Fermented calcium butyrate supplementation in post-peak laying hens improved ovarian function and tibia quality through the“gut-bone” axis

The compromised egg quality and leg abnormality during the end of the laying cycle(after 40 weeks)have been leading to poor animal welfare and substantial economic losses. Therefore, the effects of fermented calcium(Ca) butyrate, produced by fermentation by Clostridium butyricum, on production,eggshell quality, and tibial property of hens were explored. A total of 192 Hy-line brown laying hens at50-week-old were assigned to a basal diet or the basal diet with 300 mg/kg of the fermented Ca butyrate from 50 to 58 weeks of age. Each treatment had 6 replicates with 16 hens each. The diet supplemented with 300 mg/kg fermented Ca butyrate notably increased egg weight, ovarian follicle number, and eggshell strength(P = 0.072) as compared to the basal diet, which were associated with cytokine secretion, toll-like receptor signaling pathways, and intestinal immunity based on the RNA-seq data from the granulosa. Dietary Ca butyrate inclusion decreased the expression of ileal tumor necrosis factor-alpha and serum pro-inflammatory cytokine concentration, as well as increased the content of serum immunoglobulin A when compared to the basal diet(both P 0.05). In addition, the diet with fermented Ca butyrate reduced the number of osteoclasts in the proximal tibia and the level of C-terminal cross-linked telopeptide of type I collagen, a bone resorption marker(P < 0.05), whereas it tended to increase the concentration of the procollagen type I N-terminal propeptide that reflects bone formation marker in serum. Moreover, the layers fed fermented Ca butyrate diets possessed higher(P < 0.05) bone area and trabecular number of the proximal tibia, yield load, and ultimate load than those that consumed basal diets. Background Commercial diets are frequently formulated to meet or exceed nutrient levels including those of lim-iting essential amino acids(AA)covering potential individual variations within the herd.However,the provision of dietary excess of AA,such as Lys,may lead to reduced appetite and growth in pigs.The mechanisms modulat-ing these responses have not been extensively investigated.This study evaluated the effect of Lys dietary excesses on performance and satiety biomarkers in post weaning pigs.Methods Twenty-four pigs aged 21 d and weighing 6.81±0.12kg(mean±SEM)were individually housed and offered 1 of 4 dietary treatments for 3weeks:a diet containing a standardized ileal digestible Lys reaching 100%(T0),120%(T1),150%(T2)or 200%(T3)of the NRC(2012)requirements.At the end of the experiment,blood samples from the cephalic vein of the T0 and T3 groups were obtained for AA analysis.In addition,primary intestinal cultures from T0 pigs were used,following their humane killing,to evaluate the effect of Lys on gut hormone secretion and AA sensors gene expression under ex vivo conditions.Results Feed intake was linearly reduced(P<0.001)and the weight gain to feed ratio reduced(P<0.10)with increased dietary levels of Lys during the third-and first-week post weaning,respectively.Cholecystokinin con-centration(P<0.05)and the metabotropic glutamate receptor 1 and the solute carrier family 7 member 2(P<0.10)gene expression was enhanced in proximal jejunum tissues incubated with Lys at 20mmol/L when compared to the control(Lys 0mmol/L).Plasma Lys and Glu(P<0.05)concentration increased in the T3 compared to T0 pigs.In contrast,plasma levels of His,Val,Thr,Leu(P<0.05)and Gln(P<0.10)were lower in T3 than T0 pigs.Conclusion The present results confirm that excess dietary Lys inhibits hunger in pigs.Moreover,the results provide evidence of pre-and post-absorptive mechanisms modulating these responses.Lys dietary excesses should be nar-rowed,when possible,to avoid negative effects of the AA on appetite in pigs.

Physiological strategies in wild rodents:immune defenses of commensal rats

The importance of issues associated with urban/commensal rats and mice(property damage,management costs,and health risks)press upon research on these animals.While the demography of commensal rodents is mostly studied,the need for understanding factors influencing their natural morbidity/mortality is also stressed.In this respect,more attention is expected to be paid to immunity,the physiological mechanism of defense against host survival threats(pathogens,parasites,diseases).Commensal rats and mice carry numerous pathogens that evoke diverse immune responses.The state of immunity in commensal house mice is studied in great detail,owing to the use of laboratory strains in biomedical research.Because commensal rats are,compared to mice,carriers of more zoonotic agents,rats’immunity is studied mainly in that context.Some of these zoonotic agents cause chronic,asymptomatic infections,which justified studies of immunological mechanisms of pathogen tolerance versus clearance regulation in rats.Occurrence of some infections in specific tissues/organs pressed upon analysis of local/regional immune responses and/or immunopathology.A survey of immunological activity/responses in commensal rats is given in this review,with mention of existing data in commensal mice.It should throw some light on the factors relevant to their morbidity and lifespan,supplementing the knowledge of commensal rodent ecology.

Seasonal Variation of Hearing Sensitivity in the Red-eared Slider(Trachemys scripta elegans)

Seasonal variation of hearing sensitivity has been observed in many vertebrate groups with obvious vocal behaviors.Circulating hormones,conspecific calling signals,and temperature are potential factors that drive these plasticity patterns.Turtles have a hearing range that appears to be limited to under 1.5 kHz and are often thought to be non-vocal;thus,they are commonly neglected in vocal communication research.In this study,we aimed to determine whether the auditory phenotype exhibits seasonal variation in sensitivity and to analyze the potential factors driving such variation patterns in turtles.We measured hearing sensitivity and sex hormone levels in female(estradiol)and male(testosterone and dihydrotestosterone)Red-eared sliders(Trachemys scripta elegans)during spring and winter.The results showed that auditory brainstem response(ABR)thresholds were significantly lower in spring than in winter at a frequency range of 0.5-0.9 kHz.The hearing-sensitivity bandwidth was wider,and the ABR latency was significantly shorter in spring than in winter.No significant differences were found in estradiol,testosterone,and dihydrotestosterone levels in T.scripta elegans between spring and winter.This study is the first to reveal the seasonal variation of peripheral hearing sensitivity in turtles,a special animal group with limited hearing range and less vocalization.Temperature variations may be used to explain these seasonal effects,but further research is required to confirm our findings.

animals must strike a balance between anti-predation behavior and other essential behaviors,such as foraging.Within the same species,strategies may vary on individuals’risk-taking preferences,and in this process the environment is a determinant,in addition to predator regime.The Crested Ibis(Nipponia nippon)exhibits such tendency.This is an endangered species,once inhabiting exclusively in China’s Qinling Mountain.This used to be the sole remaining wild population.However,over recent decades,this population has expanded.A portion has relocated to breed in the lower plain area,which is characterized by elevated level of human activities and landscape complexity.We used flight initiation distance(FID)as an indicator of the ibises’risk-taking preference,particularly their response to human proximity.Additionally,we examined the environmental factors influencing their foraging site selection,including altitude,terrain openness,human activity intensity and human construction.Our findings revealed a significantly shorter FID among individuals relocating to plain habitats,indicating a higher tolerance of human proximity.The results showed that FID decreased with distance to the nearest human settlement.Another finding is that FID was independent of instant human activity intensity and environmental factors(altitude and terrain openness).These different may arise from various combinations of human activity,predation risk,and food abundance within the two habitats.These results provide insights into the in situ conservation of the threatened species within the context of global urbanization.

Antifertility effects of EP-1 (quinestrol and levonorgestrel) on Pacific rats (Rattus exulans)

Pest rodents pose a serious threat to island biodiversity.Fertility control could be an alternative approach to control the impact of rodents on these islands.In this study,we examined the antifertility effects of EP-1 baits containing quinestrol(E)and levonorgestrel(P)using a dose of 50 ppm E and P at three different ratios(E:P ratio=1:2,1:1,and 2:1)on Pacific rats(Rattus exulans)in the Xisha Islands,Hainan,China.Compared to the control group,all animals in EP-1 treatment groups showed significantly decreased food intake and body weight.In treated males,there were obvious abnormalities in testis structure and a significant decrease of relative seminal vesicle weight,but no significant effect on relative uterine and ovarian weights(g kg-1 body weight),or ovarian structure in females.Adding 8%sucrose to the original 50-ppm baits(E:P ratio=1:1)significantly increased bait palatability for males and females.This dose induced uterine edema and abnormalities of ovarian structure in females but had no significant negative effect on the relative testis,epididymis,and seminal vesicle weights(g kg-1 body weight)or sperm density in males.In summary,50-ppm EP-1(1:1)baits have the potential to disrupt the fertility of females,and 8%sucrose addition to the EP-1 baits(E:P ratio=1:1)could improve bait palatability.This dose disrupted the testis structure in males.Future studies are needed to improve bait acceptance and assess the antifertility effects of EP-1(1:1)on Pacific rats in captive breeding trials and underfield conditions.

animal body size variation is of particular interest in evolutionary biology,but the genetic basis remains largely unknown.Previous studies have shown the presence of two parallel evolutionary genetic clusters within the fish genus Epinephelus with evident divergence in body size,providing an excellent opportunity to investigate the genetic basis of body size variation in vertebrates.Herein,we performed phylotranscriptomic analysis and reconstructed the phylogeny of 13 epinephelids originating from the South China Sea.Two genetic clades with an estimated divergence time of approximately 15.4 million years ago were correlated with large and small body size,respectively.A total of 180 rapidly evolving genes and two positively selected genes were identified between the two groups.Functional enrichment analyses of these candidate genes revealed distinct enrichment categories between the two groups.These pathways and genes may play important roles in body size variation in groupers through complex regulatory networks.Based on our results,we speculate that the ancestors of the two divergent groups of groupers may have adapted to different environments through habitat selection,leading to genetic variations in metabolic patterns,organ development,and lifespan,resulting in body size divergence between the two locally adapted populations.These findings provide important insights into the genetic mechanisms underlying body size variation in groupers and species differentiation.

Warming intensified the effects of nitrogen addition on N_(2)O emissions from alpine meadow in the northern Qinghai-Tibet Plateau

Warming and nitrogen(N)addition may impact soil nitrous oxide(N_(2)O)emissions,but the relationship between plant community composition and soil microbial activities remains unclear.For a two-year field study in the Qinghai-Tibet Plateau,open-top chambers were used to quantify the effects of warming,N-addition,and their interactions on N_(2)O emissions.We found that the N-addition greatly increased N_(2)O emissions by 77.4%in 2018 when compared to the control group.In contrast,warming showed little effect on N_(2)0 emissions but did increase the activity of enzymes associated with soil nitrification and denitrification.A combined effect of warming and N-addition of resulted in 208.6%(2018)and 90.8%(2019)increase in N_(2)0 emissions,respectively,compared to the individual treatments of warming or N-addition.Global warming in alpine meadows is causally linked to increased legume biomass which is further intensified with the N-addition.Intensified legume biomass(p<0.05),soil moisture(p<0.001)and enzyme activity(p<0.001)had a positive effect on N_(2)0 emissions,while diminished microbial carbon/nitrogen(MBC/MBN)(p<0.05)correlated with reduced N_(2)O emissions.Final results indicated that N-addition has a positive effect on N_(2)O emissions,and the addition of warming further intensifies this effect.The increased dominance of legumes and microbial N content contributes to this effect.These outcomes suggest that warming and atmospheric N deposition can stimulate N_(2)O emissions of alpine meadows inthefuture.

Bats are the second-most diverse group of mammals in the world,and bat flies are their main parasites.However,significant knowledge gaps remain regarding these antagonistic interactions,especially since diverse factors such as seasonality and host sex can affect their network structures.Here,we explore the influence of such factors by comparing species richness and composition of bat flies on host bats,as well as specialization and modularity of bat–bat fly interaction networks between seasons and adult host sexes.We captured bats and collected their ectoparasitic flies at 10 sampling sites in the savannahs of AmapáState,northeastern region of the Brazilian Amazon.Despite female bats being more parasitized and recording greater bat fly species richness in the wet season,neither relationship was statistically significant.The pooled network could be divided into 15 compartments with 54 links,and all subnetworks comprised>12 compartments.The total number of links ranged from 27 to 48(for the dry and wet seasons,respectively),and female and male subnetworks had 44 and 41 links,respectively.Connectance values were very low for the pooled network and for all subnetworks.Our results revealed higher bat fly species richness and abundance in the wet season,whereas specialization and modularity were higher in the dry season.Moreover,the subnetwork for female bats displayed higher specialization and modularity than the male subnetwork.Therefore,both seasonality and host sex contribute in different ways to bat–bat fly network structure.Future studies should consider these factors when evaluating bat–bat fly interaction networks.
