Three-dimensional laser writing inside silicon remains today inaccessible with the shortest infrared light pulses unless complex schemes are used to circumvent screening propagation nonlinearities.Here,we explore a new approach irradiating silicon with trains of femtosecond laser pulses at repetition rates up to 5.6 THz that is order of magnitude higher than any source used for laser processing so far.This extremely high repetition rate is faster than laser energy dissipation from microvolume inside silicon,thus enabling unique capabilities for pulse-to-pulse accumulation of free carriers generated by nonlinear ionization,as well as progressive thermal bandgap closure before any diffusion process comes into play.By space-resolved measurements of energy delivery inside silicon,we evidence changes in the interplay between detrimental nonlinearities and accumulation-based effects.This leads to a net increase on the level of space-time energy localization.The improvement is also supported by experiments demonstrating high performance for 3D laser writing inside silicon.In comparison to repeated single pulses,irradiation with trains of only four-picosecond pulses with the same total energy leads to an apparent decrease of the energy threshold for modification and drastic improvements on the repeatability,uniformity,and symmetricity of the produced features.The unique benefits of THz bursts can provide a new route to meet the challenge of 3D inscription inside narrow bandgap materials.

Water and air quality of eight seawater swimming pools using chlorine disinfection was measured during four sampling campaigns, spread on one full-year, and in four thalassotherapy centers located in Southeast of France. Concentrations of trihalomethanes（THMs） in air and in water as well as concentrations of parameters, including nonpurgeable organic carbon（NPOC）, free residual chlorine（Cl_f）, pH, Kjeldhal Nitrogen（KN）, salinity,conductivity, bromide ions and, water and air temperature, were measured. Water and air samples were collected in triplicates morning — at the opening of the pools —, noon and night — at the closing of the pools —, in summer and winter. Data analysis was performed by Principal Component Analysis（PCA） and rotated component matrix, from both data quality and other parameters such as TOC, aromaticity（UV_（254））, pH, hygrometry, and free residual chlorine（Cl_f）. This statistical analysis demonstrates a high correlation between TOC, Cl_fand UV_（254） and THM levels found in air and water, particularly for the major ones（CHBr_3in water：300.0 μg/L mean, 1029.0 μg/L maximum; CHBr_3 in air： 266.1 μg/m^3 mean,1600.0 μg/m^3 maximum, and CHClBr_2 in water： 18.9 μg/L mean, 81.0 μg/L maximum;CHClBr_2 in air： 13.6 μg/m^3 mean, 150.0 μg/m^3maximum）. These high levels of bromoform（CHBr_3） are particularly worrisome in such health institutions, even these levels do not exceed the Permissible Exposure Limit（PEL） of 5 mg/m^3 as an 8 hour time-weighted average currently fixed by various administrations, such as Occupational Safety and Health Administration（OSHA）.

Glucocorticoids(GCs)are endogenous hormones that are crucial for the homeostasis of the organism and adaptation to the external environment.Because of their anti-inflammatory effects,synthetic GCs are also extensively used in clinical practice.However,almost all cells in the body are sensitive to GC regulation.As a result,these mediators have pleiotropic effects,which may be undesirable or detrimental to human health.Here,we summarize the recent findings that contribute to deciphering the molecular mechanisms downstream of glucocorticoid receptor activation.We also discuss the complex role of GCs in infectious diseases such as sepsis and COVID-19,in which the balance between pathogen elimination and protection against excessive inflammation and immunopathology needs to be tightly regulated.An understanding of the cell type-and context-specific actions of GCs from the molecular to the organismal level would help to optimize their therapeutic use.

Laser-induced damage(LID)on high-power laser facilities is one of the limiting factors for the increase in power and energy.Inertial confinement fusion(ICF)facilities such as Laser Mégajoule or the National Ignition Facility use spectral broadening of the laser pulse that may induce power modulations because of frequency modulation to amplitude modulation conversion.In this paper,we study the impact of low and fast power modulations of laser pulses both experimentally and numerically.The MELBA experimental testbed was used to shape a wide variety of laser pulses and to study their impact on LID.A 1D Lagrangian hydrodynamic code was used to understand the impact of different power profiles on LID.

Scanning fluorescence microscopes are now able to image large biological samples at high spatial and temporal resolution.This comes at the expense of an increased light dose which is detrimental to fluorophore stability and cell physiology.To highly reduce the light dose,we designed an adaptive scanning fluorescence microscope with a scanning scheme optimized for the unsupervised imaging of cell sheets,which underly the shape of many embryos and organs.The surface of the tissue is first delineated from the acquisition of a very small subset(~0.1%)of sample space,using a robust estimation strategy.Two alternative scanning strategies are then proposed to image the tissue with an improved photon budget,without loss in resolution.The first strategy consists in scanning only a thin shell around the estimated surface of interest,allowing high reduction of light dose when the tissue is curved.The second strategy applies when structures of interest lie at the cell periphery(e.***.adherens junctions).An iterative approach is then used to propagate scanning along cell contours.We demonstrate the benefit of our approach imaging live epithelia from Drosophila melanogaster.On the examples shown,both approaches yield more than a 20-fold reduction in light dose-and up to more than 80-fold-compared to a full scan of the volume.These smart-scanning strategies can be easily implemented on most scanning fluorescent imaging modality.The dramatic reduction in light exposure of the sample should allow prolonged imaging of the live processes under investigation.

The preparation of medium-sized benzo[b]azocines has always been challenging because of inherently unfavorable enthalpy and entropy factors.This report presents a novel approach for accessing 8-membered seleno-benzo[b]azocines via electrochemically-driven seleno-cyclization.This method enables room-temperature preparation of various structurally diverse medium-sized seleno-benzo[b]azocines.The facile deselenation of the seleno-cyclization products to generate functionalized dienes is an additional benefit of this indispensable reaction.Mechanistic insights are presented based on radical inhibition experiments and cyclic voltammetry measurements,which elucidate the radical pathway.Finally,density functional theory calculations further rationalize the rate-determining step and the unique chemoselectivity observed in this transformation.

初始T细胞活化是由T细胞受体(TCR)及共刺激分子与其配体结合引起的,其中CD28共刺激信号在促进初始T细胞的活化过程中起关键作用.本文研究了小鼠CD4^(+)T细胞中CD28与B7配体的结合活性在CD28介导的信号通路激活中的调控机制.结果发现虽然初始CD4^(+)T细胞表面CD28-B7的本底结合力很低,但当同步给予TCR刺激可触发CD28胞内域激酶SRC酪氨酸磷酸化,并由此显著增强CD28-B7的结合力.此外通过CD28胞内域缺失的小鼠模型,进一步验证CD28胞内结构域的存在是TCR刺激信号增强CD28-B7结合力的必要条件.据此,本研究证实CD28胞内域在其配体结合过程中承担了关键性的调控作用,并为TCR和CD28相互作用介导初始T细胞活化提供了新的证据.

In this paper, the authors prove an analogue of Gibbons' conjecture for the extended fourth order Allen-Cahn equation in R^N, as well as Liouville type results for some solutions converging to the same value at infinity in a given direction. The authors also prove a priori bounds and further one-dimensional symmetry and rigidity results for semilinear fourth order elliptic equations with more general nonlinearities.

Finding effective therapies for hepatocellular carcinoma(HCC)is an ongoing challenge for both researchers and clinicians.Enthusiasm for new treatment options emerging from in vitro,preclinical,and clinical studies is often tempered by limited patient benefits and associated side effects,tolerated in some cases due to the lack of alternatives.The complexity of identifying effective treatment options resides in several features of HCC,notably the extraordinary heterogeneity in molecular alterations among different patients(1-3),the lack of consistent alterations associated with oncogene addictions(4),and a so-called“non-oncogene addiction”by which tumorigenicity results from the action of non-mutated genes(5,6).This complex configuration also prevents identification of unequivocal alterations used to design therapies,as for some other cancer types.Current HCC treatments are based on the Barcelona Clinic Liver Cancer(BCLC)guidelines according to the stage of lesions among other parameters.For HCC at early stages(30-40%of HCC patients),treatments include lesion resection,liver transplantation,radiofrequency ablation(RFA)and microwave ablation(MWA),achieving a median overall survival(OS)of more than 60 months.For HCC at intermediate stages(~20%of HCC patients),the advised treatment is generally transarterial chemoembolization(TACE),achieving an OS of 20 months.For HCC at advanced stages(~40%of HCC patients),targeted therapies are the most frequent treatments,leading to an OS of 11 months with most targeted monotherapies.For HCC at terminal stages,only palliative care is provided(~10%of HCC patients),with less than 3 months OS(7,8).
