This study was to evaluate the effect of supplementing the diet of broilers with Neolamarckia cadamba leaf extract(NCLE) on meat quality by evaluating antioxidant parameters and the expression of genes in the p38 mitogen-activated protein kinase/nuclear factor-erythroid 2-related factor 2/antioxidant responsive element(p38 MAPK/Nrf2/ARE) signaling pathway, coupled with LC-MS-based metabolomic analysis. A total of 480 one-day-old male broilers were randomly allocated to four treatment groups—a control(CON)group, which was fed a basal diet, and three NCLE treatment groups, which were fed the basal diet supplemented with 100, 200, or 400 mg/kg NCLE(N1, N2, and N3 groups, respectively) for 42 d. Compared with the CON group, meat quality was improved in the N2 and N3 groups, as evidenced by the higher p H45min(P < 0.05) and lower shear force(P < 0.05) in breast muscle(BM) and lower drip loss at48 h(P < 0.05) in leg muscle(LM). Moreover, BM antioxidant capacity was significantly enhanced in the N3group, characterized by an increase in the total antioxidant capacity(T-AOC), the concentrations of glutathione peroxidase(GSH-Px) and catalase(CAT), and the relative mRNA expression of p38 MAPK,extracellular-signal regulated kinase 1/2(ERK1/2), c-Jun N-terminal kinase(JNK), Nrf2, CAT, and GSH-Px(P < 0.05). Similarly, LM in the N3 group displayed higher T-AOC, increased GSH-Px and CAT concentrations,reduced malonaldehyde contents(P < 0.05), and upregulation of the relative mRNA levels of JNK, Nrf2,heme oxygenase, CAT, and superoxide dismutase(SOD)(P < 0.05). Metabolomics analysis revealed that Darabinono-1,4-lactone and lyso-PAF C-16-d4 were negatively correlated with shear force and cooking loss(P < 0.05) and displayed increased abundance in BM of the N3 group. Natural product elansolid A belongs to one type of polyketide macrolactone with promising antibiotic ***-viously,the first total synthesis of elansolid A in 28 longest linear sequence(LLS)and 41 steps in total has been ***,the simplified analog of elansolid A,featured with a cyclohexyl-fused 19-memebered macrolactone,was proposed,and its asymmetric total synthesis based on a convergent strategy and key reactions exemplified by desymmetric alcoholysis of anhydride,Pd-catalyzed Stille coupling,Suzuki-Miyaura coupling as well as Mukaiyama macrolactonization was finished.

Ten novel isocoumarins,including four pairs of enantiomers,were isolated from Artemisia dubia ***(Asteraceae).Compounds 1,2 and 3a/3b possessed a unique 6/6/6-tricyclic system comprising an unusual 1-(2-methylcyclohexyl)propan-1-one moiety fused with isocoumarin core *** 4a/4b were characterized as an unexpected 2,5-dimethylcyclohexan-1-one scaffold,and compounds 5a/5b and 6a/6b were rare 1,*** structures and absolute configurations were elucidated through spectroscopic data,X-ray crystallography,ECD and NMR calculations with DP4+*** biosynthetic pathways were proposed from the naturally occurring *** pharmacological analysis suggested that the targets of compound 1 were significantly enriched in the cell cycle and Pl3K-Akt signaling *** molecular docking revealed that compound 1 had high binding affinity with CDK2(total score:6.8717).Furthermore,compounds 1 and 2 exhibited inhibitory activity on three human hepatoma cell lines,with inhibitory ratios of 85.1% and 84.5%(HepG2),88.2% and 87.3%(Huh7),and 69.2% and 69.1%(SK-Hep-1)at 200μmol·L^(-1),respectively.

Nineteen new cadinane-involving sesquiterpenoid dimers,artemordins A-S(1-—19),together with 13 known SDs(20-32)were isolated from Artemisia *** structures and absolute configurations were established by comprehensive spectral analyses,X-ray single crystal diffraction,theoretical ECD,and NMR ***,artemordins A-F(1—6)were the first examples of two cadinane units constructed by unprecedented C-3−C-15′or C-3−C-13′single bond with an oxido-rearranged 6/5/6/6 fused ring system;artemordins G-K(7-11)were biogenetically connected by[4+2]cycloaddition reaction and artemordins G-J(7-10)possessed a novel 5/6/6/6/6/6/5-heptacyclic fused ring *** L-S(12-19)were formed by esterification,which involved three different types of *** assay suggested that the most active compounds,artemordins B and H(2 and 8),exhibited inhibitory activities on three hepatoma cell lines with IC50 values of 26.9 and 25.1μmol/L(HepG2),29.5 and 18.3μmol/L(Huh7),19.7 and 15.7μmol/L(SK-Hep-1).

CRISPR/Cas9,presently the most widely used genome editing technology,has provided great potential for functional studies and plant ***,the strict requirement for a protospacer adjacent motif(PAM)has hindered the application of the CRISPR/Cas9 system because the number of targetable genomic sites is ***,the engineered variants Cas9-NG,SpG,and SpRY,which recognize non-canonical PAMs,have been successfully tested in plants(mainly in rice,a monocot).In this study,we evaluated the targeted mutagenesis capabilities of these Cas9 variants in two important Brassica vegetables,Chinese cabbage(Brassica rapa ***)and cabbage(Brassica oleracea ***).Both Cas9-NG and SpG induced efficient mutagenesis at NGN PAMs,while SpG outperformed Cas9-NG at NGC and NGT *** achieved efficient editing at almost all PAMs(NRN>NYN),albeit with some self-targeting activity at transfer(T)-DNA *** SpRY-induced mutants were detected in cabbage plants in a PAM-less ***,an adenine base editor was developed using SpRY and TadA8e deaminase that induced A-to-G conversions within target sites using non-canonical ***,the toolboxes developed here induced successful genome editing in Chinese cabbage and *** work further expands the targeting scope of genome editing and paves the way for future basic research and genetic improvement in Brassica.

Five stems of rapeseed with abundant black microsclerotia were collected from Huangyuan County of Qinghai Province,China,and fungal isolates were obtained from the *** were identified based on morphology,molecular features and specific PCR *** results showed that the 10 fungal isolates belonged to Verticillium longisporum lineage A1/*** of the 10 isolates(HW7-1)was tested for virulence on three species of rapeseed,including *** Zhongshuang 9,*** Qingyou 9 and *** Tayou 2 by conidia inoculation of HW7-1 on roots of young *** seedlings were inoculated with *** conidia or water *** seedlings of these treatments were transplanted in culture mix and incubated in a growth chamber(20℃).Results suggested that the control seedlings of three cultivars appeared quite healthy,while the seedlings inoculated with HW7-1 turned yellowing leaves,seedling stunting or even death after 22 days *** was re-isolated from he yellow leaves,thus fulfilling Koch's ***,compared to the control treatments,inoculation with HW7-1 caused flowering delay and seed yield reduction on Tayou 2 with production of microsclerotia on the *** our knowledge,this is the first report of *** lineage A1/D3 on rapeseed in northwestern China.

his study sought to determine the effects of rosemary leaf powder(RP) on laying performance, egg quality, serum indices, gut barrier function, and cecal microbiota and metabolites of late-phase laying hens. A total of 84 “Jing Tint 6” laying hens at 65-week old were randomly divided into 2 groups and fed either a basal diet(CON) or a basal diet supplemented with 0.3% RP. Our study revealed that RP improved the Haugh unit and decreased yolk n-6/n-3 polyunsaturated fatty acid(PUFA) ratio of laying hens,increased serum superoxide dismutase(SOD), jejunal activities of SOD and catalase(CAT), and jejunal zonula occludens-1(ZO-1) expression, as well as decreased serum tumor necrosis factor-a(TNF-a) level and jejunal TNF-a m RNA expression. Rosemary leaf powder markedly enhanced(P < 0.05) cecal abundances of Rikenellaceae, Rikenellaceae＿RC9＿gut＿group, and Turicibacter, tended to promote(P = 0.076)butyrate concentration, and reduced(P < 0.05) cecal abundances of Erysipelatoclostridiaceae, Sutterellaceae, Fusobacteriaceae, Campylobacteraceae, Sutterella, Campylobacter, and Fusobacterium, which were closely linked with Haugh unit, yolk n-6/n-3 PUFA ratio, serum SOD and TNF-a. In addition, RP altered the metabolic functions of cecal microbiota and enhanced the abundances of butyratesynthesizing enzymes, including lysine 2,3-aminomutase, β-lysine 5,6-aminomutase, and 3-oxoacid Co A-transferase. Together, 0.3% RP has the potential to enhance egg quality by partially modulating serum antioxidant status, jejunal barrier function, and cecal microbiota structure and metabolites,indicating that RP could be considered a promising feed additive to promote the production performance of late-phase laying hens.

Stomatal regulation is critical for mangroves to survive in the hyper-saline intertidal zone where water stress is severe and water availability is highly ***,very little is known about the stomatal sensitivity to vapour pressure deficit(VPD)in mangroves,and its co-ordination with stomatal morphology and leaf hydraulic *** measured the stomatal response to a step increase in VPD in situ,stomatal anatomy,leaf hydraulic vulnerability and pressure-volume traits in nine true mangrove species of five families and collected the data of genome *** aimed to answer two questions:(1)Does stomatal morphology influence stomatal dynamics in response to a high VPD in mangroves?with a consideration of possible influence of genome size on stomatal morphology;and(2)do leaf hydraulic traits influence stomatal sensitivity to VPD in mangroves?We found that the stomata of mangrove plants were highly sensitive to a step rise in VPD and the stomatal responses were directly affected by stomatal anatomy and hydraulic ***,denser stomata was correlated with faster stomatal closure at high VPD across the species of Rhizophoraceae,and stomata size negatively and vein density positively correlated with genome *** negative leaf osmotic pressure at the full turgor(πo)was related to higher operating steady-state stomatal conductance(gs);and a higher leaf capacitance(Cleaf)and more embolism resistant leaf xylem were associated with slower stomatal responses to an increase in *** addition,stomatal responsiveness to VPD was indirectly affected by leaf morphological traits,which were affected by site salinity and consequently leaf water *** results demonstrate that mangroves display a unique relationship between genome size,stomatal size and vein packing,and that stomatal responsiveness to VPD is regulated by leaf hydraulic traits and stomatal *** work provides a quantitative framework to better understand of stomatal regulation in mangroves
