Leaf-color mutations have been studied extensively in ***,to better understand the complex mechanisms underlying the formation of leaf color,it is essential to continue discover novel genes involved in the process of leaf color *** this study,we identified a variegated-leaf(vg)mutant in tomato that exhibited defective phenotypes in thylakoids and *** clone the vg locus,an F2population was constructed from the cross between the vg mutant(Solanum lycopersicum)and the wild tomato LA1589(***).Using the map-based cloning approach,the vg locus was mapped on chromosome 7 and narrowed down to a 128 kb region that contained 21 open reading frames(ORFs).The expression levels of ORF9,ORF10,and ORF13 were significantly lower in vg than in the wild-type plants,while the ORF11 transcript level was elevated in *** then mutagenized ORF9,ORF10,and ORF13 by the CRISPR/Cas9 system in the wild-type tomato background and found that only the ORF10 mutation reproduced the phenotype of variegated leaves,indicating that ORF10 represents VG and its down-regulated expression was responsible for the variegated leaf ***10 encodes a thylakoid formation protein and its mutant lines showed reduced levels of chlorophyll synthesis and *** together,these results suggest that VG is necessary for chloroplast development,chlorophyll synthesis,and photosynthesis in tomato.

Persistent luminescence nanoparticles (PLNPs) are a series of emerging luminescent nanomaterials which can emit persistently after ceasing the external excitation. Due to the long decay time of persistent luminescence, the background autofluorescence in complex sample and tissues can be effectively eliminated, thus significantly improving the sensitivity of bioanalysis. Besides, such a long decay time of luminescence also makes PLNPs valuable for long-term bioimaging. Benefiting from these merits, PLNPs have been widely used for biomedical applications, especially biosensing and bioimaging. In this review,we conclude the progress in the application of PLNPs at biosensing and bioimaging in recent years, and also provide our understanding of the prospects.

The achievement of a functional cure for chronic hepatitis B(CHB)remains limited to a minority of patients treated with currently approved *** primary objective in developing new anti-HBV drugs is to enhance the functional cure rates for CHB.A critical prerequisite for the functional cure of CHB is a substantial reduction,or even eradication of covalently closed circular DNA(cccDNA).Within this context,the changes in cccDNA levels during treatment become as a pivotal *** have previously analyzed the factors influencing cccDNA dynamics and introduced a preliminary classification of hepatitis B treatment strategies based on these *** this review,we employ a systems thinking perspective to elucidate the fundamental aspects of the HBV replication cycle and to rationalize the classification of treatment strategies according to their impact on the dynamic equilibrium of *** upon this foundation,we categorize current anti-HBV strategies into two distinct groups and advocate for their combined use to significantly reduce cccDNA levels within a well-defined timeframe.

Formins are well-known regulators that participate in the organization of the actin cytoskeleton in organisms. The Arabidopsis thaliana L. genome encodes 21 formins, which can be divided into two distinct subfamilies. However, type II formins have to date been less well characterized. Here, we cloned a type II formin, AtFH16, and characterized its biochemical activities on actin and microtubule dynamics. The results show that the FH1 FH2 structure of AtFH16 cannot nucleate actin polymerization efficiently, but can bind and bundle microfilaments. AtFH16 FHIFH2 is also able to bind and bundle microtubules, and preferentially binds microtubules over microfilaments in vitro, in addition, AtFH16 FHIFH2 co-localizes with microtubules in onion epidermal cells, indicating a higher binding affinity of AtFH16 FHIFH2 for microtubules rather than microfilaments in vivo. In conclusion, AtFH16 is able to interact with both microfilaments and microtubules, suggesting that AtFH16 probably functions as a bifunctional protein, and may thus participate in plant cellular processes.

Knee osteoarthritis(OA)is a debilitating condition with limited long-term treatment *** therapeutic potential of mesenchymal stem cells(MSCs),particularly those derived from bone marrow aspirate concentrate,has garnered attention for cartilage repair in *** the iliac crest is the traditional site for bone marrow harvesting(BMH),associated morbidity has prompted the exploration of alternative sites such as the proximal tibia,distal femur,and proximal *** paper reviews the impact of different harvesting sites on mesenchymal stem cell(MSC)yield,viability,and regenerative potential,emphasizing their relevance in knee OA *** iliac crest consistently offers the highest MSC yield,but alternative sites within the surgical field of knee procedures offer comparable MSC characteristics with reduced *** integration of harvesting techniques into existing knee surgeries,such as total knee arthroplasty,provides a less invasive approach while maintaining thera-peutic ***,variability in MSC yield from these alternative sites underscores the need for further research to standardize techniques and optimize clinical *** directions include large-scale comparative studies,advanced characterization of MSCs,and the development of personalized harvesting ***,the findings suggest that optimizing the site of BMH can significantly influence the quality of MSC-based therapies for knee OA,enhancing their clinical utility and patient outcomes.

Lanthanide-doped upconversion nanoparticles （UCNPs） offer unique advantages in term of low autofluorescence, high signal-to-noise ratio and deep tissue penetration, and have attracted considerable attention in biomedical applications. DNA, beyond the properties of self-assembly, also exhibits multiple functions such as molecular recognition, drug loading capacity and therapeutic effect. In this regard, the combination of UCNPs and DNA offers a promising and powerful platform for potential applications in biosensing, bioimaging and disease therapy. In this review, we mainly introduce recent progresses of DNA-functionalized upconversion materials, providing an overview of the design and applications in biosensing, bioimaging and therapy. The challenges and future perspectives are also discussed, aiming to promote their applications in material science and biomedicine.

The mangrove rivulus Kryptolebias marmoratus a small cyprinodont fish native to tropical and subtropical waters from Florida to Brazil, is one of two known self-fertilizing, hermaphroditic vertebrates of which K. marmoratus displays androdioecy, a complex system of reproduction in which hermaphrodites and males are present. This study describes the behavioral repertoires observed during dyadic interactions in the laboratory. Kryptolebias marmoratus exhibited 23 distinctive acts or behaviors. Acts were divided into four categories： aggressive, submissive, neutral, and reproductive. Leading and following behaviors played important roles in the behavioral repertoires of these fish. In hermaphrodite-male dyads, males exclusively initiated the reproductive process and actively pursued hermaphrodites. When hermaphrodites were paired, there was no evidence that they behaved like other simultaneous hermaphrodites that alternate sexual roles （e.g. serranids）. Hermaphrodites were extremely aggressive towards one another, and the aggressor established dominance rapidly. Male-male dyads were divided into two subdivisions based on the presence or absence of the caudal ocellus on one fish. A caudal ocellus on one male appeared to signal the possibility of a potential mating partner to males lacking it. Pairings of males without an ocellus were similar to hermaphrodite-hermaphrodite dyads in that both members of the pair were aggressive towards one another. These observations may be indicative of interactions taking place in natural communities or assemblages of fish in which both males and hermaphrodites occur and provide evidence on the role of dyadic interactions in the mixed-mating strategies [Current Zoology 56 （1）： 6-17, 2010].

A new crystal of N,N＇-bis（4-nitrophenylcarbamothioyl）isophthalamide DMF solva-te has been prepared at room temperature and characterized by elemental analysis and IR,MS,1H NMR spectra and X-ray single-crystal *** complex crystallizes in monoclinic,space group P21/c with a = 11.2093（12）,b = 22.081（2）,c = 13.9640（15） ,β= 112.128（2）°,V = 3201.8（6） 3,C28H30N8O8S2,Mr = 670.72,Dc = 1.391 Mg/m3,Z = 4,μ= 0.228 mm-1,F（000） = 1400,the final R = 0.0483 and wR = 0.1411（I 2σ（I））.The molecular packing in the crystal is the result of N-H…O hydrogen bonding.

The crystal structure of the title compound（C24H22N2O5, Mr = 418.44） has been determined by single-crystal X-ray diffraction. The crystal is of orthorhombic, space group P21/c with a = 7.751（2）, b = 12.392（4）, c = 21.326（6）, V = 2048.4（1）3, Z = 4, Dc = 1.357 g/cm3, F（000） = 880, μ（MoKα） = 0.096 mm-1, the final R = 0.0731 and wR = 0.1982 for 3335 observed reflections（I 〉 2σ（I））. Intermolecular C（24）–H（24B）···O（1） hydrogen bond is observed in the structure.
