Biological synthesis of silver and gold nanoparticles using Costus pictus leaf extract （CPLE） and their potential in vitro antioxidant and catalytic activities were reported here. Formation of Costus pictus silver （CPAgNPs） and gold （CPAuNPs） nanoparticles was confirmed by UV-visible spectroscopy and their spherical shape by scanning electron microscopy. The synthesized nanoparticles gave strong signals for silver and gold in energy dispersive X-ray spectroscopy. The CPAgNPs and CPAuNPs had an average size of 46.7 and 37.2 nm, respectively, as determined by dynamic light scattering particle size analyzer. Fourier transform infrared spectroscopy （FTIR） analysis indicated involvement of amine and carbonyl groups in the formation of CPAgNPs and CPAuNPs. Thermal stability of synthesized nanoparticles was assessed by thermogravimetric analysis-differential scanning calorimetry. CPAgNPs, CPAuNPs and CPLE exhibited tremendous antioxidant activity when being assessed by various in vitro assays, and their activity was comparable to standard antioxidants. CPAgNPs, CPAuNPs and CPLE also caused degradation of dyes like methylene blue and methyl red. While CPAgNPs, CPAuNPs and CPLE caused respective 85%, 42% and 30% degradation of methylene blue, they showed less activity against methyl red. These observations signify that such green methods open up new avenues in nanobiotechnology for the synthesis of nanoparticles with extensive industrial and biomedical applications.

Collection and preservation of urinary proteins, using a fluff pulp diaper

Change is the most fundamental property of a biomarker. In contrast to the blood, which is under homeostatic controls, urine reflects changes in the body earlier and is more sensitive, thus making it a better biomarker source. Moreover, drawing blood from infants and toddlers is difficult and not tolerated well. For patients limited by language, communicating their chief complaint is difficult. Thus, monitoring biomarkers in urine can provide valuable clues for the diagnosis of diseases, especially pediatric diseases. Collecting urine from young children and some adult patients is more challenging than collecting it from healthy ***, we propose a method that uses a fluff pulp diaper to collect urine. Urinary proteins are then eluted and adsorbed onto a piece of nitrocellulose membrane, which can be dried and stored in a vacuum bag. SDS-PAGE and LC-MS/MS analysis indicated that this method is reproducible, and similar proteins were identified as those obtained by an acetone precipitation method. With this simple and economical method, it is possible to collect and preserve urine samples from infants, toddlers, and patients with special needs, even for large-scale biomarker studies.

Long Non-coding RNAs （lncRNAs）, usually derived from intergenic regions or introns, play distinct and important roles in diverse biological processes. Up to now, many types of non-coding RNAs have been identified across spe- cies. They are highly heterogeneous in their lengths, struc- tures, and functions. Aberrant regulation of lncRNAs has been shown to be associated with human diseases, such as cancer, but the mechanisms have remained largely unknown （Rinn and Chang 2012）. A recent study from Mendell and colleagues published in Cell dissected a novel pathway in which a specific IncRNA, termed NORAD, protects cells from becoming aneuploid （Lee et al., 2016）, which is known to drive the occurrence and development of cancer, among other diseases.

Adinbitor was cloned from Agkistrodon halys brevicaudus stejneger and characterized as a novel disintegrin. In this study, total RNA was extracted from venom gland and used in RT-PCR to generate a cDNA which is 219 bp long. The sequence encoded a polypeptide composed of 73 amino acids, including 12 cysteines, an RGD motif, and the signature motif of disintergrin. Recombinant Adinbitor (rAdinbitor) was expressed in E. coli and purified by using the His?Bind affinity chromatography. The IC50 for inhibiting human platelet aggregation and bFGF-induced proliferation of ECV304 cells was 6 μΜ and 0.89 μΜ respectively. Furthermore, Adinbitor significantly inhibited angiogenesis both in vivo and in vitro. Taken together, these results suggested that Adinbitor had typical functions of disintegrins.

The baker's yeast Saccharomyces(S.)cerevisiae is a single-celled eukaryotic model organism widely used in research on life *** a unicellular organism,*** has some evident limitations in application to ***,yeast prions are extensively studied and they are known to share some hallmarks with mammalian prion protein or other amyloidogenic proteins found in the pathogenesis of Alzheimer's,Parkinson's,or Huntington's ***,the yeast *** has been widely used for basic research on aggregation properties of proteins in cellulo and on their ***,a yeast-based study revealed that some regions of mammalian prion protein and amyloidβ1–42 are capable of induction and propagation of yeast *** is one of the examples showing that evolutionarily distant organisms share common mechanisms underlying the structural conversion of prion proteins making yeast cells a useful system for studying mammalian prion *** has also been used to design novel screening systems for anti-prion compounds from chemical *** assays are cheap in maintenance and safe for the researcher,making them a very good choice to perform preliminary screening before further characterization in systems engaging mammalian cells infected with *** this review,not only classical red/white colony assay but also yeast-based screening assays developed during last year are *** analysis and research carried out using yeast prions force us to expect that prions are widely present in ***,the last few years brought us several examples indicating that the mammalian prion protein is no more peculiar protein–it seems that a better understanding of prion proteins nature-wide may aid us with the treatment of prion diseases and other amyloid-related medical conditions.

The transfection of NIH 3T3 cells was performed with DNAs from 2 duck primary hepatic carcinomas (DHC 40K, 9K) and I tumor-adjacent liver tissue (TAL, 9N). Transfectants were found from 40K, 9K and 9N DNAs. The secondary transfectants were obtained after transfection of RAT-1 cells with DNAs from primary transfectants. After hybridization with Ha-ras, Ki-ras, N-ras and mht oncogenes, it was found that duck mht (5.2 and 3.2 kb EcoRI fragments) and duck Ha-ras (3.4 kb EcoRI fragments) were present in all these *** is the first report on transforming genes in duck primary hepatic cancer as well as tumoradjacent liver tissue

Huntington’s disease（HD）is a currently incurable,late onset,progressive,ultimately fatal neurological disorder（Bates et al.,2015）.We have recently published the results of comprehensive genetic interaction tests aimed at identification of histone methyltransferases and demethylases involved in HD pathogenesis in a Drosophila model of the disease（Song et al.,2018）.

Human acute premyeloid leukemia cell cDNA expression library was constructed to screen acute premyeloid leukemia tumor antigen. Total RNA and purified mRNA were extracted from human premyeloid cell line NB4. First and second strands of cDNA were synthesized by reverse transcription. After blunting, the cDNA fragments were ligated with EcoR Ⅰ adapters. Then the cDNAs were digested with Xho Ⅰ, and less than 400 bp cDNA fragment was removed by Sephacryl S400 spin column, the remaining were ligated with λZAP vector. The recombinants were packaged in vitro , and a small portion of packaged phage was used to infect E. coli XL1 Blue MRF' for titration. The recombinants were examined by color selection. In order to evaluate the size of cDNA inserts and the diversity of library, the pBK CMV phagemid was excised from the ZAP express vector by using ExAssist helper phage with XLOLR strain, and then the pBK CMV phagemid was digested by Xho Ⅰ and EcoR Ⅰ. The results showed that the NB4 cell line cDNA library consisting of 1.65×10 6 recombinant bacteriophages was constructed with the recombinant ratio of 99.6 %. The average length of the recombinant exogenous inserts was about 1.7 kb. It was concluded that the constructed cDNA library are deserved to screen target clones.

EXPRESSION OF N-ras GENE IN HUMAN PRIMARY HEPATOCARCINOMA

Poly(A)~+ RNA was isolated from 9 specimens of human primary hepatic carcinoma, 1 non-tumorous liver tissue adjacent to cancer and 1 normal liver tissue samples. The Oligo-dT cellulose-purified poly(A)~+ RNAs were subjected to formaldehyde agarose gel electrophoresis, Northern transfer and hybridization with various ancogene probes. Two RNA species, 5.6 kb and 2.2 kb wore identified by N-ras gone hybridization in 6 out of 9 mRNA samples from primary hepatic carcinoma specimen. N-ras specific mRNA was not detectable in mRNA samples from normal human liver and tumor surrounding cirrhotic tissue. No detectable hybridization of mRNA from hepatoma and normal liver with Ki-ras or Ha-ras was *** human N-ras gene has been identified in DNA of mouse transfeetants transformed with PHC DNA, it strongly suggests that N-ras gene might be responsible for the transforming activity of part of cases of human liver cancer.
