Biotinylated dextran amine tracing of nerve tracts determines regeneration of corticospinal tracts after neural stem cell transplantation

We investigated regeneration of the corticospinal tract and rearrangement of corticospinal nerves after spinal cord injury by biotinylated dextran amine （BDA） nerve tract tracing after neural stem cell transplantation. Neural stem cell transplantation increased motor function scores of rats at 3 weeks after spinal cord transection injury at the thoracic 10 segment. A proportion of BDA-labeled corticospinal tract regenerated through the spinal cord injury site at 12 weeks after transplantation. Electron microscopy revealed that the regenerated BDA-labeled nerve terminals formed new synaptic connections with neurons at the distal end of the injured site. These findings indicate that BDA nerve tract tracing effectively provides anatomic and morphological evidence of recovery after spinal cord injury.

肌肉减少症(简称“肌少症”)是一种与增龄相关、以肌肉含量减少、肌肉力量下降和(或)躯体功能障碍为主要特征的老年综合征,可增加衰弱、跌倒、残疾甚至死亡等不良结局的发生风险,疾病负担沉重。规范肌少症的诊断及治疗对开展临床工作和促进我国健康老龄事业发展具有重要意义,但目前我国尚缺乏基于循证医学证据的肌少症临床诊疗指南。基于此,由国家老年疾病临床医学研究中心(湘雅医院)牵头,在中华医学会老年医学分会组织下,遵循《中国制订/修订临床诊疗指南的指导原则(2022版)》和国际实践指南报告规范(RIGHT)等指南制订指导文件,遴选出肌少症领域最为关注的12个重要临床问题。采用推荐意见分级的评价、制定及评估(GRADE)体系评估证据强度,并运用德尔菲共识法确定推荐强度,最终形成18条关于肌少症诊疗的循证医学推荐意见。本指南旨在提高我国肌少症诊疗的科学性,并推动我国肌少症科研工作的深入发展,以期最终提升以患者为中心的医疗服务质量。

Objective To explore the therapeutic effect of naringin on colorectal cancer(CRC)and the related *** Cell counting kit-8(CCK-8)assay and annexin V-FITC/PI assay were used to detect the effect of naringin(50–400µg/mL)on cell proliferation and apoptosis of CRC cells,*** scratch wound assay and transwell migration assay were used to assess the effect of naringin on CRC cell ***-week-old male nude mice were injected with HCT116 cells subcutaneously to establish the tumor xenograft *** was injected intraperitoneally at 50 mg/(kg·d),with solvent and 5-fluorouracil treatment as *** width and length of the tumors were measured and recorded every 6 days,and tumor tissues were photographed and weighed on the last day of the 24-d observation *** staining for caspase-3,proliferating cell nuclear antigen and TUNEL assay were used to evaluate the effect of naringin on cell proliferation and apoptosis in tumor *** body weight,food and water intake of mice were recorded,and the major organs in different treatment groups were weighed on the last day and stained with hematoxylin and eosin for histological ***,the routine blood indicators were *** CCK-8 and annexin V-FITC/PI results confirmed that naringin(100,200,and 400µg/mL)could inhibit proliferation and promote *** scratch wound assay and transwell migration assay results confirmed the inhibitory activity of naringin against CRC cells *** vivo results demonstrated the inhibitory effect of naringin on tumor growth with good *** Naringin inhibited colorectal carcinogenesis by inhibiting viability of CRC cells.

BACKGROUND： Studies have shown that latency changes of some elements in a somatosensory evoked potential （SEP） and motor evoked potential （MEP） can reflect electrical activity of cerebral cortical neurons and conduction of white matter nerve fibers. However, there is a paucity of information regarding the dynamic observation of SEP and MEP following cerebral ischemic injury. OBJECTIVE： To explore SEP and MEP changes following acute ischemic stroke, and investigate the role of evoked potentials in monitoring brain function in stroke. DESIGN, TIME AND SETTING： A randomized, controlled, animal experiment was performed at the Chongqing Key Laboratory of Neurology, Affiliated Hospital of Chongqing Medical University from September 2007 to August 2008. MATERIALS＂ Hydrogen blood flow detector was purchased from Soochow University Medical Instrument Co., China, and Power lab system was purchased from AD Instruments, Inc., USA. METHODS A total of 36 healthy, adult, male, Sprague Dawley rats were randomly assigned to four groups （n = 9）, including three ischemia groups （12, 24 and 72 hours of ischemia） and a sham-surgery group. The rat model of acute ischemic stroke was established by middle cerebral artery occlusion （MCAO） in the left hemisphere. MAIN OUTCOME MEASURES＂ SEP and MEP of the left limbs were detected, and cerebral blood flow was measured by the hydrogen cleaning method. RESULTS： The latency of positive wave 1 （P1）, negative wave 1 （N1） and positive wave 2 （P2） waves in SEP, and latency of negative wave 1,2 （N1, N2） waves in MEP were significantly prolonged with increasing ischemic duration following MCAO （P 〈 0.01）, but cerebral blood flow was significantly decreased （P 〈 0.05, or P 〈 0.01）. CONLUSION： Ischemic stroke prolongs the latency of SEP waves （P1, N1, P2） and MEP waves （N1, N2）, and cerebral cortical evoked potential may correlate with cerebral blood flow changes. Objective To develop a plasma pharmacological method evaluates the effect of Da Huang Zhe Chong capsule on platelet aggregation and its mechanism, which is a representative Traditional Chinese Medicine Patent Prescription Promoting blood circulation by removing blood stasis. Methods Platelets specimens from healthy volunteers made serum and plasma with medicine, while platelet PRP were separated, which were divided into 8groups,i.e. auto-serum, allo-serum, serum with Da Huang Zhe Chong capsule , serum with aspirin, auto-plasma, plasma with Da Huang Zhe Chong capsule, plasma with aspirin, every group added to serum and plasma to hatch. After ADP and adrenalin were added into the specimens and hatched, the effects of specimens on platelet aggregation were observed. Results After ADP adrenalin were added, all the serum groups did not present platelet aggregation,while all the plasma group presented platelet aggregation. P1, P5, Pmax, t and TM have no significant difference (P＞0. 05) between auto-plasma group and allo-plasma group induced by ADP and adrenalin. P1, P5, t and Pmax have significant differences (P＜0. 01) and TM decreased significantly (P＜0. 05) comparing plasma group with Da Huang Zhe Chong capsule and plasma group of aspirin to allo-plasma group. P1, t and Pmax have significant difference (P＜0.05), and P5 and TM are simulate comparing plasma group with Da Huang Zhe Chong capsule to plasma group of aspirin. P1, P5, t and Pmax have significant differences (P＜0. 01), P1, TM have also significant(P＜0. 05), comparing plasma group of Da Huang Zhe Chong capsule with plasma of aspirin to allo-plasma group induced by adrenalin. BACKGROUND： To this date, specific molecular markers for early diagnosis and prognosis monitoring of craniocerebral injury in clinical medicine do not exist. Therefore, differential detection of specific proteins might play an important role in diagnosis and treatment of this type of brain injury. OBJECTIVE： To compare differential cerebral cortical protein expression of craniocerebral injury patients and normal subjects through the use of proteomics. DESIGN： Contrast observation. SETTING： Department of Neurosurgery, Xiangya Hospital of Central South University. PARTICIPANTS： Ten patients （6 males and 4 females, 20 58 years old）, with severe craniocerebral injury, were selected at the Department of Neurosurgery, Xiangya Hospital of Central South University, from June 2004 to December 2006. All patients were diagnosed with CT test and Glasgow test （scores 〈 8）. Surgery was performed 4-12 hours after craniocerebral injury, and injured cortical tissues of the frontal and temporal lobes were resected for sampling. At the same time, control cortical tissues were collected from frontal and temporal lobes of 2 epileptic patients who underwent hippocampus-nucleus amygdala resection, and 2 lateral ventricular tumor patients who underwent tumor resection. The participants and their relatives provided confirmed consent, and this study received confirrned consent from the local ethics committee. METHODS： Ten samples from injured patients and 4 normal samples were compared through the use of proteomics. Total protein was separated by using two-dimensional electrophoresis with immobilized pH gradients, and the differential protein expressions were compared using image analysis after blue-sliver staining. Differential protein spot expressions were analyzed with a matrix-assisted laser desorption/ionization time of flight mass spectrometry （MALDI/TOF MS） and electrospray ionization-quadrupole time of flight mass spectrometry （ESI-Qq TOF MS）.
