Objective: Radix Bupleuri and Radix Paeoniae Alba are referred to as Chai-Hu and Bai-shao, respectively, in Chinese. We used molecular docking and network pharmacological approaches to explore the active components of this Chinese herb combination which is commonly used for treating Alzheimer's disease(AD), the Chai-Hu and Bai-shao pair(CBP). Materials and Methods: The active compounds and action targets of Chai-Hu and Bai-shao were identified in the Traditional Chinese Medicine systems Pharmacology database, and the Therapeutic Target Database, Drug Bank, Gen Cards, Dis Ge NET, and Online Mendelian Inheritance in Man databases were used to identify therapeutic targets in patients with AD. To identify a common target of CBP in the treatment of AD, we used the Metascape tool to perform Gene Ontology function enrichment and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses. In addition, CBP-AD protein–protein interaction(PPI) and herb-component-target networks were created using Cytoscape 3.8.2. Molecular docking was verified using Auto Dock tools. Results: We identified 20 active compounds of CBP, 202 active targets, 1,323 disease targets, and 117 active compound disease intersection targets;the PPI network showed that the top eight key targets were AKT1, interleukin(IL) 6, JUN, MAPK1, tumor necrosis factor, TP53, vascular endothelial growth factor, and epidermal growth factor. These act mainly on the fluid shear stress, atherosclerosis, IL-17, hypoxia-inducible factor-1, and nonalcoholic fatty liver disease pathways, among others, which control immune inflammation, cell proliferation, and apoptosis. Based on molecular docking results, the active components of CBP bind well to its key targets. Conclusion: Our results indicated that CBP was effective for treating AD by reducing the expression of inflammatory factors, promoting cell proliferation and differentiation, and preventing nerve cell death and anti-neuronal cell apoptosis.

Monitoring water quality in an aquatic system is a crucial objective in pursuing sustainable development,despite the challenges posed by water scarcity and the impact of climate *** objective of the present study was to examine the water quality of the Tigris River along its course from the point of entry into Baghdad city to its exit,particularly in the presence of Phthalic Acid Esters(PAEs).In both dry and wet seasons and from five sites of the river,nine environmental factors(Turbidity,Total dissolved solids,pH,dissolved oxygen,Chloride,nitrate,phosphate,sulfate,and Biological oxygen demand)and three PAEs(di-n-butyl phthalate(DBP),di-isononyl phthalate(DiNP),and di-2-ethyl hexyl phthalate(DEHP))were *** revealed that the river has high levels of turbidity and Total dissolved solids,slightly alkaline,and well-aerated(dissolved oxygen>7 mg/L).Chloride,nitrate,phosphate,and sulfate values varied between seasons,as high values were recorded in the wet *** oxygen demand levels were higher during the dry *** highest value of total PAEs(2.20μg/L)was observed during the dry season,and the lowest value was in the wet season(1.16μg/L).The HQ results revealed that the river was ranked as a moderate risk *** the water quality index and environmental risk index classified the water quality as a poor category,with a high-risk score in the study sites within Baghdad ***,despite the acceptable limit of PAEs in the river's water(s presence is an early alarm to withdraw attention to its environmental *** findings highlight the need for appropriate measures to control and remove pollutants to ensure a safe and healthy water supply in the Tigris River.

Objective To explore the rapid antidepressant potential and the underlying mechanism of Chaihu shugan san(Css)in female *** Liquid chromatography mass spectrometry(LC-Ms)/Ms was used to determine the content of main components in Css to determine its *** C57BL/6J mice were randomly divided into 4 groups,including control(saline),vehicle(saline),Css(4 g/kg)and ketamine(30 mg/kg)*** were subjected to irregular stress stimulation for 4 weeks to establish the chronic mild stress(CMs)model,then received a single administration of *** hours later,the behavioral tests were performed,including open field test,tail suspension test(TsT),forced swimming test(FsT),novelty suppression feeding test(NsF),and sucrose preference test(sPT).Western blot analysis was used to detect the expression levels of N-methyl-D-aspartate receptor(NMDA)subtypes[N-methyl-D-aspartate receptor 1(NR1),NR2A,NR2B],synaptic proteins[synapsin1 and post synaptic density protein 95(PsD95)],and brain-derived neurotrophic factor(BDNF).Moreover,the rapid antidepressant effect of Css was tested by pharmacological technologies and optogenetic interventions that activated glutamate receptors,*** Compared with the vehicle group,a single administration of Css(4 g/kg)reversed all behavioral defects in TsT,FsT,sPT and NsF caused by CMs(Pss also significantly decreased the expressions of NMDA subtypes(NR1,NR2A,NR2B)at 2 h in hippocampus of mice(all Psimilar to ketamine,Css increased levels of synaptic proteins and BDNF(Pssant effects of Css were blocked by transient activation of NMDA receptors in the hippocampus(all Psion Rapid antidepressant effects of Css by improving behavioral deficits in female CMs mice depended on rapid suppression of NMDA receptors and activation of synaptic proteins.
