Succinylation modification:a potential therapeutic target in stroke

Stroke is a leading cause of mortality and disability *** cell death triggered by the compromised supply of blood oxygen and glucose is one of the major pathophysiology of strokeinduced brain *** mitochondrial energy metabolism is observed minutes after stroke and is closely associated with the progression of ***,a new type of posttranslational modification,known as lysine succinylation,has been recognized to play a significant role in mitochondrial energy metabolism after ***,the role of succinylation modification in cell metabolism after stroke and its regulation are not well *** aimed to review the effects of succinylation on energy metabolism,reactive oxygen species generation,and neuroinflammation,as well as Sirtuin 5 mediated desuccinylation after *** also highlight the potential of targeting succinylation/desuccinylation as a promising strategy for the treatment of *** succinylation level is dynamically regulated by the nonenzymatic or enzymatic transfer of a succinyl group to a protein on lysine residues and the removal of succinyl catalyzed by *** evidence has suggested that succinylation can regulate the metabolic pathway through modulating the activity or stability of metabolic ***,especially Sirtuin 5,are characterized for their desuccinylation activity and have been recognized as a critical regulator of metabolism through desuccinylating numerous metabolic *** between succinylation and desuccinylation has been implicated in the pathophysiology of *** agents that enhance the activity of Sirtuin 5 have been employed to promote desuccinylation and improve mitochondrial metabolism,and neuroprotective effects of these agents have been observed in experimental stroke ***,their therapeutic efficacy in stroke patients should be validated.

Obeticholic acid(OCA),a farnesoid X receptor(FXR)agonist with favorable effects on fatty and glucose metabolism,has been considered the leading candidate drug for nonalcoholic steatohepatitis(NASH)***,its limited effectiveness in resolving liver fibrosis and lipotoxicity-induced cell death remains a major ***,a newly recognized form of cell death characterized by uncontrolled lipid peroxidation,is involved in the progression of *** oxide(NO)is a versatile biological molecule that can degrade extracellular *** this study,we developed a PEGylated thiolated hollow mesoporous silica nanoparticles(MSN)loaded with OCA,as well as a ferroptosis inhibitor liproxsatin-1 and a NO donor S-nitrosothiol(ONL@MSN).Biochemical analyses,histology,multiplexed flow cytometry,bulk-tissue RNA sequencing,and fecal 16S ribosomal RNA sequencing were utilized to evaluate the effects of the combined nanoparticle(ONL@MSN)in a mouse NASH *** with the OCA-loaded nanoparticles(O@MSN),ONL@MSN not only protected against hepatic steatosis but also greatly ameliorated fibrosis and ***@MSN also displayed enhanced therapeutic actions on the maintenance of intrahepatic macrophages/monocytes homeostasis,inhibition of immune response/lipid peroxidation,and correction of microbiota *** findings present a promising synergistic nanotherapeutic strategy for the treatment of NASH by simultaneously targeting FXR,ferroptosis,and fibrosis.

Protective effect of hydrogen-rich saline on ischemia/reperfusion injury in rat skin flap

Objective: Skin damage induced by ischemia/reperfusion (I/R) is a multifactorial process that often occurs in plastic surgery. The mechanisms of I/R injury include hypoxia, inflammation, and oxidative damage. Hydrogen gas has been reported to alleviate cerebral I/R injury by acting as a free radical scavenger. Here, we assessed the protective effect of hydrogen-rich saline (HRS) on skin flap I/R injury. Methods: Abdominal skin flaps of rats were elevated and ischemia was induced for 3 h; subsequently, HRS or physiological saline was administered intraperitoneally 10 min before reperfusion. On postoperative Day 5, flap survival, blood perfusion, the accumulation of reactive oxygen species (ROS), and levels of cytokines were evaluated. Histological examinations were performed to assess inflammatory cell infiltration. Results: Skin flap survival and blood flow perfusion were improved by HRS relative to the controls. The production of malondialdehyde (MDA), an indicator of lipid peroxidation, was markedly reduced. A multiplex cytokine assay revealed that HRS reduced the elevation in the levels of inflammatory cytokines, chemokines and growth factors, with the exception of RANTES (regulated on activation, normal T-cell expressed and secreted) growth factor. HRS treatment also reduced inflammatory cell infiltration induced by I/R injury. Conclusions: Our findings suggest that HRS mitigates I/R injury by decreasing inflammation and, therefore, has the potential for application as a therapy for improving skin flap survival.

A new neuroprotective bakkenolide from the rhizome of Petasites tricholobus

A new bakkenolide-type compound, bakkenolide-Va（1）, was isolated from the rhizome of Petasites tricholobus. The structure was characterized on the basis of various NMR （1H, 13C, 1H-1H COSY, HMQC, HMBC and NOESY） and mass spectrometry data. The neuroprotective activity of bakkenolide-Va was investigated in primarily cultured neurons treated by hypoxia-ghicose deprivation and various types of oxidative insults.

Exercise induces tissue hypoxia and HIF-1α redistribution in the small intestine

Background:Exercise induces blood flow redistribution among tissues,leading to splanchnic *** epithelial cells are positioned between the anaerobic lumen and the highly metabolic lamina propria with an oxygen ***-inducible factor(HIF)-la is pivotal in the transcriptional response to the oxygen ***:In this study,the pimonidazole hydrochloride staining was applied to observe the tissue hypoxia in different organs,which might be affected by the blood flow *** HIF-la luciferase reporter ROSA26 oxygen-dependent degradation domain(ODD)-Luc/^+mouse model(ODD domain-Luc;female,n=3-6/group) was used to detect the HIF-la expression in the *** used 3 swimming models:moderate exercise for 30 min,heavy-intensity exercise bearing 5% bodyweight for 1.5 h,and long-time exercise for 3 ***:We found that 1 session of swimming at different intensities could induce tissue hypoxia redistribution in the small intestine,colon,liver and kidney,but not in the spleen,heart,and skeletal *** data showed that exercise exacerbated the extent of physiological hypoxia in the small ***,using ODD-Luc mice,we found that moderate exercise increased the in vivo HIF-1α level in the small *** postexercise HIF-1α level was gradually decreased in a time-dependent ***,the redistribution of tissue hypoxia and the increase of HIF-la expression were not related to the exercise intensity and ***:This study provided evidence that the small intestine is the primary target organ for exercise-induced tissue hypoxia and HIF-la redistribution,suggesting that HIF-1α may be a potential target for the regulation of gastrointestinal functions after exercise.

Changes of Immunoreactive β-Endorphin in Plasma, Pituitary and Hypothalamus of Rats during Oxygen-Induced Convulsions

We observed for the first time the differences of immunoreactive β-endorphin(IR -β- EP) content in plasma, pituitary and hypothalamus of rats under various conditionsusing radioimmunoassay (RIA) and the effects of naloxone and β - endorphin (β- EP) antiserumon initial time of convulsions (ITC), severity of convulsions(SOC) and mortality on surface(MOS) of rats to hyperbaric oxygen(HBO). The results suggest thatβ- EP may partici-pate in the course of oxygen - induced convulsions and be one of endogenous convulsion - causingagents.

Lung macrophages are involved in lung injury secondary to repetitive diving

This study aimed to establish an animal model of decompression-induced lung injury(DILI)secondary to repetitive diving in mice and explore the role of macrophages in DILI and the protective effects of high-concentration hydrogen(HCH)on *** were divided into three groups:control group,DILI group,and HCH *** were exposed to hyperbaric air at 600 k Pa for 60 min once daily for consecutive 3 d and then experienced *** HCH group,mice were administered with HCH(66.7%hydrogen and 33.3%oxygen)for 60 min after each hyperbaric *** function tests were done 6 h after decompression;the blood was harvested for cell counting;the lung tissues were harvested for the detection of inflammatory cytokines,hematoxylin and eosin(HE)staining,and immunohistochemistry;western blotting and polymerase chain reaction(PCR)were done for the detection of markers for M1 and M2 *** results showed that bubbles formed after decompression and repeated hyperbaric exposures significantly reduced the total lung volume and functional residual ***,repetitive diving dramatically increased proinflammatory factors and increased the markers of both M1 and M2 *** inhalation improved lung function to a certain extent,and significantly reduced the pro-inflammatory *** effects were related to the reduction of M1 macrophages as well as the increase in M2 *** study indicates that repetitive diving damages lung function and activates lung macrophages,resulting in lung *** inhalation after each diving may be a promising strategy for the prevention of DILI.
