A novel human gene, named E9730 (a clone number of fetal liver cDNA library), has been identified from more than 1.,000 expressed sequence tags (ESTs) based on our large scale sequencing of human fetal liver cDNA libraries. Although sequencing of this novel human gene indicates that it is a leucine zipper protein, the function of E9730 and its homologous genes among species is unknown yet. To find out physiological functional clue of E9730, the yeast two-hybrid system was used to screen the E9730-interac-ting protein(s), and one clone containing a cDNA insert with almost the entire coding sequence (amino acids 39-335) of human Jabl (Jun-activating domain binding protein 1. that interacted specifically with E9730 was identified. A specific association between Jabl and E9730 was shown by co-immunoprecipitation and co-localization experiments. Syrian hamster ernbryo(SHE) cells are irradiated by a particles emitted from Plutonium-238 in *** frequencies of donor DNA derived from passaged cells,including monocloned cells from 20th passage cells,after irradiation are parallel with their tumorigenicity of donor cells. After trans formation, expressions of H-ras and c-myc genes are in the state of activation, which might be resulted from occurrence of mutation in H-ras gene and hypomethylation of c-myc gene respectively, even though no alteration was observed on their restriction fragment length polymorphisms(RELP) patterns of both genes. On the other hand,expression of p53 gene is also found to be in the state of activation in transformant, but no heavy staining of p53 protein appeared in the transformant-derived tumor with p53 specific McAb HD 200. This might be result from the partlal deletion of p53 cDNA in transformant by reverse transcriptional polymerase chain reaction(RT PCR). These results imply that activation of H-ras and c-myc genes coupled with inactivation of p53 turnor suppressor gene play important role in a particle-induced cell trans formation.

O6-methylguanine-DNA methyltransferase (MGMT) gene expression in 6 Mer+ (HeLa S3, SMMC-7721.SGC-7901. B-239, AGZY83-a, and Cc80 1. and 2 Mer- (SHG-44 , and HeLa MR) human tumor cell lines was examined. Southern blot analysis revealed no deletion, amplification, or rearrangement of the MGMT gene in these cell lines. However ,～ 1. 0 kb transcripts were detected in the 6 Mer+ cell lines but not in the 2 Mer-cell lines by Northern blot analysis. Furthermore,a rough correlation between MGMT activity and mRNA level in these cell lines was observed. These results suggest that transcriptional regulation of the MGMT gene is the molecular basis of the absence of MGMT activity in Mer-cell lines.

In this study, we investigated the genes related to the transformation of immortalized human fetal tracheal fibroblast cell line induced by alpha particles by means of differential display mRNA method. The result revealed that there were 23 DNA fragments that were expressed intensively in alphaSHTF cells (SHTF cells forming clone on agar after irradiated by alpha particles emitted by 238Pu) only and not in SHTF (SV40-immortalized human fetal tracheal fibroblast) cells. Northern dot confirmed two fragments, C1.-5, C23-1.which showed intensive mRNA expression in alphaSHTF cells, but not in SHTF cells. The length of the C1.-5 fragment was 31.bp. Searching in BLAST database revealed that the C1.-5 fragment might be an unknown sequence.

The complete amino acid sequence of human augmenter of liver regeneration (hALR) was reported by deduction from nucleotide sequence of its complementary DNA . The cDNA for hALR was isolated by screening a human fetal liver cDNA library and the sequencing of this insert revealed an open reading frame encoding a protein with 1.5aa and highly homologous (87% ) with rat ALR encoding sequence. The recombinant hALR expressed from its cDNA in transient expression experiments in cos-7 cells could stimulate DNA synthesis of HTC hepatoma cell in the dose-dependent and heat-resistant way. Northern blot analysis with rat ALR cDNA as probe confirmed that ALR mRNA was expressed in the normal rat liver at low level and that dramatically increased in the regenerating liver after partial hepatectomied rat. This size of hALR mRNA is 1.4 kb long and expressed in human fetal liver, kidney and testis. These findings indicated that liver itself may be the resource of ALR and suggested that ALR seems to be an im-portant paracrined regulator of liver regeneration.

The primary Syrian hamster embryo(SHE) cells were used to study the oncogenic transformation by ￣(238)pu α particles or X-rays alone or in combination with a chemical promoter phorbol *** curves of SHE cells following exposure to α-particles or X-rays were fitted to single-or multi-target models,respectively. Model parameters were: Do = 0. 55 Gy. n = 1.for α particles 4 Do = 1.44 Gy. Dq = 3.0 Gy. n=7.7 for *** of α particles or X-rays induced cell transformation was dose-dependant.α particles were more efficient in inducing cell transformation than that of X-rays. The enhancement of SHE cell transformation by phorbol 1.-myristate 1.-acetate(PMA) following exposure to α particles of 0. 25-1. 00 Gy was observed.

Primary rat tracheal epithelium (RTE) cells can be transformed in vitro by a-particles irradiation. Oneout of 3 isolated morphologically trans formed colonies gave rise to an immortal cell line and ultimately be-came neoplastic by successive passaging. Cytogenetic analysis was performed on the cell line in order to un-derstand how the specific chromosome alterations participate in the process of neoplastic transformation ofRTE cells. The 5th, 1.th, 39th passage cells and the colony cells isolated from soft agar (SA) culturing ofthe 4oth passage cells were analyzed. The results showed that the 2Oth passage cells were nontumorigenic,but the 4Oth passage cells were tumorigenic. The increase of the length of the long arrn of chromosome 1.(1.q+ ) was frequent at the 5th passage (67. 8% ), especially in SA cells in which it occurred in a clonalfashion (1.0% ) Therefore, 1.q+ might be the critical gene lesion after a-particles irradiation. Mono-somy of chromosome 8 that occurred frequently at passage 5 (54% ) and 40 (53% ) might be correlatedwith earlier transformation of RTE cells. In addition, three marker chromosomes (Ml, M2 and M3)showed a passage-dependent increase’ In particular M1.and M2 that were highly frequent at passage 39(90% ) suggested that they might play an important role in the process of cell transformation.

Objective To search novel method for diagnosis and therapy of B-lymphoma, specific small mo-lecular peptide ligands against binding site of tumor cells were screened and its effects on signal transduction and cell apoptosis were tested. Methods Specific peptide ligands were screened by binding with site of human B lymphoma cell(OC1.Y8)using peptide-bead libraries. The identified peptides were characterized with responsible cellsby rebinding test. The role of tyrosine phosphorylation of peptide ligand was tested by Western blot; and its apoptosispromoting role was observed by confocal fluorescent microscope. Results Specific peptide ligand was able to bind specifically to site on cell surface and enter into cytoplasm. Tetrameric peptide ligand was able to strongly trigger signal transduction resulting in tyrosine phosphorylation and cellular apoptosis in OC1.Y8 cell line. Conclusion Screened peptide ligand can effectively bind with OC1.Y8 cell, stimulate cellular tyro-sine phosphorylation and induce cellular apoptosis.

Objective: A method for separating mRNAs by means of the polymerase chain reaction (differential display mRNA), and identifying the genes related to radiation-induced lung cancer was introduced. Methods: The RNAs were isolated from two pairs of samples, SV40-immortalized human fetal tracheal fibroblast cell (SHTF) versus αSHTF cell (transformed SHTF cell induced by α particles) and lung cancer tissue versus normal lung tissue obtained from one miner, and amplified by RTPCR. The differential expressed gene fragments were displayed by autoradiograph or silver nitrate stain. Results: The differential display mRNA method was established using both cell and tissue samples. The bands stained by silver nitrate were clearer than those on X-ray film. The rate of reamplification of differentially expressed gene fragments stained by silver nitrate is 80%, higher than that by autoradiograph, 50%. Conclusion: Differential display mRNA method was established successfully on both cell and tissue samples. The modified method for staining band increased the rate of reamplification and established the basis for confirming relative genes.
