficiency to 86%.We also demonstrated that PTGE worked well(more than 50%transfection efficiency)in multiple Brassica species including cabbage,Pak Choi,Chinese kale,and ***,PTGE was used for validating the activities of CRISPR/Cas9 vectors containing targets in Chinese cabbage,cabbage,and pak choi,demonstrating the broad applicability of the established PTGE for genome editing in Brassica crops.

Histopathological diagnosis is a crucial part of surgical treatment,as it determines further treatment decisions and prognosis for *** to the heterogeneity of tumors,traditional histopathological diagnosis relies on histological and cytological morphological analysis of cryo-slides by well-trained pathologists,which is subjective,time-consuming,and lacks considerable ***,we engineer a series of tumor receptor-seeking dyes to automatically label and visually depict the tumor contour with antibody-free *** meso-chlorine on the cyclohexyl ring of dyes can covalently react with specific thiol groups in various tumor signature proteins to form stable dye@protein *** dye@protein complexes have significantly amplified fluorescence signals in tumor sites,enabling highresolution delineation of tumormargins and providing histological morphology information to potentially determine tumor *** orthogonal effect not only avoids errors caused by individual signature proteins,but also amplifies the fluorescence difference between tumor and normal ***,our method is easy to operate with a minimal detection time of only 4 *** tumor receptor-seeking dyes successfully assess tumor-positive margins from 66 resected breast tumor samples and 25 intraoperative cancer patients with 100%accuracy,providing a newgeneration technique for intraoperative pathologic diagnosis of tumor biopsy.

The Impact of Blood Transfusion on the Efficiency of Stem Cell Transplants

Background: While blood product transfusion is essential for managing hematologic deficits in Allogenic Hematopoietic stem cell transplant (AHSCT) recipients, it has risks including infectious disease transmission, alloimmunization, and transfusion reactions. These risks have sparked an ongoing debate regarding the overall impact of transfusions on patient outcomes. Thus, this study aimed to evaluate the impact of Red Blood Cells (RBCs) and/or platelet transfusion on the infection incidence and overall survival in AHSCT patients. Methods: We performed a retrospective analysis of clinical and laboratory data of sixty adult patients with primary malignant hematological disorder who had undergone AHSCT. Participants’ data were categorized into two groups;Group 1 (low transfusion group) consisted of patients receiving 10 units. Quantitative data were expressed as mean ± SD. The t-test of significance and Chi-square (χ2) test were used, with p ≤ 0.05 considered significant. Result: A total of 60 patients’ data was included. In Group 1, out of 30 patients, 13 (43.33%) developed infections. In contrast, Group 2 had 21 (70%) out of 30 patients develop infections. Group 1 had a higher survival rate (57.8%) than Group 2 (transfusion > 10 units) (46.2%) with a chi-square value = 23.56, and p-value Conclusion: The volume of blood product transfusions has a considerable impact on patient outcomes, particularly infection and survival rates. Additional long-term prospective studies and larger randomized controlled trials are needed to strengthen the evidence for determining transfusion protocols for these patients.

The mucosal barrier is crucial for intestinal homeostasis,and goblet cells are essential for maintaining the mucosal barrier *** proviral integration site for Moloney murine leukemia virus-1(PIM1)kinase regulates multiple cellular functions,but its role in intestinal homeostasis during colitis is ***,we demonstrate that PIM1 is prominently elevated in the colonic epithelia of both ulcerative colitis patients and murine models,in the presence of intestinal *** PIM1 leads to decreased goblet cells,thus impairing resistance to colitis and colitis-associated colorectal cancer(CAC)in ***,PIM1 modulates goblet cell differentiation through the Wnt and Notch signaling ***,PIM1 interacts with histone deacetylase 2(HDAC2)and downregulates its level via phosphorylation,thereby altering the epigenetic profiles of Wnt signaling pathway ***,these findings investigate the unknown function of the PIM1-HDAC2 axis in goblet cell differentiation and ulcerative colitis/CAC pathogenesis,which points to the potential for PIM1-targeted therapies of ulcerative colitis and CAC.
