Japanese encephalitis virus(JEV) is a flavivirus transmitted by mosquitoes that causes severe encephalitis in humans and animals. It has been suggested that AXL, a transmembrane protein, can promote the replication of various flaviviruses,such as dengue(DENV), Zika(ZIKV), and West Nile(WNV) viruses. However, the effect of AXL on JEV infection has not yet been determined. In the present study, we demonstrate that AXL is down-regulated after JEV infection in the late stage. JEV NS2B-3 protein specifically interacted with AXL, and promoted AXL degradation through the ubiquitin–proteasome pathway. AXL-degradation increased cell apoptosis by disrupting phosphatidylinositol 3-kinase(PI3 K)/Akt signal transduction. In addition, the degradation of AXL promoted JEV release to supernatant, whereas the virus in the cell lysates decreased. The supplementation of AXL ligand Gas6 inhibited the JEV-mediated degradation of AXL. Altogether,we discover a new function of NS2B-3 during the process of JEV replication, and provide a new insight into the interactions between JEV and cell hosts.

Since late 2011,outbreaks of pseudorabies virus(PRV)have occurred in southern China causing major economic losses to the pig *** previously reported that variant PRV forms and recombination in China could be the source of continued ***,we analyzed samples from intensive pig farms in eastern China between 2017 and 2019,and sequenced the main glycoproteins(gB,gC,gD,and gE)to study the evolution characteristics of *** on the gC gene,we found that PRV variants belong to clade 2 and detected a founder effect during by the PRV *** addition,we detected inter-and intra-clade recombination;in particular,inter-clade recombination in the gB genes of strains FJ-ZXF and FJ-W2,which were recombinant with clade 1 *** also found specific amino-acid changes and positively selected sites,possibly associated with functional *** analysis of the emergence of PRV in China illustrates the need for continuous monitoring and the development of vaccines against specific variants of PRV.

Osteoporosis is a common degenerative metabolic bone disease in caged laying *** egg production mobilizing large amounts of Ca from bone for eggshell formation,consequently leading to Ca deficiency,has been recognized as a critical factor causing osteoporosis in commercial laying *** aim of this study was to examine the effect of Ca deficiency on the function of the gut microbiotaebone axis and related egg production traits and bone health in laying ***-four 48-week-old laying hens were fed a control diet(Control,3.72%)or a low Ca diet(LC,2.04%)for 60 d(n=12).Compared to the Control hens,the LC hens had higher levels of alkaline phosphatase and tartrate resistant acid phosphatase(P<0.05)with lower bone strength,eggshell thickness,and eggshell strength(P<0.05).In addition,the LC hens had higher plasma estradiol concentrations,while having lower concentrations of interleukin-1(IL-1)and *** LC hens also had a lower pH value in the ileum with an increased Ca *** principal co-ordinates analysis showed significantly separate cecal microbiota populations between the Control and LC *** Prevotellaceae_UCG-001,Subdoligranulum,Peptococcus,and Eubacterium_hallii_group(P<0.05)were higher,while the CHKC1001 and Sutterella(P<0.05)were lower at the genus level in the LC *** addition,Prevotellaceae_UCG-001,Subdoligranulum and Eubacter-ium_hallii_group had a negative correlation,while Sutterella was positively correlated with ileal pH *** transcriptome analysis revealed that the low Ca diet caused 20 and 31 genes to be signifi-cantly up-and down-regulated,*** gene expressions of cystic fibrosis transmembrane conductance regulator,solute carrier family 26 member 3 of the anion exchangers,and mitogen-activated protein kinase 12 of pro-inflammatory factors were lower in the LC birds,which was corre-lated with the lower ileal pH *** results suggest that the hens with low Ca diet-induced osteoporosis have an incr

Antimicrobial resistance is undoubtedly one of the greatest global health threats. The emergence of multidrug-resistant(MDR) Gram-positive pathogens, such as methicillin-resistant Staphylococcus aureus(MRSA), vancomycin-resistant Enterococcus faecium(VRE), and β-lactamase-resistant Streptococcus pneumonia, has severely limited our antibiotic arsenal. Numerous ribosome-targeting antibiotics, especially pleuromutilins, oxazolidinones, and streptogramins, are viewed as promising alternatives against aggressive MDR pathogens. In this study, we identified a new adenosine triphosphate(ATP)-binding cassete(ABC)-F family determinant, srp A, in Streptococcus suis(S. suis) by means of a comparative analysis of the whole-genome sequences of tiamulin(TIA)-resistant and TIA-sensitive bacteria. Functional cloning confirmed that the deduced gene can mediate cross-resistance to pleuromutilins, lincosamides, and streptogramin A in S. suis and S. aureus. A sequence alignment revealed that Srp A shares the highest amino acid identity with Vga(E)(36%) and shows canonical characteristics of ABC-F family *** Srp A-ribosome docked compounds, the extended loop region of Srp A approaches the valnemulinbinding pocket in the ribosome peptidyl-transferase center and competes with bound valnemulin. A detailed mutational analysis of the loop residues confirmed that this domain is crucial for Srp A activity,as substitutions or truncations of this region affect the efficiency and specificity of antibiotic *** antibiotics accumulation indicated that Srp A does not act as an efflux pump, while a ribosome binding assay supported the protective effects of Srp A on the ribosome by preventing antibiotic binding as well as displacing bound drugs. These findings clarify the mechanisms underlying resistance to ribosomal antibiotics.

Three immunizing haptens of bisphenol A(BPA), including two new haptens, were used to produce highly sensitive and specific polyclonal antibodies. The spacer arms of haptens for coupling to the protein carrier were located at different positions in BPA, and different length spacer arms were tested. Highly sensitive polyclonal antibodies were obtained and characterized using indirect competitive enzyme-linked immunosorbent assay(ic ELISA). Under optimized conditions, the half maximal inhibitory concentration(IC50) value of the best polyclonal antibody was 2.1 mg·L^(-1), based on coating heterogeneous antigens, and this optimal polyclonal antibody was highly sensitive toward BPA and displayed negligible crossreactivity with bisphenol B and bisphenol E. A sensitive ic ELISA method utilizing the polyclonal antibody was developed for the determination of BPA in milk. In spiked samples(5, 10 and 20 mg·L^(-1)), the recovery ranged from 80% to 102% with a coefficient of variation(CV) value below 15.8%. The limit of detection of ic ELISA was1.95 mg·L^(-1). These results indicate that the ic ELISA method is suitable for the detection of BPA in milk.

Evaluating the efficacy of an attenuated Streptococcus equi ssp. zooepidemicus vaccine produced by multi-gene deletion in pathogenicity island SeseCisland_4

Streptococcus equi ssp. zooepidemicus(SEZ) is a pathogen associated with a wild range of animal species. Frequent outbreaks have occurred in recent years in pigs, horses, goats and dogs which is liable to infect humans. There is a lack of efficient vaccines against this disease and the occurrence of antibiotic resistance may render drug therapies ineffective. In this study, gene deletion mutant(ΔSEZ) in pathogenicity islands SeseCisland_4 was constructed. The mutant ΔSEZ had a 52-fold decrease in 50% lethal dose(LD_(50)) and had less capacity to adhere epithelial cells. Importantly, immunization of mice with attenuated vaccine ΔSEZ at the dose of 10~2 colony-forming units(CFU) mL^(–1) elicited a significant humoral antibody response, with an antibody titer of 1:12 800. Therefore, 10~2 CFU mL^(–1) might be used as the appropriate immune dose for the attenuated vaccine ΔSEZ, which provided mice with efficient protection against virulent SEZ. In addition, the hyperimmune sera against 10~2 CFU m L–1 attenuated vaccine ΔSEZ could confer significant protection against virulent SEZ infection in the passive immunization experiment and exhibited efficient bactericidal activity in the whole blood assay. Meanwhile, no viable bacteria was detected in blood when mice were immunized with ΔSEZ at the dose of 10~2 CFU mL^(–1) via hypodermic injection. Thereafter, the mutant ΔSEZ at the dose of 10~2 CFU mL^(–1) could confer significant protection in mice and had less negative effects on host, which could be an effective attenuated vaccine candidate for the prevention of SEZ.
