Anti-cd TCR antibody-expanded cd T cells:a better choice for the adoptive immunotherapy of lymphoid malignancies

Cell-based immunotherapy for lymphoid malignancies has gained increasing attention as patients develop resistance to conventional *** T cells,which have major histocompatibility complex(MHC)-unrestricted lytic activity,have become a promising candidate population for adoptive cell transfer *** previously established a stable condition for expanding cd T cells by using anti-cd T-cell receptor(TCR)*** this study,we found that adoptive transfer of the expanded cd T cells to Daudi lymphoma-bearing nude mice significantly prolonged the survival time of the mice and improved their living *** further investigated the characteristics of these antibody-expanded cd T cells compared to the more commonly used phosphoantigen-expanded cd T cells and evaluated the feasibility of employing them in the treatment of lymphoid *** but sustained proliferation of human peripheral blood cd T cells was observed upon stimulation with anti-cd TCR *** to phosphoantigen-stimulated cd T cells,the antibody-expanded cells manifested similar functional phenotypes and cytotoxic activity towards lymphoma cell *** is noteworthy that the anti-cd TCR antibody could expand both the Vd1 and Vd2 subsets of cd T *** in vitro-expanded Vd1 T cells displayed comparable tumour cell-killing activity to Vd2 T ***,owing to higher C–C chemokine receptor 4(CCR4)and CCR8 expression,the Vd1 T cells were more prone to infiltrate CCL17-or CCL22-expressing lymphomas than the Vd2 T *** the peripheral blood cd T cells from lymphoma patients further confirmed that the anti-cd TCR antibody-expanded cd T cells could be a more efficacious choice for the treatment of lymphoid malignancies than phosphoantigen-expanded cd T cells.

IGF2BP3 Enhances the Growth of Hepatocellular Carcinoma Tumors by Regulating the Properties of Macrophages and CD8^(+)T Cells in the Tumor Microenvironment

Background and Aims:Overexpression of IGF2BP3 is associated with the prognosis of hepatocellular carcinoma(HCC).However,its role in regulating tumor immune microenvironment(TME)is not well ***,we investigated the effects of IGF2BP3 on macrophages and CD8^(+)T cells within the TME of ***:The relationship between IGF2BP3 and immune cell infiltration was analyzed using online bioinformatics *** of IGF2BP3 in mouse hepatoma cell line Hepa1-6 was established using CRISPR/Cas9 *** vitro cell coculture and subcutaneously implanted hepatoma mice model were used to explore the effects of IGF2BP3 on immune *** of CCL50l transforming growth factor beta 1(TGF-β1)was detected with quantitative real-time polymerase chain reaction,western blotting,and enzyme-linked immunosorbent *** binding of IGF2BP3 and its target RNA was verified by trimolecular fluorescence complementation system and RNA immunoprecipitation followed by quantitative or semiquantitative polymerase chain ***:IGF2BP3 expression was elevated in HCC and was positively correlated with macrophage *** with higher IGF2BP3 expression and lower macrophage infiltration had a better survival *** found that IGF2BP3 could bind to the mRNA of CCL5 or TGF-β1,increasing their expression,and inducing macrophage infiltration and M2 polarization while inhibiting the activation of CD8^(+)T ***,inhibition of IGF2BP3 combined with anti-CD47 antibody treatment significantly suppressed the growth of hepatoma in Hepa1-6 xenograft tu-mor ***:IGF2BP3 promoted the infiltration and M2-polarization of macrophages and suppressed CD8^(+)T activation by enhancing CCL5 and TGF-β1 expression,which facilitated the progression of Hepa1-6 xenograft tumor.

EXPRESSION OF P120ctn IN NON-SMALL-CELL LUNG CANCER: A CLINICOPATHOLOGICAL STUDY

Objective: To investigate the expression of p120ctn in non-small-cell lung cancer (NSCLC) and its relationship with clinicopathological factors and prognosis. Methods: p120ctn expression was tested by immunohistochemistry for 80 tumors from patients with non-small-cell lung cancer. Correlations were investigated between p120ctn immunostaining in primary tumors and clinicopathological characteristics and survival. Results: Abnormal expression of p120ctn was found in 68/80(85%) tumors in which 43 cases had cytoplasmic staining. Abnormal staining of p120ctn was related with high TNM stage (P=0.003) and nodal metastasis (P=0.024). However, there was no correlation between altered expression with poor differentiation and histological type. According to Kaplan-Meier survival estimate, the expression of p120ctn was related to the poor survival (P=0.015) of patients. A Cox regression analysis revealed that p120ctn expression was a significant independent factor in the prediction of survival for patients with non-small-cell lung cancer (P=0.008). Conclusion: altered expression of p120ctn was found in non-small-cell lung cancers and was correlated with lymph node metastasis and prognosis. From a practical point of view, the expression of p120ctn can be of prognostic value for patients with non-small-cell lung cancer.

In this study, the distribution of five Alzheimer's disease(AD)-related single nucleotide polymorphisms(SNPs) in the Han population was examined in combination with the evaluation of clinical cognition and brain pathological analysis. The associations among SNPs,clinical daily cognitive states, and postmortem neuropathological changes were analyzed in 110 human brains from the Chinese Academy of medical Sciences/Peking Union medical college(CAMS/PUMC) Human Brain *** ?4(OR = 4.482, P = 0.004), the RS2305421 GG genotype(adjusted OR = 4.397, P = 0.015), and the RS10498633 GT genotype(adjusted OR = 2.375,P = 0.028) were associated with a higher score on the ABC(Ab plaque score, Braak NFT stage, and CERAD neuritic plaque score) dementia scale. These results advance our understanding of the pathogenesis of AD,the relationship between pathological diagnosis and clinical diagnosis, and the SNPs in the Han population for future research.

Sialic acids as terminal entities of larger glycans linked to proteins and lipids are involved in multiple different pathological and physiological *** characterisation of sialoglycoconjugates is required to understand their biological ***,a comprehensive sialylation analysis of sialoglycoconjugates has remained *** this study,we employ a natural biomaterial,poplar catkin derived from white poplar tree（Populus tomentosa Carr.）,to develop a novel capturing microtip for selective and efficient enrichment of sialoglycopeptides,without losses of sialic acid residues and water molecules from *** electron microscopy and Fourier-transform infrared spectroscopy analysis,along with Maule and Wiesner staining assays,indicated that the main components on the outer layer of the poplar catkin are syringyl and guaiacyl lignins which play a key role in enriching sialoglycopeptides from complex peptide mixture.

Chaotic Dynamics of a Periodically Modulated Josephson Junction

We study the chaotic dynamics of a periodically modulated Josephson junction with damping. The general solution of the first-order perturbed equation is constructed by using the direct perturbation technique. It is theoretically found that the boundedness conditions of the general solution contain the Melnikov chaotic criterion. When the perturbation conditions cannot be satisfied, numerical simulations demonstrate that the system can step into chaos through a period doubling route with the increase of the amplitude of the modulating term. Regulating specific parameters can effectively suppress the chaos.

DNA-protein interaction at erythroid important regulatory elements of MEL cells bv in vivo footprinting

Using ligation-mediated PCR method to study the status of DNA-protein interaction at hypersensitive site 2 of locus control Region and βmaj promoter of MEL cell line before and after induction, MEL cell has been cultured and induced to differentiation by Hemin and DMSO, then the live cells have been treated with dimethyl sulfate. Ligation mediated PCR has been carried out following the chemical cleavage. The results demonstrate that before and after induction, the status of DNA-protein interaction at both hypersensitive site 2 and βmaj promoter change significantly, indicating that distal regulatory elements (locus control region, hypersensitive sites) as well as proximal regulatory elements (promoter, enhancer) of β-globin gene cluster participate in the regulation of developmental specificity.

Batroxobin inhibits astrocyte activation following nigrostriatal pathway injury

EXPRESSION OF MULTIDRUG RESISTANCE-ASSOCIATED PROTEIN (MRP) AND ITS RELATIONSHIP WITH CLINICOPATHOLOGICAL FACTORS IN NON-SMALL CELL LUNG CANCER

Objective: To investigate the relationship between the expression of multidrug resistance-associated protein (MRP) and clinicopathological factors and prognosis. Methods: The expression of MRP in 62 cases with non-small cell lung cancer (NSCLC) was detected using immunohistochemistry method. The expression of MRP in 30 cases of NSCLC and corresponding normal lung tissues were detected using immunohistochemistry and Western Blot. Results: this study of tumor tissues confirmed the plasma membrane and/or cytoplasm locations of MRP. There was apparent difference between normal lung tissues and NSCLC in MRP. The survival analysis of 62 NSCLC showed that the mean survival time of the patients with negative MRP expression was 69.8117.41 months and that of patients with positive MRP expression, 25.384.46 months. Log-rank test suggested that the difference between them was significant (P=0.0156). It was also found that in squamous cell lung cancer the statistically significant difference between the mean survival time of patients with positive MRP expression and those with negative MRP expression (P=0.0153). Multivariate Cox model analysis suggested that the survival time was significantly related to expression of MRP (P=0.035) and lymphatic metastasis (P=0.038). Conclusion: MRP expression in NSCLC is significantly higher compared with normal lung tissues. The mean survival time of patients with negative MRP was relative longer and expression of MRP was an independent factor for prognosis.

Effect of electroacupuncture in Weijing points on gastrointestinal interdigestive migrating motor complex and brain gut peptides release in dogs

Interdigestive gastrointestinal migrating motor complex (MMC) activities were recorded by strain gauge implanted on the serosa in 7 conscious dogs. We studied the effects of electroacupuncture (EAP) Weijing points Zusanli (S36), Tianshu (S25), Liangmen (S21) on MMC and release of motilin and gastrin, and compared them with that of EAP Pangguangjing points. The results indicated that EAP Weijing points could not only strengthen MMC contractions in antrum, duodenum and proximal jejunum, but also increase plasma concentration of motilin and gastrin. Anti-motilin serum, proglumide, atropine, or hexamethonium could markedly block the effect of EAP on reinforcing MMC contraction and release of motilin and gastrin. We could get the conclusions that such enhancing effect of EAP Weijing points on MMC and brain-gut peptides release is mediated by motilin and gastrin, on which both cholinergic nerve and sympathetic nerve play important roles.
