Objective: Baicalin is a major active ingredient from Radix Scutellariae, and has various pharmacological effects, such as anti-inflammatory, anti-tumor, anti-virus, and antioxidant effects. Presently, the available baicalin dosage forms are tablets and capsules for the treatment of acute and chronic hepatitis. Due to its poor water solubility, low lipid solubility, and obvious first pass effect, baicalin has very low oral bioavailability. To improve its solubility and mitigate its first-past effect, baicalin-loaded microemulsion-based gel（B-MEG） was prepared for improving transdermal delivery of baicalin. Methods: The formulation of baicalin-loaded microemulsions（B-ME） was determined by the equilibrium solubility and the pseudo-ternary phase diagram method. With ethyl oleate acting as oil phase, tween 80 as surfactant, and glycerin as cosurfactant, water was added dropwise to the oil phase to prepare microemulsion at room temperature under magnetic stirring. HPLC method was used to determine the content of baicalin in B-ME. Central composite design and split-split plot design were used to screen out and optimize the preparation methods for B-ME and B-MEG. The amount of the gel matrix and the amount and type of the transdermal enhancer were examined to select the appropriate composition of B-MEG. Franz diffusion cell was used to evaluate the microemulsion and gel transdermal profile in vitro. Transmission electron microscopy, atomic force microscopy and laser particle size analyzer were used to characterize the shape and size of B-ME and B-MEG. Xylene-induced mouse ear edema model（model control group, 2 mg/kg compound acetic acid dexamethasone ointment, 25 mg/kg, 100 mg/kg B-MEG and free-baicalin MEG groups, for 4 days） was used to observe the anti-inflammatory effects of B-MEG. Spectro-Microscopy of Living Plant cells

Spectro-microscopy, a combination of fluorescence microscopy with spatially resolved spectroscopic techni- ques, provides new and exciting tools for functional cell biology in living organisms. This review focuses on recent devel- opments in spectro-microscopic applications for the investigation of living plant cells in their native tissue context. The application of spectro-microscopic methods led to the recent discovery of a fast signal response pathway for the brassi- nosteroide receptor BRI1 in the plasma membrane of living plant cells. Moreover, the competence of different plant cell types to respond to environmental or endogenous stimuli was determined in vivo by correlation analysis of different optical and spectroscopic readouts such as fluorescence lifetime （FLT）. Furthermore, a new spectro-microscopic technique, fluorescence intensity decay shape analysis microscopy （FIDSAM）, has been developed. FIDSAM is capable of imaging low- expressed fluorophore-tagged proteins at high spatial resolution and precludes the misinterpretation of autofluorescence artifacts. In addition, FIDSAM provides a very effective and sensitive tool on the basis of F6rster resonance energy transfer （FRET） for the qualitative and quantitative determination of protein-protein interaction. Finally, we report on the quan- titative analysis of the photosystem I and II （PSI/PSII） ratio in the chloroplasts of living Arabidopsis plants at room tem- perature, using high-resolution, spatially resolved fluorescence spectroscopy. With this technique, it was not only possible to measure PSI/PSII ratios, but also to demonstrate the differential competence of wild-type and carbohydrate-deficient plants to adapt the PSI/PSII ratio to different light conditions. In summary, the information content of standard microscopic images is extended by several dimensions by the use of spectro-microscopic approaches. Spectral Response and Photoelectrochemical Properties of Cd1-xZnxSe Films

Cd1-xZnxSe films with different zinc contents are deposited by an electron beam evaporation technique onto glass substrates for applications in solid-state photovoltalc devices. The structural, optical and photoelectrochemical （PEC） properties of Cd1-xZnxSe films are studied. The host material Cd1-xZnxSe is prepared by the physical vapor deposition method of electron beam evaporation technique （PVD： EBE） under a pressure of 1 × 10^-5 mbar. The x-ray diffractogram indicates that these alloy films are polycrystalline in nature, hexagonal structure with strong preferential orientation of the crystallites along （002） direction. The optical properties shows that the band gap Eg varies from 2.08 to 2.84eV as zinc content varies from 0.2 to 0.8. A PEC cell of the configuration n-Cd1-xZnxSe/Na2S-S-NaOH is fabricated and the dynamic current-voltage characteristics in the dark atmosphere have been examined at room temperature. It has been found that both Voc and Iac decrease with the photoelectrode composition x. Efficiency η and fill factor （FF） also show similar variations. The material properties would be altered and excellently controlled by controlling the system composition x.

Phytosphinganine Affects Plasmodesmata Permeability via Facilitating PDLP5-Stimulated Callose Accumulation in Arabidopsis

Plant plasmodesmata (PDs) are specialized channels that enable communication between neighboring cells. The intercellular permeability of PDs, which affects plant development, defense, and responses to stimuli, must be tightly regulated. However, the lipid compositions of PD membrane and their impact on PD permeability remain elusive. Here, we report that the Arabidopsis sld1 sld2 double mutant, lacking sphingolipid long-chain base 8 desaturases 1 and 2, displayed decreased PD permeability due to a significant increase in callose accumulation. PD-located protein 5 (PDLP5) was significantly enriched in the leaf epidermal cells of sld1 sld2 and showed specific binding affinity to phytosphinganine (t18:0), suggesting that the enrichment of t18:0-based sphingolipids in sld1 sld2 PDs might facilitate the recruitment of PDLP5 proteins to PDs. The sld1 sld2 double mutant seedlings showed enhanced resistance to the fungal-wilt pathogen Verticillium dahlia and the bacterium Pseudomonas syringae pv. tomato DC3000, which could be fully rescued in sld1 sld2 pdlp5 triple mutant . Taken together, these results indicate that phytosphinganine might regulate PD functions and cell-to-cell communication by modifying the level of PDLP5 in PD membranes.

Effect of propofol on immune function, stress response and hemodynamics in patients undergoing colorectal cancer surgery

Objective:To investigate the effects of propofol on immune function, stress response and hemodynamics in patients undergoing colorectal cancer ***: 90 patients who were undergoing laparoscopic radical resection of colorectal cancer were randomly divided into propofol group (n=45) and sevoflurane group (n=45). The propofol group was anesthetized with propofol and the control group was given the same dose of sevoflurane. Changes in HR, SBP and DBP were observed before anesthesia, 1 h after induction, and at the end of surgery. The levels of CD19+, NK cells, MDA, SOD, insulin and cortisol were measured before anesthesia, at the end of operation and 24 h after ***: Compared with before anesthesia, the levels of HR, SBP and DBP decreased at 1 h after induction (allPcell ratio decreased at the end of surgery and 24 h after surgery, while MDA level increased at the end of surgery and 24 h after surgery (Pcells were (7.23±0.83)% and (15.17±1.21)% ,which were higher than the sevoflurane group (allPcells in the propofol group were (9.68±0.96) % and (19.58±1.86)%, which were higher than those in the sevoflurane group (all P<0.05). Compared with before anesthesia, MDA levels increased and SOD levels decreased at the end of surgery (P<0.05). At the end of the propofol group, the MDA level was (6.15±0.62) mmol/L, which was lower than that of the sevoflurane group (P<0.05).
