The objective of this prospective study of the risks of treatment failure in patients with drug-susceptible pulmonary tuberculosis （PTB） was to provide reference data to help develop a disease control strategy. Participants were recruited in eight provinces of China from October 2.08 to December 2.10. A total of 1447 patients with drug-susceptible PTB and older than 15 years of age were enrolled.

BACKGROUND Endoscopic submucosal dissection(ESD)is an established technique for the treatment of early gastrointestinal ***,multi-day(M-D)admission is required for patients undergoing ESD due to potential *** To evaluate the feasibility of a same-day(S-D)discharge strategy for ESD of the esophagus or *** The data of patients who underwent esophageal or gastric ESD were retrospectively collected from January 2.18 to December 2.2. at Peking University Cancer *** propensity score matching(PSM)method was applied to balance the unevenly distributed patient baseline characteristics between the S-D and M-D *** and postoperative parameters were compared between the matched *** Among the 479 patients reviewed,470 patients,including 91 in the S-D group and 379 in the M-D group,fulfilled the inclusion and exclusion *** PSM,78 patients in each group were paired using the 1:1 nearest available score match *** significant difference was found between groups with respect to intraoperative and postprocedural major adverse events(AEs).tumor size,complete resection rate,and procedural duration were comparable between the *** S-D group demonstrated a significantly shorter length of hospital stay(P<0.001)and lower overall medical expenses(P<0.001)compared with the M-D *** The S-D discharge strategy may be feasible and effective for esophagogastric ESD,and the procedural-related AEs can be managed successfully.

3D genomics mainly focuses on the 3D position of single genes at the cell level, while spatial genomics focuses more on the tissue level. In this exciting new era of 3D/spatial genomics, half-century old FISH and its derivative methods, including Tn5-FISH, play important roles. In this review, we introduce the Tn5-FISH we developed recently, and present six different applications published by our collaborators and us, based on(Tn5-)FISH, which can be either general BAC clone-based FISH or Tn5-FISH. In these interesting cases,(Tn5-)FISH demonstrated its vigorous ability of targeting sub-chromosomal structures across different diseases and cell lines(leukemia, m ESCs(mouse embryonic stem cells), and differentiation cell lines). Serving as an effective tool to image genomic structures at the kilobase level,Tn5-FISH holds great potential to detect chromosomal structures in a high-throughput manner, thus bringing the dawn for new discoveries in the great era of 3D/spatial genomics.

Special Expression of Thy-1 in Different Malignant tumors

Objective： To investigate the expression status of Thy-1 in malignant cancerous tissues and to evaluate whether Thy-1 could be a potential tumor marker. Methods： Immunohistochemistry method was used to examine Thy-1 expressions in different malignant tumor tissues. We used a monoclonal antibody specific for Thy-1 and the indirect Catalysis Signal Amplification method. With the help of tissue microarray （TMA）, we examined expression the status of Thy-1 in 1451 different types of malignant tumor tissues, 144 normal tissues and benign lesions. Results： All the malignant tumor cells were grouped as one group （malignant cells group） and normal or benign tumor tissue cells as another group （normal cells group）. Among the entire 1451 malignant cases group, positive Thy-1 expression with diffuse and strong staining was observed in 734 （734/1451; 50.59%） cases. In the benign cases only 6 chronic cervicitis cases showed weak staining for Thy-1. All the normal tissue showed negative staining. One-Way ANVOA analysis showed F value between these two groups was 147.2.9 （P〈0.0001）. Conclusion： A significantly stronger expression of Thy-1 in malignant tumors was observed. Special overexpression of Thy-1 in malignant tumors suggests that Thy-1 might be a potential novel tumor marker with respect to cancer progression. Chronic cervicitis has some relationship with cervix carcinoma. When it develops into atypical hyperplasia, it will be a precancerous lesion for cervix carcinoma. In this research we found weak staining for Thy-1 in chronic cervicitis lesion, so Thy-1 may play a crucial role in carcinogenesis for cervix carcinoma. The research about the relationship between Thy-1 expressions in cancer cells and in precancerous lesion will provide some clues to understand the mechanism for carcinogenesis process.

Expression and significance of cyclooxygenase-2.in human lung cancer

Objective: We aimed to examine the expression of cyclooxygenase-2.and the relationship with the pathological types, TNM stage, lymph node metastasis, the degree of differentiation, smoking and the survival. Methods: Immunohistochemical staining method was used to examine the expression of cyclooxygenase-2.of 12. cases of lung cancer and three control groups. The data were statistically analyzed. Results: Compared with the health group, cyclooxygenase-2.was over expressed in the inflammatory tissue(P = 0.036), lung adenocarcinoma(P = 0.005) and squamous carcinoma(P = 0.047). Compared with patients without lymph node metastasis, cyclooxygenase-2.was over expressed(P = 0.033) in patients with lymph node metastasis. Compared with high differentiation group, cyclooxygenase-2.was over expressed(P = 0.004) in low differentiation group. Compared with non-smokers, the expression of cyclooxygenase-2.increased in smokers(P = 0.000). The median survival time of patients that the expression of cyclooxygenase-2.were negative was 9 months(95% CI, 5.6–12.4 months). The median survival time of patients that the expression of cyclooxygenase-2.were positive was 5 months(95% CI, 3–7 months). They was statistical difference(P = 0.001). Conclusion: Overexpression of cyclooxygenase-2.is associated with pathological types, TNM stage, lymph node metastasis, degrees of differentiation, smoking and prognosis in lung cancer.

Cancer stem cells(CSCs)exhibit highly aggressive and metastatic features and resistance to chemotherapy and *** hydrocarbon receptor(AhR)expression varies among non-small cell lung cancers(NSCLCs),and the mechanisms that support abnormal AhR expression in CSCs remain ***,we identified ubiquitin carboxyl terminal hydrolase L3(UCHL3),a DUB enzyme in the UCH protease family,as a bona fide deubiquitylase of the AhR in ***3 was shown to interact with,deubiquitylate,and stabilize AhR in a manner dependent on its deubiquitylation ***,we showed that UCHL3 promotes the stem-like characteristics and potent tumorigenic capacity of NSCLC ***3 increased AhR stability and the binding of AhR to the promoter regions of the“stemness”genes ATP-binding cassette subfamily G member 2.ABCG2.,KLF4,and *** of UCHL3 markedly downregulated the“stemness”genes ABCG2.KLF4,and c-Myc,leading to the loss of selfrenewal and tumorigenesis in ***,the UCHL3 inhibitor TCID induced AhR degradation and exhibited significantly attenuated efficacy in NSCLC cells with stem cell-like ***,UCHL3 was shown to indicate poor prognosis in patients with lung *** general,our results reveal that the UCHL3 deubiquitylase is pivotal for AhR protein stability and a potential target for NSCLC-targeted therapy.

The median postoperative survival time of the 199 patients was 39 months and the 5-year survival rate was 26% which was decreased with increase of tumor-stage. In comparison of the survival time of patients in Stage Ⅰ and those in Stage Ⅲa there was a significant difference (P<0.01). There were no significant differences in survival rate of 3 and 5 years between the patients receiving chemotherapy prior to or after surgical resection. The improvement in survival was documented by surgical resection combined with chemotherapy or chemoradiotherapy for LSCLC. The effect of multimodality treatment is correlated with tumor P-TNM staging the involvement of lymph node especially that of the mediastinal lymph node is a negative factor influencing the prognosis. Surgical resection as an initial management followed by chemotherapy or chemoradiotherapy may be indicated in LSCLC patients of Stage Ⅰ Stage Ⅱ and some Stage Ⅲa as the cancer can be resected completely.

Immune checkpoints are the crucial regulators of immune system and play essential roles in maintaining self-tolerance,preventing autoimmune responses,and minimizing tissue damage by regulating the duration and intensity of the immune ***,immune checkpoints are usually overexpressed in cancer cells or noninvasive cells in tumor tissues and are capable of suppressing the antitumor *** on substantial physiological analyses as well as preclinical and clinical studies,checkpoint molecules have been evaluated as potential therapeutic targets for the treatment of multiple types of *** the last few years,extensive evidence has supported the immunoregulatory effects of traditional Chinese medicines(TCMs).The main advantage of TCMs and natural medicine is that they usually contain multiple active components,which can act on multiple targets at the same time,resulting in additive or synergistic *** strong immune regulation function of traditional Chinese medicine on immune checkpoints has also been of great *** example,Astragalus membranaceus polysaccharides can induce anti-PD-1 antibody responses in animals,and these antibodies can overcome the exhaustion of immune cells under tumor immune ***,many other TCM molecules could also be novel and effective drug candidates for the treatment of ***,it is essential to assess the application of immune checkpoints in the development of new drugs and *** this review,we focus on research progress in the field of immune checkpoints based on three topics:(1)immune checkpoint targets and pathways,(2.development of novel immune checkpoint-based drugs,and(3)application of immune checkpoints in the development of TCMs.
