Overexpression of chaperonin containing TCP1, subunit 3 predicts poor prognosis in hepatocellular carcinoma

AIM:To investigate the value of chaperonin containing TCP1,subunit 3(CCT3) to predict the prognosis of patients with hepatocellular carcinoma(HCC) and determine its function in HCC ***:CCT3 expression levels were examined in human non-cancerous liver tissues and a variety of HCC cell lines by quantitative real-time PCR and ***3 expression was suppressed by small interfering *** effects of reducing CCT3 expression in HCC cells were ***3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyl tetrazolium bromide(MTT) assay,cell counting experiment,cell cycle assay,apoptosis assay and invasion assay were employed to evaluate cell functions in *** was performed on HCC *** addition,CCT3 expression in HCC specimens was also assessed at the protein and mRNA *** between clinicopathological characteristics and prognosis were analyzed,along with the possible mechanisms involved in CCT3's function in HCC ***:The expression levels of CCT3 mRNA and protein were upregulated in HCC cell lines in contrast to adjacent non-cancerous *** CCT3 expression not only suppressed cell proliferation in cell counts,MTT assay,cell cycle assay and induced cell apoptosis(P cancer cells in HCC specimens(58of 104 patients,55.8%) was associated with poor prognosis in HCC patients(3-year survival rate,55.5%vs 84.2%,P = 0.020) after *** analyses showed that signal transducer and activator of transcription 3(STAT3) activation was decreased even when stimulated by interleukin-6 after knocking down CCT3 in the HepG2 cell ***:Overexpression of CCT3 in the nuclei of cancerous cells is associated with HCC ***3 may be a target that affects the activation of STAT3 in HCC.

Expression of the MAGE-1 gene in human hepatocellular carcinomas

To further investigate the expression of MAGE 1 gene in hepatocellular carcinoma (HCC) Methods The tumors and adjacent liver tissue from 45 HCC patients and liver tissue from 28 non HCC patients (16 with liver cirrhosis and 12 with normal liver) were characterized by RT PCR A 421?bp PCR product from a cDNA fragment spanning exons 1, 2 and 3 was sequenced The HLA type was assayed by standard ELISA in 43 HCC patients Results Thirty two of 45 tumor tissues from HCC patients expressed MAGE 1 mRNA (71 1%) In contrast, MAGE 1 mRNA was not detected in adjacent tissues Three were found to have point mutations at 3 identical sites resulting in the substitution of two amino acid residues The most frequent HLA types in 43 HCC patients were: HLA A2, 53 5%; A11, 25 6%; A24, 20 9%; A33, 20 9%; HLA B13, 28 3% and B35, 23 2% Expression of HLA A33 (20 9%) was higher in HCC patients than that predicted in the normal Chinese population (8 8%) There was no discernable correlation between MAGE 1 expression and α FP level, tumor size and hepatitis B or C virus infection The identification of peptides which are restricted by haploptypes other than A1 should increase the opportunity for peptide based immunotherapy Conclusions This study shows that MAGE 1 mRNA is highly expressed in HCC tumor tissue in Chinese patients Previously unreported point mutations in the MAGE 1 gene are described and may also provide additional opportunities for immunotherapy

Early urinary candidate biomarker discovery in a rat thioacetamide-induced liver fibrosis model

Biomarker is the change associated with the disease. Blood is relatively stable because of the homeostatic mechanisms of the body. However, urine accumulates changes of the body, which makes it a better early biomarker source. liver fibrosis is a reversible pathological condition, whereas cirrhosis, the end-stage of liver fibrosis, is irreversible. Consequently, noninvasive early biomarkers for fibrosis are desperately needed. In this study, differential urinary proteins were identified in the thioacetamide liver fibrosis rat model using tandem mass tagging and two-dimensional liquid chromatography tandem mass spectrometry. A total of 766 urinary proteins were identified, 143 and 118 of which were significantly changed in the TAA 1-week and3-week groups, respectively. Multiple reaction monitoring(MRM)-targeted proteomics was used to further validate the abundant differentially expressed proteins. A total of 40 urinary proteins were statistically significant, 15 of which had been previously reported as biomarkers of liver fibrosis, cirrhosis or other related diseases and 10 of which had been reported to be associated with the pathology and mechanism of liver fibrosis. These differential proteins were detected in urine before the alanine aminotransferase and aspartate transaminase changes in the serum and before fibrosis was observed upon hematoxylin and eosin(HE) and Masson's staining.

Background:The use of laparoscopic(LLR)and robotic liver resections(RLR)has been safely performed in many institutions for liver tumours.A large scale international multicenter study would provide stronger evidence and insight into application of these techniques for huge liver tumours≥10 ***:This was a retrospective review of 971 patients who underwent LLR and RLR for huge(≥10 cm)tumors at 42 international centers between ***:One hundred RLR and 699 LLR which met study criteria were *** comparison between the 2 approaches for patients with huge tumors were performed using 1:3 propensity-score matching(PSM)(73 vs.219).Before PSM,LLR was associated with significantly increased frequency of previous abdominal surgery,malignant pathology,liver cirrhosis and increased median *** PSM,RLR and LLR was associated with no significant difference in key perioperative outcomes including media operation time(242 vs.290 min,P=0.286),transfusion rate rate(19.2%vs.16.9%,P=0.652),median blood loss(200 vs.300 mL,P=0.694),open conversion rate(8.2%vs.11.0%,P=0.519),morbidity(28.8%vs.21.9%,P=0.221),major morbidity(4.1%vs.9.6%,P=0.152),mortality and postoperative length of stay(6 vs.6 days,P=0.435).Conclusions:RLR and LLR can be performed safely for selected patients with huge liver tumours with excellent *** was no significant difference in perioperative outcomes after RLR or LLR.
