This study aimed to determine the regulatory mechanism of dietary zinc lactate(ZL)supplementation on intestinal oxidative stress damage in a paraquat(PQ)-induced piglet ***-eight piglets(mean body weight 9.51±0.23 kg)weaned at 28 d of age were randomly divided into control,ZL,PQ,and ZL+PQ groups(n=7 in each group).The ZL-supplemented diet had little effect on growth performance under normal physiological ***,under PQ challenge,ZL supplementation significantly improved average daily gain(P<0.05)and reduced the frequency of *** improved intestinal morphology and ultrastructure by significantly increasing the expression level of the jejunal tight junction protein,zonula occludens-1(ZO-1)(P<0.05),and intestinal zinc transport and absorption in PQ-induced piglets,which reduced intestinal *** supplementation also enhanced the expression of antioxidant and antiinflammatory factor-related genes and decreased inflammatory cytokine expression and secretion in PQinduced ***,ZL treatment significantly inhibited the activation of constitutive androstane receptor(CAR)signaling(P<0.01)in PQ-induced piglets and altered the structure of the gut microbiota,especially by significantly increasing the abundance of beneficial gut microbes,including UCG_002,Ruminococcus,Rikenellaceae_RC9_gut_group,Christensenellaceae_R_7_group,Treponema,unclassified_Christensenellaceae,and unclassified_Erysipelotrichaceae(P<0.05).These data reveal that pre-administration of ZL to piglets can suppress intestinal oxidative stress by improving antioxidant and anti-inflammatory capacity and regulating the crosstalk between CAR signaling and gut microbiota.

Integrins are a highly complex family of receptors that, when expressed on the surface of cells, can mediate reciprocal cell-to-cell and cell-to-extracellular matrix(ECM) interactions leading to assembly of integrin adhesion complexes(IACs) that initiate many signaling functions both at the membrane and deeper within the cytoplasm to coordinate processes including cell adhesion, migration, proliferation, survival, differentiation, and metabolism. All metazoan organisms possess integrins, and it is generally agreed that integrins were associated with the evolution of multicellularity, being essential for the association of cells with their neighbors and surroundings, during embryonic development and many aspects of cellular and molecular biology. Integrins have important roles in many aspects of embryonic development, normal physiology, and disease processes with a multitude of functions discovered and elucidated for integrins that directly influence many areas of biology and medicine, including mammalian pregnancy, in particular implantation of the blastocyst to the uterine wall, subsequent placentation and conceptus(embryo/fetus and associated placental membranes) development. This review provides a succinct overview of integrin structure, ligand binding, and signaling followed with a concise overview of embryonic development, implantation, and early placentation in pigs, sheep, humans, and mice as an example for rodents. A brief timeline of the initial localization of integrin subunits to the uterine luminal epithelium(LE) and conceptus trophoblast is then presented, followed by sequential summaries of integrin expression and function during gestation in pigs, sheep, humans, and rodents. As appropriate for this journal, summaries of integrin expression and function during gestation in pigs and sheep are in depth, whereas summaries for humans and rodents are brief. Low-fishmeal and protein-saving diets are two prominent nutritional strategies utilized to address challenges related to the scarcity and sustainability of protein sources in aquaculture. However, these diets have been associated with adverse effects on the growth performance, feed utilization, and disease resistance of aquatic animals. To mitigate these challenges, exogenous protease has been applied to enhance the quality of diets with lower protein contents or fishmeal alternatives, thereby improving the bioavailability of nutritional ingredients. Additionally, protease preparations were also used to enzymatically hydrolyze fishmeal alternatives, thus enhancing their nutritional utilization. The present review aims to consolidate recent research progress on the use of protease in aquaculture and conclude the benefits and limitations of its application, thereby providing a comprehensive understanding of the subject and identifying opportunities for future research.

Crossed beak is a complex mode of inheritance with prevalence ranging from 0.2 to 7.4% in at least 12 chicken strains *** reveal the intrinsic factors causing crossed beaks,genes expression patterns in bilateral mandibular condyle between affected and normal birds were characterized by RNA sequencing analysis in the present *** beak was induced by short length of unilateral mandibular ramus,and a total of 110differentially expressed genes were up-or down-regulated in the affected(short)mandibular condyle side as compared to the normal *** anhydrase 2(CA2)and Carbonic anhydrase 13(CA13)were enriched in the carbonate dehydratase activity,and high-expressed in mandibular condyle and osteoblasts(P<0.05).However,both were low-expressed in short mandibular condyle side of affected birds(P<0.05).The carbonate dehydratase inhibitor experiments confirmed that there is positive association between the calcification and carbonic anhydrase *** analysis with cetylpyridinium chloride showed a decrease in calcification when the cells were transfected with an anti-CA13 *** research suggested that CA2 and CA13 are down-calcified in shortside mandibular condyle,and caused mandibular ramus to grow ***2 and CA13 have the critical role in crossed beaks by regulating calcification of mandibular condyle.

Evaluating the effects of temperature variations on animals plays an important role in understanding the threat of climate *** effects of developmental temperature on offspring performance are critical in evaluating the effects of warming temperatures on the fitness of ovipa-rous species,but the physiological and biochemical basis of this developmental plasticity is largely *** this study,we incubated eggs of the turtle Pelodiscus sinensis at low(24℃),medium(28℃),and high(32℃)temperatures,and evaluated the effects of developmental temperature on offspring fitness,and metabolic enzymes in the neck and limb muscles of *** hatchlings from eggs incubated at the medium temperature showed better fitness-related performance(righting response and swimming capacity)and higher activities of metabolic enzymes(hexokinase,HK;lactate dehydrogenase,LDH)than hatchlings from the eggs incubated at high or low *** addition,the swimming speed and righting response were significantly correlated with the HK activities in limb(swimming speed)and neck(righting response)muscles,suggesting that the developmental plasticity of energy metabolic pathway might play a role in determining the way incubation temperature affects offspring *** the fitness-related performance and the activities of metabolic enzymes,we predict that the P sinensis from high latitude would not face the detrimental effects of climate warming until the average nest temperatures reach 32℃.

This study was conducted to explore the regulatory mechanism of leucine(Leu)on lipid metabolism of finishing ***-four Duroc×Landrace×Large cross pigs with an average body weight of68.33±0.97 kg were randomly allocated into 3 treatment groups with 8 replicates per group(1 pig per replicate).The dietary treatments were as follows:control group(CON),0.25%Leu group and 0.50%Leu *** experimental period was 42 *** results showed as follows.(1)Compared with the CON,0.25%and 0.50%Leu increased(P<0.01)the average daily gain(ADG),while the average backfat thickness(ABT)and the ratio of feed intake to body weight gain(F:G ratio)were decreased(P<0.05).(2)In the 0.25%Leu group,the relative mRNA expression levels of sterol regulatory element binding protein-1c(SREBP1c),recombinant fatty acid transport protein 1(FATP1),chemerin and peroxisome proliferatoractivated receptorγ(PPARγ)were decreased but the level of fatty acid binding protein 4(FABP4)and fatty acid translocase(FAT/CD36)were increased in backfat *** the 0.25%Leu group,the protein levels of p-Rictor,p-Raptor,p-eIF4E-binding protein 1(p-4EBP1),p-silent mating type information regulator 2homolog 1(p-SIRT1)and acetylation ribosome s6 protein kinase 1(Ac—S6K1)were increased(P<0.05).(3)Compared to the CON,the diversity of gut microbiota in the 0.25%Leu group was *** component analysis showed that the relative abundance of Bacteroidetes,Lactobacillus and Desulfovibrio was higher in the 0.25%Leu group than the CON,but the relative abundance of Firmicutes,Treponema and Shigella was lower than in the CON(P<0.05).(4)Four different metabolites were screened out from the serum of finishing pigs including allolithocholic acid(alloLCA),isolithocholic acid(isoLCA),ursodeoxycholic acid(UDCA)and hyodeoxycholic acid(HDCA),which correlate to various degrees with the above *** conclusion,Leu could promote adipose tissue lipolysis of finishing pigs through the mTOR-SIRT1 signaling pathway,a

Antimicrobial resistance in the laying hen production industry has become a serious public health *** antimicrobial resistance and phylogenetic relationships of the common conditional pathogen Enterococcus along the laying hen production chain have not been systematically clarified.105 Enterococcus isolates were obtained from 115 environmental samples (air,dust,feces,fies,sewage,and soil) collected along the laying hen production chain (breeding chicken,chick,young chicken,and commercial laying hen).These Enterococcus isolates exhibited resistance to some clinically relevant antibiotics,such as tetracycline (92.4%),streptomycin (92.4%),and erythromycin (91.4%),and all strains had multidrug resistance *** genome sequencing characterized 29 acquired antibiotic resistance genes (ARGs) that conferred resistance to 11 classes of antibiotics in 51pleuromutilin-resistant Enterococcus isolates,and lsa(E),which mediates resistance to pleuromutilins,always co-occurred with lnu(B).Alignments with the Mobile Genetic Elements database identified four transposons (Tn554,Tn558,Tn6261,and Tn6674) with several ARGs(erm(A),ant(9)-la,fex(A),and optrA) that mediated resistance to many clinically important ***,we identified two new transposons that carried ARGs in the Tn554 family designated as Tn7508 and Tn7492.A complementary approach based on conventional multi-locus sequence typing and whole genome single nucleotide polymorphism analysis showed that phylogenetically related pleuromutilin-resistant Enterococcus isolates were widely distributed in various environments on different production *** results indicate that environmental contamination by antimicrobial-resistant Enterococcus requires greater attention,and they highlight the risk of pleuromutilin-resistant Enterococcus and ARGs disseminating along the laying hen production chain,thereby warranting effective disinfection.

Avian pathogenic Escherichia coli(APEC)belonging to extraintestinal pathogenic ***(ExPEC)can cause severe infections in extraintestinal tissues in birds and humans,such as the lungs and ***(microcin production regulation,locus A,herein renamed AbsR,a blood survival regulator),a member of the MarR(multiple antibiotic resistance regulator)transcriptional regulator family,governs the expression of capsule biosynthetic genes in human ExPEC and represents a promising druggable target for ***,a deep understanding of the AbsR regulatory mechanism as well as its regulon is *** this study,we present a systems-level analysis of the APEC AbsR regulon using ChIP-Seq(chromatin immunoprecipitation sequencing)and RNA-Seq(RNA sequencing)*** found that AbsR directly regulates 99 genes and indirectly regulates 667 ***,we showed that:1)AbsR contributes to antiphagocytotic effects by macrophages and virulence in a mouse model for systemic infection by directly activating the capsular gene cluster;2)AbsR positively impacts biofilm formation via direct regulation of the T2SS(type II secretion system)but plays a marginal role in virulence;and 3)AbsR directly upregulates the acid tolerance signaling system EvgAS to withstand acid stress but is dispensable in ExPEC ***,our data indicate that the role of AbsR in virulence gene regulation is relatively conserved in ExPEC ***,this study provides a comprehensive analysis of the AbsR regulon and regulatory mechanism,and our data suggest that AbsR likely influences virulence primarily through the control of capsule ***,we found that AbsR severely represses the expression of the type I-F CRISPR(clustered regularly interspaced short palindromic repeats)-Cas(CRISPR associated)systems,which could have implications in CRISPR biology and application.
