Objective:To determine the molecular characterization of Polymerase complex(PA,PB1 and PB2) genes of H9N2 avian influenza viruses and the genetic relalionsliip of Iranian H9N2 viruses and other Asian ***:The Polymerase complex(PA,PBl and PB2) genes from seven isolates of H9N2 viruses isolated from commercial chickens in Iran during 2008-2009 were amplified(by RT-PCR method) and *** sequences(Open Reading Frame: orf) of the PA,PBl and PB2 genes were used for phylogenetic tree ***:Most PB2 and PA genes of the H9N2 viruses isolated in 2008-2009 belonged to the unknown avian sublineage which grouped with the 2004 Pakistani H7N3 *** PBl genes of Iranian viruses indicated greater genetic diversity and shared a high level of similarity to PBl genes from either HS or H7 subtypes with compared to established H9N2 Eurasian ***:Our findings demonstrated that the H9N2 viruses in Iran exhibit striking reassortment which has led to the generation of new genotypes.

Dear Editor,Infectious bursal disease (IBD) is one of the most important diseases of the poultry. The IBD virus (IBDV), a nonenveloped virus belonging to the Birnaviridae family with a genome consisting of two segments of double-stranded RNA (segments A and B), targets B lymphocytes of bursa of Fabricious leading to immunosuppression. In Pakistan,poultry farming is the second biggest industry and IBD is the second biggest disease threating the poultry ***.

Leucocytozoonosis was found in three layer farms in chickens with suspected fatty liver or fatty liver hemorrhagic syndrome in Korea between 2009 and *** layer chicken flocks showed both mortality and decreased egg production for one or two weeks when they were between 59 and 82 weeks *** the necropsy,the most prominent gross lesions were found in the liver,which was enlarged,had a fragile texture,exhibited yellowish discolorations,and had various hemorrhagic *** reactions associated with megaloschizonts specific for Leucocytozoon caulleryi were prominent upon microscopic examination of the liver without significant *** addition,the ovaries and uterus were the most affected organs for Leucocytozoon caulleryi multiplication,which led to decreased egg *** studies with formalin-fixed,paraffin-embedded tissues were performed in search of a partial region of the cytochrome b gene for hemosporidian *** on these results,the causal agent was determined to be closely related to Leucocytozoon caulleryi reported in Japan and *** this study,we describe recently re-occurring leucocytozoonosis in layer chickens,which required histopathology for disease *** prevent outbreaks and maintain chicken health and egg production,layer chickens need to be monitored for symptoms of leucocytozoonosis.

Mycoplasma synoviae （MS） is an economically important pathogenic agent of chickens, causing air sacculitis and synovitis. The diagnosis and monitoring of M. synoviae infection is usually made using serological assays, while confirmative diagnosis is made by isolation and identifcation of the organism, because of the cross reaction between M. gallisepticum and M. synoviae. This study was conducted from 2011 to 2013 during which l l broiler breeder flocks were sampled. These farms were located in all regions of morocco, The sampling was conducted as follows： Farms were visited on day one old chicks ＂day of importation from Europe＂. 20 to 60 ＂chicks＂ were randomly sampled. At the age of 8, 16, 32 and 56 weeks, 60 blood samples and 60 tracheal swabs were collected at each sampling. The serological screening was performed using Rapid Slide Agglutination （RSA） according the OIE protocol and Indirect EL1SA （IDEXX） according the manufacturer＇s instructions. The molecular diagnosis was performed using a commercial kit of a duplex real time PCR （Life Biotechnology）. The results revealed that one day old chicks were negative to MS by RSA and PCR, however they have a variable stock of maternal antibodies （Ig-Y） detected by iELISA. The seroprevalence found by RSA is variable and increase with the age （Sth week： 55%, 16th week： 91%, 32tb and 56th week： 100%）, the same profile was traced by PCR （Sth week： 36%, 16th week： 64%, 32th week： 82%, 56th week： 100%）, however, all farms were positive by iELISA, from the first day to 56th weeks. These results show that MS infections are very frequent and very widespread among poultry breeder flocks, and showed a perfect agreement between serological tests and Real time PCR starting from 32th week of age.

The Shangcheng stout salamander(Pachyhynobius shangchengensis) is a poorly known stream dwelling hynobiid salamander from China. We studied the reproduction, development and growth response of the salamander to different diets in captivity. Two females each produced two unique, striated egg sacs, with each containing 18 and 20, and 22 and 32 white eggs with an average diameter of 3.3 mm. Hatchlings(n = 27) had front- and hind-limb buds with interdigit membranes and a large yolk sac, and developed cornified finger and toe tips during further development. Metamorphosis occurred between d 441 and d 454 after hatching at an average total length of 94.8 mm(n = 21). These reproductive and developmental traits fit in well with the ancestral state reconstruction in hynobiids. If corrected for dry mass, feed conversion ratios obtained by feeding bloodworms(Chironomidae) to P. shangchengensis larvae and mealworms(Tenebrio molitor) to postmetamorphs, were very low(0.33 and 0.34 respectively), equaling a very high mass conversion efficiency.

The highly pathogenic avian influenza H5N1 pandemic motivated countries around the World to be prepared for outbreaks within their borders. Well ahead of the outbreak, in Jordan contingency plans were prepared involving all relevant bodies in both the private and public sectors and a practice was simulated to test the effectiveness of the task force and the applicability of the plan. The highly pathogenic H5N1 influenza virus was detected and confirmed in one turkey in a backyard flock comprising 12 turkeys and 25 chickens in Kufranjah, 6 km from Ajloun city, on 23 March 2006. The samples were confirmed by the following day. Control measures were implemented immediately according to the national contingency plan and complete clean up achieved by 27 March 2006. A 3 km area around the index case was designated as a control zone, with a 10 km area around the holding further designated as a surveillance zone. The objective was to stamp out the infection and approximately 20,000 birds were destroyed including all commercial and backyard flocks in the control zone. All the human contacts were examined, treated and discharged from the hospital. The immediate response from all Jordanian sectors involved in the control strategy was positive as a result of increasing awareness and training programs that were implemented six months before the occurrence of the outbreak. A total estimated economic loss posed by this outbreak including compensation was almost $US169 million. Following the outbreak, passive surveil-lance for the virus was undertaken and training and capacity building continued. Efforts made in the preparation for an outbreak paid dividends and the need for a department responsible for monitoring migratory and wild birds in Jordan was recognized. Legislation regarding biosecurity of poultry farms and poultry transport vehicles should be revised.

Infectious bursal disease(IBD),caused by IBD virus(IBDV),is one of the most devastating and immunosuppressive diseases of the poultry and has been a constraint on the sustainable poultry production around the globe including *** the disease is threatening the poulty industry,the nature of predominant strains of IBDV in Pakistan remained *** this study,an epidemiology survey was conducted in the main chicken-farming regions of *** batch of Pakistan IBDVs genes simultaneously covering both VP1 and VP2 were amplified,sequenced,and *** unique segment-reassortant IBDVs(vv-A/Uniq-B),carrying segmentA from vvIBDV and segment B from one unique ancestor,were identifed as one important type of circulating strains in *** data also discovered the characteristic molecular features of Pakistan IBDVs,which will contribute to scientific vaccine selection and effective prevention of the disease.

The production of cells capable of expressing gene(s) of interest is important for a variety of applications in biomedicine and biotechnology, including gene therapy and a novel method of stem cell therapy in the various diseases. Achieving high levels of transgene expression for the longer period of time, without adversely affecting cell viability and differentiation capacity of the cells, is crucial. In the present study, we investigated the efficiency of plasmid vector for the production of transgenic cMSCs and examined any functional change of cells after transfection. To do so first we have collected bone marrows from the adult goats and cultured them for isolation of mesenchymal stem cells (cBM-MSCs). These cells were characterized using MSC specific markers including differentiation into osteocytes and adipocytes. Transfection with plasmid vector did not adversely affect cBM-MSCs morphology, viability or differentiation potential, and transgene expression levels were unaffected beyond passage 12th. The results indicated that we have been able to generate transgenic caprine MSC (tcBM-MSC) and transfection of cBM-MSCs using plasmid vector resulted in very high and stable transfection efficiency. This finding may have considerable significance in improving the efficacy of MSC-based therapies and their tracking in animal model.

Objective:To determine the macroscopic and microscopic lesions of various dosages of gentamicin in the partridge ***:Fertile chukar partridge eggs were allocated into four *** 1:saline-injected group whose individuals were administered by sterile physiological saline solution of 0.2 mL/egg inserted into yolk *** 2,3 and 4 whose individuals were similarly administered by gentamicin sulfate at a dosage of 80 mg/kg egg-weight once,twice and three times,***:Results showed that the embryos were congested and stunted in the gentamicininjected *** in feet,wings and feather development were accompanied by microscopic lesions in brain,meninges,heart,lungs,liver and *** lesions were noticed as edema,undeveloped tissues,necrosis and degeneration in the affected ***:Based on acquired results,it is concluded that gentamicin at above-described dosages causes toxicopathological effects to the partridge embryo in a dose dependent manner.

Frequent Mutation of rs13281615 and Its Association with PVT1 Expression and Cell Proliferation in Breast Cancer

The q24 band of chromosome 8 （8q24） is frequently amplified in human cancers including breast cancer, and several SNPs （single nucleotide polymorphisms） at 8q24, including rs13281615, have been identified for their association with cancer risks. These SNPs are in a ＂gene desert＂ region, and their functions in cancer development remain to be illustrated, although several of the SNPs appear to influence the genes in the ＂desert＂ in a long-range manner, including the v-myc avian myelocytomatosis viral oncogene homolog （MYC） and the non- protein coding plasmacytoma variant translocation 1 （PVT1）, both of which have been implicated in human cancers. In the current study, we examined rs13281615 for its potential role in breast cancer using normal and cancer tissues from 121 Chinese women with breast cancer. In addition to confirming the association of the GG genotype of rs 13281615 with breast cancer risk, we found that germline GG genotype was significantly associated with estrogen receptor （ER） positivity, higher tumor grade and higher proliferation index. We also found frequent somatic mutations （22/121 or 18.2%） of this SNP in breast cancer. Interestingly, the majority of the mutations （17/22 or 77%） involved a G→ A change, resulting in a decrease in the number of cancers with the GG risk genotype and subsequent loss of GG association with higher tumor grade and proliferation index in cancers. Furthermore, PVT1 expression was increased in cancers, and the increase was associated with the GG genotype of rs13281615. These results suggest that the GG genotype of SNP rs13281615 plays a role in breast cancer likely by influencing PVT1 expression, and that during oncogenesis, ＂protective＂ mutations could occur.
