Eco-friendly reduced graphene oxide for the determination of mycophenolate mofetil in pharmaceutical formulations

Graphene oxide(GO) was synthesized and characterized by scanning electron microscopy(SEM), energy dispersive X-ray spectroscopy(EDX), atomic force microscopy(AFM), X-ray diffraction(XRD), Fourier transform-infrared spectroscopy(FT-IR) and thermogravimetric analysis(TGA). GO was then electrochemically reduced and used for electrochemical study of mycophenolate mofetil(MMF). The electrochemically reduced graphene oxide(ERGO) film on glassy carbon electrode(GCE) showed enhanced peak current for electrooxidation of MMF. MMF exhibited two irreversible oxidation peaks at 0.84 V(peak a_1)and 1.1 V(peak a_2). Effects of accumulation time, pH and scan rate were studied and various electrochemical parameters were calculated. A differential pulse voltammetric method was developed for the determination of MMF in bulk samples and pharmaceutical formulations. Linear relationship was observed between the peak current and concentration of MMF in the range of 40 nM–15 μM with a limit of detection of 11.3 nM. The proposed method is simple, sensitive and inexpensive and, hence, could be readily adopted in clinical and quality control laboratories.

A new polyglycerol phthalate extractant was prepared and characterized using Fourier transform infrared spectroscopy, proton nuclear magnetic resonance(~1H NMR) spectroscopy, and thermogravimetric analysis. A preliminary study of the extraction of long-lived(^(90) Sr and ^(152+154)Eu) and short-lived(^(90) Y) radionuclides by polyglycerol phthalate was performed. The influence of different diluents, concentration of the acid, type of acid, and various interfering ions was explored. The stripping of ^(152+154)Eu radionuclide was studied by using different acid solutions, and it was found that sulfuric acid was the ideal acid for this purpose. Furthermore,^(152+154)Eu was extracted from the fission products(^(90) Sr) and the extraction% of^(90) Sr was found to improve in the presence of interfering ions.

A direct drive actuator （DDA） with direct drive valves （DDVs） as the control device is an ideal solution for a flight actuation system. This paper presents a novel triple-redundant voice coil motor （TRVCM） used for redundant DDVs. The TRVCM features electrical/mechanical hybrid triple-redundancy by securing three stators along with three moving coils in the same frame. A permanent magnet （PM） Halbach array is employed in each redundant VCM to simplify the system structure. A back-to-back design between neighborly redundancies is adopted to decouple the magnetic flux linkage. The particle swarm optimization （PSO） method is implemented to optimize design parameters based on the analytical magnetic circuit model. The optimization objective function is defined as the acceleration capacity of the motor to achieve high dynamic performance. The optimal geometric parameters are verified with 3D magnetic field finite element analysis （FEA）. A research prototype has been developed for experimental purpose. The experimental results of magnetic field density and force output show that the proposed TRVCM has great potential of applications in DDA systems.

Carnuba wax consists chiefly of myricyl cerotate (MW 817.4) and small quantities of free cerotic acid (C26H52O2, Mw 396.7) and myricyl alcohol (C30H62O, mp 90°C). Of the two common extraction solvents, ethyl ether or petroleum ether, Lewkowitsch prefers the former. Concerning separation of phases, he advocates addition of small amounts of alcohol or caustic, and he also states that formation of a flocculant layer between the aqueous layer and the solvent does not interfere with the correct estimation of the unsaponifiable matter. These statements were not corroborated in the hands of this chemist. The “Unsaponifiable Matter” in oils or and fats, which consist mainly of hydrocarbons, sterols and aliphatic alcohols of high molecular mass that are not saponifiable by alkali hydroxides but are soluble in the ordinary fat solvents, and to products of saponification that are soluble in such solvents. Carnuba wax, a rather expensive wax, may be adulterated with less expensive paraffin by dishonest merchants. ASTM has a method for determining paraffinic material in carnuba wax. It uses heptanes at its boiling point to dissolve the wax, apply it to a silica gel column, and elute only the nonpolar (i.e. alkane) material. The method has the disadvantage of using a large volume of haptane, nor is it called for by either US Pharmacopeia/National Formulary (USP/NF) or Food Chemicals Codex (FCC). The test for unsaponifiable matter on pure carnuba wax will yield a result of 50.0% - 55.0%, while a higher result will betray the presence of paraffin adulterants.

Electrogenerated chemiluminescence(electrochemiluminescence, ECL) generates species at electrode surfaces, which undergoes electron-transfer reactions and forms excited states to emit light. It has become a very powerful analytical technique and has been widely used in such as clinical testing, biowarfare agent detection, and pharmaceutical analysis. This review focuses on the current trends of molecular recognition-based biosensing methods for pharmaceutical analysis since 2010. It introduces a background of ECL and presents the recent ECL developments in ECL immunoassay(ECLIA), immunosensors, enzyme-based biosensors, aptamer-based biosensors, and molecularly imprinted polymers(MIP)-based sensors. At last, the future perspective for these analytical methods is briefly discussed.

The development and delivery of high quality therapeutic products necessitates the need for highthrough-put (HTP) process development tools. Traditionally, these works requires a combination of shake flask and small-scale stirred tank bioreactor (STR) which are labor and resource intensive and time-consuming. Here we demonstrate a strategy for rapid and robust cell culture process development by evaluating and implementing the use of a new HTP disposable micro bioreactor (MBR) called AMBRTM system (Advanced Microscale Bioreactor) that has the capabilities for automated sampling, feed addition, pH, dissolved oxygen (DO), gassing and agitation controls. In these studies the performance of two monoclonal antibody (MAb) producing cell lines (MAb1 and MAb2) was evaluated both in the AMBR system and 3-L STR. We demonstrated that cell culture performance (growth and viability, production titer and product quality) were similar in both vessel systems. Furthermore, process control and feed optimization were demonstrated in an additional cell line (MAb3) in the disposable MBR and its performance confirmed at STR scale. The results indicate that the AMBR system can be used to streamline the process development effort and facilitate a rapid and robust cell culture process development effort for MAb programs in a HTP manner.
