LncRNA pol-lnc78 as a ceRNA regulates antibacterial responses via suppression of pol-miR-n199-3p-mediated SARM down-regulation in Paralichthys olivaceus

Long non-coding RNAs(lncRNAs)function as key modulators in mammalian immunity,particularly due to their involvement in lncRNA-mediated competitive endogenous RNA(ceRNA)crosstalk.Despite their recognized significance in mammals,research on lncRNAs in lower vertebrates remains limited.In the present study,we characterized the first immune-related lncRNA(pol-lnc78)in the teleost Japanese flounder(Paralichthys olivaceus).Results indicated that pol-lnc78 acted as a ceRNA for pol-miR-n199-3p to target the sterile alpha and armadillo motif-containing protein(SARM),the fifth discovered member of the Toll/interleukin 1(IL-1)receptor(TIR)adaptor family.This ceRNA network regulated the antibacterial responses of flounder via the Toll-like receptor(TLR)signaling pathway.Specifically,SARM acted as a negative regulator and exacerbated bacterial infection by inhibiting the expression of inflammatory cytokines IL-1βand tumor necrosis factor-α(TNF-α).Pol-miR-n199-3p reduced SARM expression by specifically interacting with the 3’untranslated region(UTR),thereby promoting SARM-dependent inflammatory cytokine expression and protecting the host against bacterial dissemination.Furthermore,pol-lnc78 sponged pol-miR-n199-3p to ameliorate the inhibition of SARM expression.During infection,the negative regulators pol-lnc78 and SARM were significantly down-regulated,while pol-miR-n199-3p was significantly up-regulated,thus favoring host antibacterial defense.These findings provide novel insights into the mechanisms underlying fish immunity and open new horizons to better understand ceRNA crosstalk in lower vertebrates.

New record of the feather star Palaeocomatella hiwia(Echinodermata:Crinoidea:Comatulida)from Zhenbei Seamount in South China Sea:new insights into the taxonomic position of Palaeocomatella

One specimen belonging to the family Comatellinae was collected from the Zhenbei Seamount(332.5–478.2 m)in the South China Sea in July 2022.Based on the morphological characters,the specimen was identified as Palaeocomatella hiwia McKnight,1977.It is first recorded from China Sea and redescribed in detail.This specimen differs from the original description from New Zealand for never showing syzygy at br4+5 or br5+6 on interior and br1+2 on exterior arms.However,it is much conform to the redescription to specimens from Indonesia,with only differences in position of the second syzygy and distalmost pinnule comb.Specimen is deposited in the Institute of Oceanology,Chinese Academy of Sciences.Phylogenetic analyses based on the mitochondrial c oxidase subunit I(COI)and 16S rRNA genes indicated that P.hiwia was nested within the tribe Phanogeniini and clustered with Aphanocomaster pulcher.Furthermore,P.hiwia showed same morphological features in terms of mouth placement,comb location,and number of comb teeth rows as other genera of Phanogeniini.Therefore,we suggest that the genus Palaeocomatella should be put in the tribe Phanogeniini.

Macular corneal dystrophy(MCD)is a progressive,bilateral stromal dystrophic disease that arises from mutations in carbohydrate sulfotransferase 6(CHST6).Corneal transplantation is the ultimate therapeutic solution for MCD patients.Unfortunately,postoperative recurrence remains a significant challenge.We conducted a retrospective review of a clinical cohort comprising 102 MCD patients with 124 eyes that underwent either penetrating keratoplasty(PKP)or deep anterior lamellar keratoplasty(DALK).Our results revealed that the recurrence rate was nearly three times higher in the DALK group(39.13%,9/23 eyes)compared with the PKP group(10.89%,11/101 eyes),suggesting that surgical replacement of the corneal endothelium for treating MCD is advisable to prevent postoperative recurrence.Our experimental data confirmed the robust m RNA and protein expression of CHST6 in human corneal endothelium and the rodent homolog CHST5 in mouse endothelium.Selective knockdown of wild-type Chst5 in mouse corneal endothelium(AC^(siChst5)),but not in the corneal stroma,induced experimental MCD with similar extracellular matrix synthesis impairments and corneal thinning as observed in MCD patients.Mice carrying Chst5 point mutation also recapitulated clinical phenotypes of MCD,along with corneal endothelial abnormalities.Intracameral injection of wild-type Chst5 rescued the corneal impairments in AC^(siChst5)mice and retarded the disease progression in Chst5 mutant mice.Overall,our study provides new mechanistic insights and therapeutic approaches for MCD treatment by highlighting the role of corneal endothelium in MCD development.

A sudden and unprecedented outbreak of molluscan Philine kinglipini occurred in summer 2022 in Jiaozhou Bay,shandong,China,causing substantial damage to local mariculture industry of the Manila clam Ruditapes philippinarum.Although P.kinglipini has been found in many coastal regions of China,the molecular research of P.kinglipini has not been extensively studied,making it difficult to accurately identify and track P.kinglipini samples in field using molecular methods.Samples were collected during the outbreak and their morphological features and molecular sequences were analyzed.Results show that the causative species of the outbreak was P.kinglipini.The mitochondrial genome(mt DNA)of P.kinglipini was constructed for the first time,based on which phylogenetic analysis of the mt DNAs of P.kinglipini and related species in the order Cephalaspidea was carried out.As revealed by metabarcoding analysis of 18S rDNA V4,the seasonal change of P.kinglipini and closely related Philine species was striking with peaks between April and August.Therefore,metabarcoding analysis is applicable tool for monitoring the bloom development of P.kinglipini and related species.This study generated for the first time essential molecular marker sequences and mtDNA of P.kinglipini,which provided a reference for future characterization and monitoring of its outbreaks and for phylogenetic analysis of Philine species.

Heteroatom tuning in agarose derived carbon aerogel for enhanced potassium ion multiple energy storage

The incorporation of heteroatoms into carbon aerogels(cas)can lead to structural distortions and changes in active sites due to their smaller size and electronegativity compared to pure carbon.However,the evolution of the electronic structure from single-atom doping to heteroatom codoping in cas has not yet been thoroughly investigated,and the impact of codoping on potassium ion(K+)storage and diffusion pathways as electrode material remains unclear.In this study,experimental and theoretical simulations were conducted to demonstrate that heteroatom codoping,composed of multiple heteroatoms(O/N/B)with different properties,has the potential to improve the electrical properties and stability of cas compared to single-atom doping.Electronic states near the Fermi level have revealed that doping with O/N/B generates a greater number of active centers on adjacent carbon atoms than doping with O and O/N atoms.As a result of synergy with enhanced wetting ability(contact angle of 9.26°)derived from amino groups and hierarchical porous structure,ON-CA has the most optimized adsorption capacity(−1.62 eV)and diffusion barrier(0.12 eV)of K^(+).The optimal pathway of K^(+)in ON-CA is along the carbon ring with N or O doping.As K^(+)storage material for supercapacitors and ion batteries,it shows an outstanding specific capacity and capacitance,electrochemical stability,and rate performance.Especially,the assembled symmetrical K^(+)supercapacitor demonstrates an energy density of 51.8 Wh kg^(−1),an ultrahigh power density of 443Wkg^(−1),and outstanding cycling stability(maintaining 83.3%after 10,000 cycles in 1M KPF6 organic electrolyte).This research provides valuable insights into the design of highperformance potassium ion storage materials.

Background Schistosoma japonicum is a parasitic flatworm that causes human schistosomiasis,which is a signif-cant cause of morbidity in China,the Philippines and Indonesia.Oncomelania hupensis(Gastropoda:Pomatiopsidae)is the unique intermediate host of S.japonicum.A complete genome sequence of O.hupensis will enable the funda-mental understanding of snail biology as well as its co-evolution with the S.japonicum parasite.Assembling a high-quality reference genome of O.hupehensis will provide data for further research on the snail biology and controlling the spread of S.japonicum.Methods The draft genome was de novo assembly using the long-read sequencing technology(PacBio Sequel I)and corrected with llumina sequencing data.Then,using Hi-C sequencing data,the genome was assembled at the chromosomal level.CAFE was used to do analysis of contraction and expansion of the gene family and CodeML module in PAML was used for positive selection analysis in protein coding sequences.Results A total length of 1.46 Gb high-quality O.hupensis genome with 17 unique full-length chromosomes(2n=34)of the individual including a contig N50 of 1.35 Mb and a scaffold N50 of 75.08 Mb.Additionally,95.03%of these con-tig sequences were anchored in 17 chromosomes.After scanning the assembled genome,a total of 30,604 protein-coding genes were predicted.Among them,86.67%were functionally annotated.Further phylogenetic analysis revealed that O.hupensis was separated from a common ancestor of Pomacea canaliculata and Bellamya purificata approximately 170 million years ago.Comparing the genome of O.hupensis with its most recent common ancestor,it showed 266 significantly expanded and 58 significantly contracted gene families(P<0.05).Functional enrichment of the expanded gene families indicated that they were mainly involved with intracellular,DNA-mediated transposi-tion,DNA integration and transposase activity.Conclusions Integrated use of multiple sequencing technologies,we have successfully constructed the genome at the chromosomal-level of O

The Jiangchuan Biota from the Jiucheng Member(Mb.)of the Dengying Formation(Fm.),discovered in Jiangchuan,eastern Yunnan,China,is marked by copious macrofossils at the apex of the Ediacaran strata.This fauna features benthic algae with varied holdfasts and other fossils of indeterminate taxonomic affinity and is compositionally unique compared to the Shibantan and Gaojiashan biotas of the Dengying Fm.and the Miaohe and Wenghui biotas of the Doushantuo Fm.,elsewhere in China.One novel benthic saccular macroalgal fossil,named here Houjiashania yuxiensis gen.and sp.nov.,from the Jiangchuan Biota is based on fossils that are sausage-shaped,elongate,tubular,ranging from 0.3 to 4 cm in length,and up to 0.8 cm in diameter.One terminus is blunt and rounded to an obtuse angle,the other is bent with a spread-out surface resembling a holdfast,suggesting a three-dimensional thallus.Thin,stipe-shaped outgrowths,likely vestiges of sessile saccular life forms,are prevalent in macroalgal fossils of analogous size and shape,as well as present brown algae Scytosiphonaceae,such as Colpomenia and Dactylosiphon.The new findings augment the diversity of benthic algae,such as those known from the Early Neoproterozoic Longfengshan Biota in North China.The benthic algal macrofossils in the Jiucheng Mb.add to knowledge of Late Ediacaran metaphyte diversification and offer more clues about the evolutionary positioning of primitive macroalgae.The co-occurrence of numerous planktonic and benthic multicellular algae and planktonic microbes might have facilitated ecologically the more extensive later Cambrian explosion evidenced by the Chengjiang Biota in Yunnan.

Correlative species distribution models(SDMs)are important tools to estimate species’geographic distribution across space and time,but their reliability heavily relies on the availability and quality of occurrence data.Estimations can be biased when occurrences do not fully represent the environmental requirement of a species.We tested to what extent species’physiological knowledge might influence SDM estimations.Focusing on the Japanese sea cucumber Apostichopus japonicus within the coastal ocean of East Asia,we compiled a comprehensive dataset of occurrence records.We then explored the importance of incorporating physiological knowledge into SDMs by calibrating two types of correlative SDMs:a naïve model that solely depends on environmental correlates,and a physiologically informed model that further incorporates physiological information as priors.We further tested the models’sensitivity to calibration area choices by fitting them with different buffered areas around known presences.Compared with naïve models,the physiologically informed models successfully captured the negative influence of high temperature on A.japonicus and were less sensitive to the choice of calibration area.The naïve models resulted in more optimistic prediction of the changes of potential distributions under climate change(i.e.,larger range expansion and less contraction)than the physiologically informed models.Our findings highlight benefits from incorporating physiological information into correlative SDMs,namely mitigating the uncertainties associated with the choice of calibration area.Given these promising features,we encourage future SDM studies to consider species physi-ological information where available.

Cyanea nozakii,a common jellyfish distributed in offshore China,has a complex trophic relationship with other zooplankton groups.However,few studies have reported the predation rates and prey selection patterns of C.nozakii medusae on different prey items.Research is also lacking on the intraguild predation of Aurelia coerulea(another common bloom jellyfish in offshore China)by C.nozakii.To address the knowledge gaps,the clearance rates of C.nozakii for different prey items,including copepods(small1000μm),fish larvae,and gelatinous prey(hydromedusae,A.coerulea ephyrae,and chaetognaths),were measured.The influence of predator size on the clearance rate was also determined.Additionally,we examined the intraguild predation of C.nozakii on A.coerulea medusae.The clearance rates of C.nozakii varied widely with prey organisms,being independent of prey concentrations.Gelatinous organisms,except for chaetognaths,were captured with considerably high efficiency,followed by fish larvae and copepods,indicating the preferential prey selection of gelatinous organisms by C.nozakii.The clearance rate increased linearly with the cross-sectional area of C.nozakii.Body size in medusae may,to some extents,underpin their capacity to capture more prey by increasing the encounter rate and capture success through ontogeny.C.nozakii preyed voraciously on A.coerulea in high feeding efficiency,but the clearance rate decreased with increasing A.coerulea(as prey)size.This phenomenon of intraguild predation suggests a speculative hypothesis of potential population regulation of A.coerulea by C.nozakii.The information regarding the feeding ecology of C.nozakii reported in this study is important for understanding plankton dynamics in marine ecosystems with extensive occurrences of this jellyfish.

The D-quadrant organizer sets up the dorsal–ventral(DV)axis and regulates mesodermal development of spiralians.Studies have revealed an important role of mitogen-activated protein kinase(MAPK)signaling in organizer function,but the related molecules have not been fully revealed.The association between fibroblast growth factor receptor(FGFR)and MAPK signaling in regulating organizer specification has been established in the annelid Owenia fusiformis.Now,comparable studies in other spiralian phyla are required to decipher whether this organizer-inducing function of FGFR is prevalent in Spiralia.Here,we indicate that treatment with the FGFR inhibitor SU5402 resulted in deficiency of organizer specification in the mollusk Lottia peitaihoensis.Subsequently,the bone morphogenetic protein(BMP)signaling gradient and DV patterning were disrupted,suggesting the roles of FGFR in regulating organizer function.Changes in multiple aspects of organizer function(the morphology of vegetal blastomeres,BMP signaling gradient,expression of DV patterning markers,etc.)indicate that these developmental functions have different sensitivities to FGFR/MAPK signaling.Our results reveal a functional role of FGFR in organizer specification as well as DV patterning of Lottia embryos,which expands our knowledge of spiralian organizers.
