Dear Editor,Plant viruses spread from the initially infected cells to adjacent cells through plasmodesmata(PD) to complete their successful infections in host plants.This cell-to-cell movement requires specific viral protein(s),known as movement protein(s)(MP).Generally,virus MPs can bind to nucleic acids and modify PD size exclusion limit(Schoelz et al.,2011).Geminiviruses are DNA viruses that cause devastating losses to food crops worldwide,and encode 4–8 classical viral proteins(Gong et al.,2021;Hu et al.,2020.

The phytohormone abscisic acid(ABA)is a 15-carbon molecule that fulfills essential functions in all plants.It regulates many as-pects of plant life,including growth and development,seed dormancy and germination,leaf senescence,and responses to various biotic and abiotic stresses(Finkelstein 2013).Due to the latter functions,ABA is also known as the"stress phytohormone."As in the case of other hormones,the level of ABA in the cells is critical for its precise regulatory functioning.

In the fall of 2019,I left home in Indonesia to study in China,my ancestral home a century ago according to my parents.I went to Shaoxing,the capital of Chinese yellow wine.Since the moment I arrived,I have learned many stories about this renowned beverage.Shaoxing locals joke that yellow wine flows through their veins,and the liquor dates back to the ancient Kingdom of Yue in the first millennium BC.

Marker-assisted selection(MAS) is an important modern breeding technique,but it has been found that the effect of the markers for quantitative trait loci(QTL) is inconsistent,leading in some cases to MAS failure and raising doubts about its effectiveness.Here the model organism Drosophila melanogaster was employed to study whether an effective marker could be found and applied to MAS.We crossed the stock carrying the y0 marker(a recessive mutation allele of the yellow gene on the X chromosome) with three other stocks carrying corresponding wild-type markers in an F2 design,and found that the y0 marker was in significant association with low body weight(Pyellow gene was deficient in the y0 stock,leading to disappearance of melanin from the cuticle and probably influencing the developmental process.The above results confirmed the existence of effective QTL markers applicable to MAS breeding schemes,and their potential application in breeding new stocks.

The sex-determining genes are found to be variable among different fish species. yellow catfish(Pelteobagrus fulvidraco) is an important aquaculture fish species in China with XX/XY sex-determining type. Recently, YY super-male yellow catfish has been successfully produced by combining hormonalinduced sex reversal method with sex chromosome-linked markers. Here, we identified a novel PDZ domain-containing gene in yellow catfish designated as pfpdz1, in whose intron the sex-linked marker was located. The coding sequence of pfpdz1 in Y chromosome was identical to that in X chromosome except a missense SNP(A/T) that changes an amino acid(E8V) in the N-terminal region. Pfpdz1 displayed male-specific expression during sex differentiation. Overexpression of pfpdz1 using additive transgenesis induces XX ovary to differentiate into testis-like tissue, while the targeted inactivation of pfpdz1 in Y chromosome using CRISPR/Cas9-mediated mutagenesis triggers ovarian differentiation. Furthermore,we demonstrated that pfpdz1 initiates testicular differentiation through upregulating expression of amh, dmrt1 and sox9a1, as well as downregulating expression of cyp19a1, foxl2 and wnt4. Our data provide functional evidence that pfpdz1 is significant for male differentiation and maintenance in yellow catfish.

Pseudomonas plecoglossicida is a rod-shaped,gram-negative bacterium with flagella.It causes visceral white spot disease and high mortality in Larimichthys crocea during culture,resulting in serious economic loss.Analysis of transcriptome and quantitative real-time polymerase chain reaction(PCR)data showed that dks A gene expression was significantly up-regulated after 48 h of infection with Epinephelus coioides(log2 FC=3.12,P<0.001).RNAi of five sh RNAs significantly reduced the expression of dks A in P.plecoglossicida,and the optimal silencing efficiency was 96.23%.Compared with wild-type strains,the symptoms of visceral white spot disease in L.crocea infected with RNAi strains were reduced,with time of death delayed by 48 h and mortality reduced by 25%.The dks A silencing led to a substantial down-regulation in cellular component-,flagellum-,and ribosome assembly-related genes in P.plecoglossicida,and the significant up-regulation of fli C may be a way in which virulence is maintained in P.plecoglossicida.The GO and KEGG results showed that RNAi strain infection in L.crocea led to the down-regulation of inflammatory factor genes in immune-related pathways,which were associated with multiple immune response processes.Results also showed that dks A was a virulence gene in P.plecoglossicida.Compared with the wild-type strains,RNAi strain infection induced a weaker immune response in L.crocea.
