Competition between anthocyanin and kaempferol glycosides biosynthesis affects pollen tube growth and seed set of Malus

Flavonoids play important roles in regulating plant growth and *** this study,three kaempferol 3-O-glycosides were identi fi ed and mainly accumulated in fl owers but not in leaves or fruits of *** Malus,fl ower petal color is normally white,but some genotypes have red fl owers containing *** biosynthesis appears to be in competition with kaempferol 3-O-glycosides production and controlled by the biosynthetic *** white fl ower Malus genotypes had better-developed seeds than the red fl ower *** fl owers,the overexpression of MYB10 in Malus domestica enhanced the accumulation of anthocyanin,but decreased that of kaempferol *** pollination the transgenic plants showed slower pollen tube growth and fewer developed *** application ofdifferent fl avonoid compounds suggested that kaempferol 3-O-glycosides,especially kaempferol 3-O-rhamnoside,regulated pollen tube growth and seed set rather than cyanidin or quercetin *** was found that kaempferol 3-O-rhamnoside might regulate pollen tube growth through effects on auxin,the Rho of plants(ROP)GTPases,calcium and the phosphoinositides signaling *** the inhibition of auxin transport,the transcription levels of Heat Shock Proteins(HSPs)and ROP GTPases were downregulated while the levels were not changed or even enhanced when blocking calcium signaling,suggesting that HSPs and ROP GTPases were downstream of auxin signaling,but upstream of calcium *** summary,kaempferol glycoside concentrations in pistils correlated with auxin transport,the transcription of HSPs and ROP GTPases,and calcium signaling in pollen tubes,culminating in changes to pollen tube growth and seed set.

Novel diagnosis and therapy for hepatoma targeting HBV-related carcinogenesis through alternative splicing of FIR (PUF60)/FIRΔexon2

Aim: Disturbed alternative splicing of far upstream element-binding protein-interacting repressor (FIR) was found to be unable to repress c-Myc transcription and so it might be important for suppressing tumor development. FIR is a splicing variant of poly (U)-binding-splicing factor (PUF60), and forms complex with other splicing factors. FIR/PUF60 is a splicing factor of U2 small nuclear ribonucleoprotein auxiliary factor family, Thus FIR/PUF60 is a multifunctional protein. The expression of exon2-lacking splicing variant of FIR, FIRΔexon2, is elevated in many cancer tissues and promotes tumor development by disabling FIR-repression to sustain c-Myc activation. FIRΔexon2, as a dominant negative of FIR, opposed apoptosis in cancer cells. FIR/FIRΔexon2 interacts with degron pocket of F-box and W (Typ) D (Asp) repeat domain-containing 7 and inhibits proteolysis of substrates proteins. Recently, FIR/PUF60 was identified as a versatile regulator of transcriptional and post-transcriptional steps in expression of hepatitis B virus (HBV) pregenomic RNA (pgRNA) expression. Methods: Small molecular chemical compounds against FIR and FIRΔexon2 were screened among 2,3275 chemicals by natural product depository array (RIKEN, Wako, Saitama, Japan). Results: Nine chemicals against FIR and four chemicals against FIRΔexon2 were identified as candidates of interacting chemicals. Interestingly, BK697 contains WD -like structure. Among them, BK697 against FIRΔexon2 inhibited hepatoma cell growth. Conclusion: Therefore, FIR (PUF60)/FIRΔexon2 is multifunctional and applicable for clinical use for HBV suppression and hepatoma treatment. Together, one clue to the development of hepatome diagnosis and therapies directed against FIR/FIRΔexon2/PUF60 with small molecular weight chemicals that inhibit HBV cccDNA replication.

In fleshy fruits,organic acids are the main source of fruit acidity and play an important role in regulating osmotic pressure,pH homeostasis,stress resistance,and fruit *** transport of organic acids from the cytosol to the vacuole and their storage are complex processes.A large number of transporters carry organic acids from the cytosol to the vacuole with the assistance of various proton pumps and ***,much remains to be explored regarding the vacuolar transport mechanism of organic acids as well as the substances involved and their *** this review,recent advances in the vacuolar transport mechanism of organic acids in plants are summarized from the perspectives of transporters,channels,proton pumps,and upstream regulators to better understand the complex regulatory networks involved in fruit acid formation.

通过 Newton类型的伪时间子迭代格式实现第 n时间层至第 n+ 1时间层的精确推进 ,发展了一种具有二阶时间精度、全隐式 LU- SGS- τTS方法求解雷诺平均 Navier- Stokes方程组( RANS) ;同时还对比了显式四步 R- K- τTS和原始 LU- SGS方法的计算结果。采用改进的Jameson中心、Van Leer和 Roe格式对 N- S方程组的对流项进行空间离散 ;迎风格式通过MUSCL插值提高格式精度 ,并采用光滑且连续可微的通量限制器消除求解过程中的数值振荡。利用上述时间方法和空间格式进行组合 ,分别对二维翼型、三维机翼激波 /边界层干扰流动中的几个极具挑战性的状态进行模拟 ,通过结果相互间及与实验结果的对比。
