Fluorescence confocal laser-scanning microscopy(LSM)is one of the most popular tools for life science *** popularity is expected to grow thanks to single-photon array detectors tailored for *** detectors offer unique single-photon spatiotemporal information,opening new perspectives for gentle and quantitative superresolution ***,a flawless recording of this information poses significant challenges for the microscope data acquisition(DAQ)*** present a DAQ module based on the digital frequency domain principle,able to record essential spatial and temporal features of *** use this module to extend the capabilities of established imaging techniques based on single-photon avalanche diode(SPAD)array detectors,such as fluorescence lifetime image scanning ***,we use the module to introduce a robust multispecies approach encoding the fluorophore excitation spectra in the time ***,we combine time-resolved stimulated emission depletion microscopy with image scanning microscopy,boosting spatial *** results demonstrate how a conventional fluorescence laser scanning microscope can transform into a simple,information-rich,superresolved imaging system with the simple addition of a SPAD array detector with a tailored data acquisition *** expected a blooming of advanced single-photon imaging techniques,which effectively harness all the sample information encoded in each photon.

Highly accurate biosensors for few or single molecule detection play a central role in numerous key fields,such as healthcare and environmental *** the last decade,laser biosensors have been investigated as proofs of concept,and several technologies have been *** here propose a demonstration of polymeric whispering gallery microlasers as biosensors for detecting small amounts of proteins,down to 400 *** have the advantage of working in free space without any need for waveguiding for input excitation or output signal *** photonic microsensors can be easily patterned on microscope slides and operate in air and *** estimate the limit of detection up to 148 nm/RIU for three different protein *** addition,the sensing ability of passive spherical resonators in the presence of dielectric nanoparticles that mimic proteins is described by massive ab initio numerical simulations.

Understanding the processes involved in how and why animals communicate has long been fascinating to scientists (Darwin 1871). This endeavor plays a key role in the pursuit of reaching a better comprehension both of the rules involved in organizing the animal societies around us (Freeberg et al. 2012) and of the evolutionary bases underlying our own vocal communication system: human language (Fitch 2010). Moreover, in recent years, the type and function of information conveyed by animal vocalizations have also attracted research interest in the field of ecology (see Sueur and Farina 2015). While these fields certainly still have room for developing new concepts and methods in order to improve our appreciation of animal vocal communication, to date, 3 main approaches have been applied which have greatly contributed to advance our knowledge on the question.

Ligninolytic Peroxidase-Like Activity of a Synthetic Metalloporphine Immobilized onto Mercapto-Grafted Crosslinked PVA Inspired by the Active Site of Cytochrome P450

A synthetic metalloporphine was immobilized onto a PVA-based and mercapto-grafted solid support, emulating the active site of cytochrome P450. Its ligninolytic peroxidase-like catalytic activity was studied. The coordinated mercapto ligand significantly affected the catalytic features of the catalyst because the oxidation of lignin-model compounds was very slow by comparison with imidazoleand pyridine-coordinated immobilized metalloporphines. Conversely, the catalyst efficiently bleached several industrial dyes and thus demonstrated promising activity for this application. Based on this altered substrate specificity the oxygen-donor catalytic route seems to be more favorable than a single electron oxidation pathway.

Field and Numerical Tests of the Two-Ponding Depth Procedure for Analysis of Single-Ring Pressure Infiltrometer Data

The two-ponding depth(TPD)analysis procedure of single-ring infiltrometer data can yield invalid results,i.e.,negative values of the field-saturated soil hydraulic conductivity or the matric flux potential,denoting failure of the two-level *** objective of this study was to test the performance of the TPD procedure in analyzing the single-ring infiltrometer data of diferent types of soils.A field investigation carried out in western Sicily,Italy,yielded higher failure rates(40%)in two clay loam soils than in a sandy loam soil(25%).A similar result,i.e.,fine-textured soils yielding higher failure rates than the coarse-textured one,was obtained using numerically simulated infiltration *** heterogeneity and reading errors were suggested to be factors determining invalid results in the *** the numerical data,allowing a less generic definition of soil heterogeneity,invalid TPD results were occasionally obtained with the simultaneous occurrence of a high random variation(standard deviation≥0.5)and a well developed structural correlation for saturated hydraulic conductivity(correlation length≥20 cm).It was concluded that a larger number of replicated runs should be planned to characterize fine-textured soils,where the risk to obtain invalid results is relatively *** rings should be used since they appeared more appropriate than the small ones to capture and average soil *** simulation appeared suitable for developing improved strategies of soil characterization for an area of interest,which should also take into account macropore effects.

Originally identified as allelochemicals involved in plant-parasite interactions, more recently, Strigolactones （SLs） have been shown to play multiple key roles in the rhizosphere communication between plants and mycorrhizal fungi. Even more recent is the hormonal role ascribed to SLs which broadens the biological impact of these relatively simple molecules. In spite of the crucial and multifaceted biological role of SLs, there are no data on the receptor（s） which bind（s） such active molecules, neither in the producing plants nor in parasitic weeds or AM fungi. Information about the putative receptor of SLs can be gathered by means of structural, molecular, and genetic approaches. Our contribution on this topic is the design and synthesis of fluorescent labeled SL analogs to be used as probes for the detection in vivo of the receptor（s）. Knowledge of the putative receptor structure will boost the research on analogs of the natural substrates as required for agricultural applications.

Entanglement distribution between distant parties is one of the most important and challenging tasks in quantum *** of photonic entangled states using optical fiber links is a fundamental building block toward quantum *** the different degrees of freedom,orbital angular momentum(OAM)is one of the most promising due to its natural capability to encode high dimensional quantum *** experimentally demonstrate fiber distribution of hybrid polarization-vector vortex entangled photon *** this end,we exploit a recently developed air-core fiber that supports OAM *** fidelity distribution of the entangled states is demonstrated by performing quantum state tomography in the polarization-OAM Hilbert space after fiber propagation and by violations of Bell inequalities and multipartite entanglement *** results open new scenarios for quantum applications where correlated complex states can be transmitted by exploiting the vectorial nature of light.

Female indris determine the rhythmic structure of the song and sustain a higher cost when the chorus size increases

Among the behavioral traits shared by some nonhuman primate species and humans there is singing. Unfortunately, our understanding of animals' rhythmic abilities is still in its infancy. Indris are the only lemurs who sing and live in monogamous pairs, usually forming a group with their offspri ng. All adult members of a group usually participate in choruses that are emitted regularly and play a role in advertising territorial occupa ncy and in tergroup spaci ng. Males and females emit phrases that have similar frequency ranges but may differ in their temporal structure. We examined whether the individuals' contribution to the song may change according to chorus size, the total duration of the song or the duration of the individual con tribution using the in ter-on set intervals within a phrase and between phrases. We found that the rhythmic structure of indri's songs depends on factors that are different for males and females. We showed that females have sigrdficantly higher variation in the rhythm of their contribution to the song and that, changes according to chorus size. Our findings indicate that female indris sustain a higher cost of singing than males whe n the nu mber of singers in creases. These results suggest that cross-species investigatio ns will be crucial to un derstanding the evoluti on ary frame in which such sexually dimorphic traits occurred.

Proteinase activated-receptors-associated signaling in the control of gastric cancer

Gastric cancer(GC)is the fourth most common cancer in the world and the second cause of cancer-related *** carcinogenesis is a multifactorial process,in which environmental and genetic factors interact to activate multiple intracellular signals thus leading to uncontrolled growth and survival of GC *** such a pathway is regulated by proteinase activated-receptors(PARs),seven transmembrane-spanning domain G protein-coupled receptors,which comprise four receptors(i.e.,PAR-1,PAR-2,PAR-3,and PAR-4)activated by various *** PAR-1 and PAR-2 are over-expressed on GC cells and their activation triggers and/or amplifies intracellular pathways,which sustain gastric *** is also evidence that expression of either PAR-1 or PAR-2 correlates with depth of wall invasion and metastatic dissemination and inversely with the overall survival of ***,data emerging from experimental models of GC suggest that both these receptors can be important targets for therapeutic interventions in GC *** contrast,PAR-4levels are down-regulated in GC and correlate inversely with the aggressiveness of GC,thus suggesting a negative role of this receptor in the control of *** this article we review the available data on the expression and role of PARs in GC and discuss whether manipulation of PAR-driven signals may be useful for interfering with GC cell behavior.

Global optimization of protein-peptide docking by a filling function method

Molecular docking programs play a crucial role in drug design and development. In recent years, much attention has been devoted to the protein-peptide docking problem in which docking of a flexible peptide with a known protein is sought. In this work we present a new docking algorithm which is based on the use of a filling function method for continuos constrained global optimization. Indeed, the protein-peptide docking position is sought by minimizing the conformational potential energy subject to constraints necessary to maintain the primary sequence of the given peptide. The resulting global optimization problem is difficult mainly for two reasons. First, the problem is large scale in constrained global optimization;second, the energy function is multivariate non-convex so that it has many local minima. The method is based on the device of modifying the original objective function once a local minimum has been attained by adding to it a filling term. This allows the overall algorithm to escape from local minima thus, ultimately, giving the algorithm ability to explore large regions in the peptide conformational space. We present numerical results on a set of benchmark docking pairs and comparison with the well-known software package for molecular docking PacthDock.
