A reliable,efficient anther culture system,the dominant technique for generating haploid plants in breeding programs,that can be used for generating transgenic poplar plants has been *** the present study,therefore,an anther culture system was developed using isolated mid-and late-uninucleate anthers of poplar(Populus simonii x ***).From a co.bination of SSR and ploidy analyses,six double haploid and two haploid lines were characterized from 86 plants grown from 16 regenerated anther cultured *** 48 months of development,two plant lines from the regenerated plants maintained their haploid level in vitro for over 2 years.A number of haploid plants from the different lines weretransferred to *** leaves of these transplants were then used as explants for transformation with the APETALA1(AP1) gene using Agrobacterium *** of AP1 in haploid poplar induced early flowering with obvious petals when ectopically *** our knowledge,this is the first report on changes in flowering time in AP1-trangenic poplar,which is important for elucidating the regulatory mechanism of tree flower development.

Objective: To in vestigate the relati on ship betwee n gene mutations and resp onse to co.po und Qinghuang Powder (复方青黄散,CQHP) in patients with myelodysplastic syndrome (MDS). Methods: Forty-three MDS patients after treatment with CQHP for 6 months were genotyped by ultra-deep targeted sequencing and the clinical data of patients were co.lected and the relationship between them was analyzed. Results: Up to 41.86% of patie nts harbored genet mutations, in most cases with more than one mutation. The most co.m on mutations were in SF3B1, U2AF1, ASXL1, and DNMT3A. After treatment with CQHP, about 88.00% of patients no longer required blood transfusion, or needed half of prior transfusions. co.lusion: CQHP is an effective treatment for patients with MDS, especially those with gene mutations in SF3B1, DNMT3A, U2AF1, and/or ASXL1.

As a traditional herbal medicine,the major alkaloids in Uncaria rhynchophylla have been proven to have blood pressure-lowering and sedative *** is essential to develop an effective method for the determination of the major alkaloids in *** this research,a rapid quantitative analysis involving multi-co.ponents analysis by a single marker strategy co.pled with co.e-shell co.umn HPLC was adopted to analyse four alkaloids(co.ynoxeine,isoco.ynoxeine,isorhynchophylline,rhynchophylline)in *** was selected as the internal reference substance,the co.tent of which was determined by the traditional external standard *** co.rection factors(RCF)between isorhynchophylline and the other three alkaloids were calculated *** results showed that the QAMS method had good robustness under different HPLC *** batches of *** were *** significant difference was observed between the results by QAMS and EMS(co.relation co.fficient>0.99,p>0.05).The QAMS method co.ld be employed as a rapid,effective technique for the quality co.trol of ***.

Background:A traditional Chinese medicine formula,Youdujing(YDJ)ointment,is widely used for treatment of human papilloma virus-related diseases,such as cervical ***,the underlying mechanisms by which active co.pounds of YDJ alleviates cervical cancer are still ***:We applied a co.prehensive network pharmaco.ogy approach to explore the key mechanisms of YDJ by integrating potential target identification,network analysis,and enrichment analysis into classical molecular docking ***,we used network and enrichment analyses to identify potential therapeutic ***,we performed molecular docking to investigate the potential active co.pounds of ***,we carried out a network-based analysis to unravel potentially effective drug ***:Network analysis yielded four potential therapeutic targets:ESRI,NFKB1,TNF,and *** docking highlighted that these proteins may interact with four potential active co.pounds of YDJ:E4,Y2,Y20,and ***,we found that Y2 or Y21 can act alone or together with E4 to trigger apoptotic cascades via the mitochondrial apoptotic pathway and estrogen ***:Our study not only explained why YDJ is effective for cervical cancer treatment,but also lays a strong foundation for future clinical studies based on this traditional medicine.

QTL Scanning for Rice Yield Using a Whole Genome SNP Array

High-throughput SNP genotyping is widely used for plant genetic studies. Recently, a RICE6K SNP array has been developed based on the Illumina Bead Array platform and Infinium SNP assay technology for genome-wide evaluation of allelic variations and breeding applications. In this study, the RICE6K SNP array was used to genotype a reco.binant inbred line （RIL） population derived from the cross between the indica variety, Zhenshan 97, and the japonica variety, Xizang 2. A total of 3324 SNP markers of high quality were identified and were grouped into 1495 reco.bination bins in the RIL population. A high-density linkage map, co.sisting of the 1495 bins, was developed, co.ering 1591.2 cM and with average length ofl.1 cM per bin. Segregation distortions were observed in 24 regions of the 11 chromosomes in the RILs. One half of the distorted regions co.tained fertility genes that had been previously reported. A total of 23 QTLs were identified for yield. Seven QTLs were firstly detected in this study. The positive alleles from about half of the identified QTLs came from Zhenshan 97 and they had lower phenotypic values than Xizang 2. This indicated that favorable alleles for breeding were dispersed in both parents and pyramiding favorable alleles co.ld develop elite lines. The size of the mapping population for QTL analysis using high throughput SNP genotyping platform is also discussed.

Dear Editor,Male infertility is a co.plex and multifactorial health problem with highly heterogeneous phenotypes,ranging from reduced sperm production to the co.plete absence of sperm in testis,and genetic factors play an important role in this problem.1 Patients with azoospermia have the highest risk of carrying genetic abnormalities,and this risk gradually decreases as sperm co.nt increases.1 Due to the high throughput and low co.t of next-generation sequencing(NGS)technology,many centers have used NGS to identify genetic defects associated with abnormal spermatogenesis.

The advent of CRISPR-Cas has revolutionized precise gene editing. While pioneering CRISPR nucleases like Cas9 and Cas12 generate targeted DNA double-strand breaks(DSB) for knockout or homology-directed repair, next generation CRISPR technologies enable gene editing without DNA DSB. Base editors directly co.vert bases, prime editors make diverse alterations, and dead Cas-regulator fusions allow nuanced co.trol of gene expression, avoiding potentially risks like translocations. Meanwhile, the disco.ery of diminutive Cas12 orthologs and Obligate Mobile Element-Guided Activity(OMEGA) nucleases has overco.e cargo limitations of adeno-associated viral vectors, expanding prospects for in vivo therapeutic delivery. Here, we review the ever-evolving landscape of cutting-edge gene editing tools, focusing on miniature Cas12 orthologs and OMEGA effectors amenable to single AAV packaging. We also summarize CRISPR therapies delivered using AAV vectors, discuss challenges such as efficiency and specificity, and look to the future of this transformative field of in vivo gene editing enabled by AAV vectors delivery.

AIM To determine whether oral administration of Bifidobacterium infantis CGMCC313-2(B. infantis CGMCC313-2) inhibits allergen-induced airway inflammation and food allergies in a mouse *** Ovalbumin(OVA)-induced allergic asthma and b-lactoglobulin-induced food allergy mouse models were used in this study. Following oral administration of B. infantis CGMCC313-2 during or after allergen sensitization, histopathologic changes in the lung and intestine were evaluated by hematoxylin and eosin(HE) staining. In the allergic asthma mouse model, we evaluated the proportion of lung-infiltrating inflammatory cells. OVAspecific IgE and IgG1 levels in serum and cytokine levels in bronchoalveolar lavage fluid(BALF) were also assessed. In the food allergy mouse model, the levels of total Ig E and cytokines in serum were *** Oral administration of B. infantis CGMCC313-2 during or after allergen sensitization suppressed allergic inflammation in lung and intestinal tissues, while the proportion of infiltrating inflammatory cells was significantly decreased in the BALF of allergic asthma mice. Moreover, B. infantis CGMCC313-2 decreased the serum levels of total Ig E in food allergy mice, and reductions in IgE and IgG1 were also observed in OVA-induced allergic asthma mice. The expression of interleukin-4(IL-4) and IL-13 in both serum and BALF was suppressed following the administration of B. infantis CGMCC313-2, while an effect on serum IL-10 levels was not *** B. infantis CGMCC313-2 inhibits the secretion of allergen-induced IgE, IL-4 and IL-13, and attenuates allergic inflammation.

The importance of structural variants(SVs)for human phenotypes and diseases is now *** a variety of SV detection platforms and strategies that vary in sensitivity and specificity have been developed,few benchmarking procedures are available to co.fidently assess their performances in biological and clinical *** facilitate the validation and application of these SV detection approaches,we established an Asian reference material by characterizing the genome of an Epstein-Barr virus(EBV)-immortalized B lymphocyte line along with identified benchmark regions and high-co.fidence SV *** established a high-co.fidence SV callset with 8938 SVs by integrating four alignment-based SV callers,including 109×Pacific Bio sciences(PacBio)co.tinuous long reads(CLRs),22×PacBio circular co.sensus sequencing(CCS)reads,104×Oxford Nanopore Technologies(ONT)long reads,and 114×Bionano optical mapping platform,and one de novo assembly-based SV caller using CCS reads.A total of 544 randomly selected SVs were validated by PCR amplification and Sanger sequencing,demonstrating the robustness of our SV *** trio-binning-based haplotype assemblies,we established an SV benchmark for identifying false negatives and false positives by co.structing the co.tinuous high-co.fidence regions(CHCRs),which co.ered 1.46 gigabase pairs(Gb)and 6882 SVs supported by at least one diploid haplotype *** high-co.fidence SV calls for a benchmark sample that has been characterized by multiple technologies provides a valuable resource for investigating SVs in human biology,disease,and clinical research.
