Background： In order to improve the efficiency of bovine sperm cryopreservation process, it is important to understand how spermatozoa respond to differences in temperature as well as the ability to recover its own metabolism. The combination between flow cytometry approach and antioxidant enzymes activity allows a more sensible evaluation of sperm cell during cryopreservation. The aim of this study was to evaluate sperm attributes and antioxidant enzymes activity during different stages of cryopreservation process. Semen samples from Holstein bulls （n = 4） were separated in 3 treatments： fresh （37 ℃）; cooled （5 ℃）; and thawed. Evaluation occurred at 0 h and 2 h after incubation. Membrane integrity, mitochondrial membrane potential （MMP） and DNA damages were evaluated by flow cytometry; activities of antioxidant enzymes such as catalase, superoxide dismutase and gluthatione peroxidase were measured by spectrofotometry. Results： There was an increase in the percentage of sperm with DNA damage in the thawed group, compared to fresh and cooled, and for 2 hs of incubation when compared to 0 h. Considering MMP, there was an increase in the percentage of cells with medium potential in thawed group when compared to fresh and cooled groups. Opposingly, a decrease was observed in the thawed group considering high mitochondrial potential. Also in the thawed group, there was an increase on cells with damaged acrosome and membrane when compared to fresh and cooled groups. Significant correlations were found between antioxidant enzymes activity and membrane or mitochondrial parameters. Conclusion： Based on our results, we conclude that cryopreservation affects cellular and DNA integrity and that the critical moment is when sperm cells are exposed to freezing temperature. Also, our study indicates that intracellular antioxidant machinery （SOD and GPX enzymes） is not enough to control cryodamage.

Dosage Effects of Fadrozole on Growth and Development of Sex-Reversed Genetic Female Chickens

Fadrozole, an aromatase inhibitor, can masculinize genetic female chickens and high-dose decreases the hatchability. Therefore, it is important to study the growth and development of sex-reversed females after hatch. Chick embryos from a population of CAU3 egg-type were treated with different concentrations of Fadrozole prior to the sexual differentiation at E3.0 （st18）. At hatch, the phenotypic sex and genetic sex were identified by vent sexing and genetic diagnosis with CHD1, respectively. Body weight and shank length of sex reversal were tested at 8 and 20 wk, respectively. Testicular development, oviduct and ovarian degeneration were observed and serum concentration of estradiol and testosterone were tested with radioimmunoassay （RIA） at 30 wk. The results showed that body weight and shank length of sex- reversed females were not significantly different between low-dose groups （0.1, 0.3, and 0.5 mg for F1, F2, and F3, respectively） and high-dose groups （1.0 and 1.3 mg for F4 and F5, respectively）（P〉0.05）. Left and right testes or ovotestes in F2, F3, F4, and F5 groups were heavier than that of in F1 group （P〈0.05）. While the gonad weight of treatment groups were less than that in male control （P〈0.05）, oviduct weight in F2, F3, F4, and F5 groups were significant differences compared with female control and F1 group （P〈0.05）. Egg number from onset of laying egg to 30 wk in F4 and F5 groups were less than in female control, F1 and F2 groups （P〈0.05）. Serum testosterone level in F5 group was significant higher compared with female control, F1, F2, F3, and F4 groups （P〈0.05）, but significant lower compared with male control （P〈0.05）. While concentration of serum estradiol in F5 group was significant lower compared with female control, F1, F2, and F4 groups （P〈0.05）. In conclusion, the concentration of Fadrozole do not affect postnatal growth of sex-reversed female chicken and the degree of sex-reversed females elevate with the increase of Fadrozole concentration at

The prolonged effect of glucagon-like peptide 2 pretreatment on growth performance and intestinal development of weaned piglets

Background： Glucagon-like peptide 2 （GLP-2） is a potent epithelium-specific intestinal growth factor. The aim of this study was to demonstrate the prolonged effect of GLP-2 on the growth performance of weaned piglets. Forty piglets weaned at the age of 28 d with an average BW of 6.8 ＋ 0.4 kg were assigned to four treatments：（i） non- challenged control; （ii） LPS-challenged control; （iii） LPS ＋ low GLP-2; and （iv） LPS ＋ high GLP-2. Piglets in groups （i）, （ii）, and （iv） were s.c. injected with PBS supplemented with human [Gly2]GLP-21-34 at doses of 0, 2 and 10 nmol/kg BW per day for seven consecutive days. BW, gain：feed ratio （G：F）, and plasma GLP-2 levels were determined on d 0 7, and 14 after weaning. Piglets were challenged with i.p. administration of Escherichia coil lipopolysaccharide （LPS） at a dose of 100 pg/kg on d 14 to induce intestinal damage. Twenty-four hours later, intestinal tract samples were collected to assess intestinal morphology and quantify enzyme activity. Results： Plasma GLP-2 levels decreased after weaning, but in the high GLP-2 group, plasma GLP-2 was maintained on d 7 and even increased to a level higher than the preweaning level on d ]4 （P 〈 0.05）. High GLP-2 treatment significantly increased the duodenal, jejunal and ileal weight, as well as the gross weight of the small intestine （SI）, and the SI weight index （P 〈 0.05）. LPS caused villous atrophy and disrupted intestinal morphology in the duodenum, jejunum and ileum. GLP-2 also significantly increased the villus height and the villus height/crypt depth ratio （VCR） of the duodenum, jejunum, and ileum （P 〈 0.05）. Histological examination revealed that in GLP-2-treated groups, the integrity of the villus was maintained, and the villus was protected against LPS-induced damage. Effect of lactic acid bacteria inoculants on alfalfa(Medicago sativa L.) silage quality:assessment of degradation(in situ) and gas production(in vitro)

Alfalfa （Medicago sativa） is difficult to ensile successfully because of the low content of moisture and water-soluble carbohydrates （WSC） in fresh alfalfa and the high buffering capacity in fresh alfalfa. Here, we conducted a study to evaluate the effects of three lactic acid bacteria （LAB） inoculants （Lactobacillus case/, lactobacillus plantarum, and Pediococcus pentosaceus） on silage quality, in sitE/ruminal degradability, and in vitro fermentation of alfalfa silage. The first cut of alfalfa was wilted, chopped, and randomly divided into four groups： the control （CON） and control mixed with three separate LAB inoculants （106 cfu g-1）. Simmental steers with a body weight of （452±18） kg and with installed rumen fistulas were prepared for in situ degradation and for in vitro gas production. LAB inoculants had a lower （P〈0.05） content of butyric acid than the CON group. Among them, the L. casei inoculated silage had a higher （P〈0.05） content of water-soluble carbohydrate （WSC） and a lower （P〈0.05） NH3-H content. The effective degradation （ED） of crude protein in LAB inoculation decreased （P〈0.05）, while the ED of acid detergent fiber increased （P〈0.05） in situ fermentation. The alfalfa silage with LAB inoculants produced more carbon dioxide （P〈0.05）. The NH3-H content of mixed incubation fluid in L. casei inoculated silage was lower （P〈0.05） compared with other groups. Therefore, this study showed that LAB inoculants could improve both ensiling quality and degradation. In particular, the L. casei inoculations exhibited better performance by limiting proteolysis during ensiling.

Impact of dietary supplementation with β-alanine on the rumen microbial crude protein supply, nutrient digestibility and nitrogen retention in beef steers elucidated through sequencing the rumen bacterial community

This study investigated the effects of β-alanine(β-Ala) on rumen fermentation, nutrient digestibility,nitrogen(N) metabolism, plasma biochemical parameters, and rumen bacterial communities in beef steers. Six steers with initial liveweight of 252.8 ± 5.2 kg and 3 treatments of supplementing with 0, 30,or 60 g β-Ala per day to basal diet were allocated in a replicated 3 × 3 Latin square design. Each experimental period was 20 d, of which the first 15 d were for adaptation and the subsequent 5 d were for sampling. The results showed that β-Ala linearly increased the ruminal concentration of microbial crude protein(MCP)(P = 0.005), but it did not affect the ruminal concentrations of ammonia N and total volatile fatty acids(P > 0.10). β-Ala also linearly increased the dry matter(DM)(P = 0.009), organic matter(OM)(P = 0.017) and crude protein(CP)(P = 0.043) digestibility, tended to decrease the acid detergent fiber digestibility(P = 0.077), but it did not affect the neutral detergent fiber digestibility(P = 0.641). β-Ala quadratically increased the relative abundance of ruminal Bacteroidota(P = 0.021) at the phylum level, and increased Prevotella(P = 0.028) and Prevotellaceae＿UCG-003(P = 0.014), and decreased the relative abundance of NK4A214＿group(P = 0.009) at the genus level. Feeding steers with β-Ala linearly increased the urinary N(P = 0.006), urea excretions(P = 0.002) and the N retention(P = 0.004), but it did not affect the N utilization efficiency(P = 0.120). β-Ala quadratically increased the plasma concentration of the total antioxidant capacity(P = 0.011) and linearly increased the plasma concentration of insulin-like growth factor-1(P < 0.001). Analysis of the Genetic Diversity and Origin of Some Chinese Domestic Duck Breeds

Twelve fluorescence-labeled microsatellite markers were used to analyze the genetic diversity of 12 domestic duck breeds and 2 wild duck breeds to determine the relationship and origin of Chinese domestic duck breeds. Gene frequency, effective number of alleles （Ne）, expected heterozygosity （He）, polymorphism information contents （PIC）, inbreeding coefficient in population （Fis）, standard genetic distance （Ds）, and genetic distance （DA） were calculated by FSTAT and distance and phylogenetic analysis after the dates which were output from the Microsatellite-Toolkit software. Genetic distances between 12 domestic duck breeds and 2 wild duck breeds were analyzed by variance analysis. Unweighted pair group method with arithmetic mean （UPGMA） and phylogenetic trees used for cluster analysis were structured. The results indicated that 11 loci had medium- or high-level genetic diversity among the 12 loci, which could be efficiently used in the detection of the genetic parameters of each population. The values of He were 0.5414 to 0.7343, those of PIC proved similar, and those of Fis were 0.1101 to 0.3381 among all populations. All breeds were clustered into three groups by UPGMA phylogenetic trees. Banzui duck was clustered into a separate group. Differences of the DA were analysed by t-test. The results showed that difference in DA between the 12 domestic duck breeds and Lvtou duck and the Banzui duck were very significant （P〈0.01）, indicating that these 12 domestic duck breeds originated from Lvtou wild duck, but not Banzui duck.

Effect of reduced energy density of close-up diets on dry matter intake,lactation performance and energy balance in multiparous Holstein cows

Energy intake prepartum is critically important to health, milk performance, and profitability of dairy cows. The objective of this study was to determine the effect of reduced energy density of dose-up diets on dry matter intake （DMI）, lactation performance and energy balance （EB） in multiparous Holstein cows which were housed in a free-stall barn and fed for ad libitum intake. Thirty-nine dry cows were blocked and assigned randomly to three groups fed a high energy density diet [HD, n = 13; 6.8 MJ of net energy for lactation （NEL）/kg; 14.0% crude protein （CP） ], or a middle energy density diet （MD, n = 13; 6.2 MJ NEh/kg; 14.0% CP）, or a low energy density diet （LD, n = 13; 5.4 MJ NEh/kg; 14.0% CP） from d 21 before expected day of calving. After parturition, all cows were fed the same lactation diet to d 70 in milk （DIM）. The DMI and NEE intake prepartum were decreased by the reduced energy density diets （P 〈 0.05）. The LD group consumed 1.3 last 24 h before calving. The milk yield and the postpartum kg/d （DM） more diet compared with HD group in the DMI were increased by the reduced energy density diet prepartum （P 〈 0.05）. The changes in BCS and BW prepartum and postpartum were not affected by prepartum diets HD group had higher milk fat content and lower lactose content compared with LD group during the first 3 wk of lactation （P 〈 0.05）. The energy consumption for HD, MD and LD groups were 149.8%, 126.2% and 101.1 % of their calculated energy requirements prepartum （P 〈 0.05）, and 72.7%, 73.1% and 7.5.2% during the first 4 wk postpartum, respectively. In conclusion, the low energy density prepartum diet was effective in controlling NF_L intake prepartum, and was beneficial in increasing DMI and milk yield, and alleviating negative EB postpartum.
