Introduction :Chronic liver disease (CLD) is a disease of public health importance. CLD is defined as a clinical syndrome of liver disease lasting for at least six months with histology showing varying degree of hepatocellular necro-inflammation and fibrosis with or without neoplastic transformation. The disease is a spectrum that manifests initially as chronic hepatitis which may progress to liver cirrhosis and ultimately hepatocellular carcinoma (HCC). The current practice in the field of Gastroenterology has shifted from invasive methods of diagnosing HCC to non-invasive methods using tumor biomarkers. Various biomarkers of HCC have been proposed, but the largest body of evidence exists with alpha-fetoprotein (AFP). Most of the studies on the combined diagnostic accuracy of AFP and des-gamma-carboxyprothrombin (DCP) were done in other populations outside Nigeria. It is necessary to determine the combined diagnostic accuracy of the two tumor markers for early detection of HCC in North-central Nigeria. The expression of miR-155 and miR-181 in gastric cancer and their effects on clinical parameters and prognostic value

Objective The aim of this study was to investigate the expression and clinical significance of miR-155 and miR-181 in gastric *** Sixty-eight patients with gastric cancer and 80 healthy volunteers were selected as *** samples of patients and volunteers were collected to detect miR-155 and miR-181 expression levels in serum and tumor *** miR-155 and miR-181 serum levels were significantly higher in patients with gastric cancer than those in the control group(P<0.05).miR-155 and miR-181 expression levels in gastric cancer tissues were 16.74±4.29 and 12.17±3.26,respectively and 3.42±0.39) and 3.06±0.69 in paracancerous tissues,respectively,and they were 1.22±0.21 and 1.08±0.35 in normal tissues,***-155 and miR-181 expression levels in cancer tissues were significantly higher than adjacent tissues,and they were lowest in normal tissues(P<0.05).High miR-155 and miR-181 expression had significant effects on differentiation,T stage,lymph node metastasis,and clinical stage(P<0.05).Cox multivariate analysis showed that onset,T stage,clinical stage,differentiation degree,lymph node metastasis,miR-155,and miR-181 were independent risk factors for prognosis of patients with gastric *** Serum miR-155 and miR-181 have a diagnostic value in gastric cancer,and they are strong signals of poor prognosis.

Objective:To study the effect of exfoliative cytology combined with CEA,CA125,CA15-3 and CA19-9 examinations on the diagnosis of malignant serous ***:236 cases of patients who were diagnosed as serous effusion during inpatient in our hospital from January of 2015 to January of 2017 were selected as research *** to biopsy and pathological examinations,the diagnostic results were cleared that there were 136 cases of patients with benign serous effusion(benign group)and 100 cases of patients with malignant serous effusion(malignant group).Two groups of patients were both given exfoliative cytology combined with CEA,CA125,CA15-3 and CA19-9 examinations for tumor markers to analyze the effect of exfoliative cytology combined with CEA,CA125,CA15-3 and CA19-9 examinations on the diagnosis of malignant serous ***:The levels of tumor markers CEA,CA125,CA15-3 and CA19-9 in the benign group were all lower than those in the malignant group,and the difference was of statistical significance(p<0.05).In addition,exfoliative cytology combined with CEA,CA125,CA15-3 and CA19-9 examinations showed a higher sensitivity,specificity and accuracy than any of the above examinations alone,and the difference was of statistical significance(p<0.05).Conclusions:Exfoliative cytology combined with CEA,CA125,CA15-3 and CA19-9 examinations can effectively improve the diagnostic accuracy of malignant serous effusion and is worthy of being spread clinically.

Objective: The aim of this study was to explore the correlation of pretreatment serum tissue polypeptide specific antigen (TPS) with prognosis in primary breast cancer. Methods: A total of 361 patients with grades I-III breast cancer had been followed up from January 2001 to February 2011. Serumal TPS level was measured by enzyme-linked immunosorbent assays (ELISA). Univariate and multivariate analyses were used to investigate associations between pretreatment TPS level and clinicopathological parameters and patient outcomes. Results: First, at the univariate analysis, the expression of TPS was related with some clinicopathological traditional prognostic factors such as tumor size (P = 0.030), histologic grade (P = 0.001) and lymph node status (P = 0.008). Second, overall survival were significantly shorter among patients with elevated pretreatment serum TPS (P = 0.038). However, finally, multivariate Cox regression indicated that the level of pretreatment serum TPS was not an independent prognostic parameter for overall survival in primarily breast cancer patients (P > 0.05). Conclusion: The expression of pretreatment serum TPS is closely correlated with clinicopathology parameters and overall survival of patients with primarily breast cancer, but its level has no independent prognostic value.

Objective: To initially explore the application of proteome technologies in study of serum, to establish two-dimen-sional gel electrophoresis (2-DE) profiles of human gastric cancer serum and paired normal serum, and to screen and identify differentially expressed proteins in poorly-differentiated gastric cancer serum and paired normal serum, in which try to find out significant biomarker candidates for gastric cancer. Methods: 2-DE was adopted to separate the total proteins of poorly-differentiated gastric cancer serum and paired normal serum. After staining and analyzing by ImageMaster 2D Elite software, the differentially expressed proteins were identified by matrix-assisted laser desorption/ionization-time of flight- mass spectrometry (MALDI-TOF-MS). Results: 2-DE serum profiles with high resolution were obtained. Five protein spots were found as differentially-expressed proteins and identified as Serpin B6 (Placental thrombin inhibitor) Cytoplasmic antiproteinase (CAP) (Protease inhibitor6) (PI-6), Septin-1 (LARP) (Serologically defined breast cancer antigen NY-BR-24), Kallikrein-6 precursor (Protease M) (Neurosin) (Zyme) (SP59), Hemoglobin beta chain, Hemoglobin beta chain and Beta-defensin 108 precursor (Beta-defensin 8) (DEFB-8). Conclusion: The differential proteins were demonstrated to present in poorly-differentiated gastric cancer serum and paired normal serum. The molecular biomarkers associated with poorly differentiated gastric cancer could be possibly identified by the high throughput screening proteome technology.

Background: The dual marker algorithm Risk of Ovarian Malignancy Algorithm(ROMA) has been widely used in the clinic for identification of equivocal pelvic masses in ovarian carcinoma. To obtain higher diagnostic efficiency, we created a new diagnostic index Risk of Ovarian Malignancy Index(ROMI) by combing thymidine kinase 1(TK1), HE4 and ***: 335 patients with pelvic masses on imaging and 46 healthy controls were enrolled. Dissociation enhanced lanthanide fluoroimmunoassay（DELFIA）, as one of the most sensitive luminescent bioassay techniques, has been widely adopted in a variety of research and medical institutions. However, commercial DELFIA using molecular probes like lanthanide（Ln3+）-chelates as biolabels often suffers from a series of disadvantages, such as a low labelling ratio of Ln3+ per biomolecule（up to 10-30）, low photostability and high cost of Ln3+-chelates. To overcome these concerns, herein we develop a unique and facile bioassay method based on the dissolution-enhanced luminescence of inorganic lanthanide nanoparticles（NPs） [1]. Due to the highly concentrated Ln3+ ions in a single Ln3+-NP, an extremely high Ln3+ labeling ratio can be achieved, which enormously amplifies the luminescent signal and thus improves the detection sensitivity as compared to that of DELFIA（Fig. 1）. Utilizing sub-10 nm NaEuF4 NPs as dissolution-enhanced luminescent nanoprobes, we demonstrate the successful in vitro detection of carcinoembryonic antigen（CEA, an important tumor marker） in human serum samples with a detection limit down to 0.1 pg/mL（0.5 f M）, which is the lowest among those ever reported（about 3 orders of magnitude improvement relative to that of DELFIA）. The proposed dissolution-enhanced luminescent bioassay, as an alternative to commercial DELFIA by simply replacing the molecular probes with inorganic lanthanide nanoprobes in its protocol, shows great promise in versatile biomedical applications such as ultrasensitive and multiplexed in vitro detection of disease markers in clinical diagnosis [2, 3].

The predictive value of tumor markers has been previously reported a lot. However, the studies focused on advanced gastric cancer are few. In this paper, we focused on investigating the connection between clinical characteristics and the tumor markers of advanced gastric cancer, therapeutic effect of chemotherapy, and prognosis. A retrospective study including 146 advanced gastric cancer who had not received any previous anticancer therapy was performed. Blood samples for CEA, AFP, CA125, CA72-4, CA242 and CA19-9 were taken from patients before chemotherapy and monthly during their treatments. The tumor markers’ clinical value can be predicted through statistical analysis. An ROC value of 0.621 was yielded by CEA in predicting lymph node metastasis in gastric cancer, which was significantly higher than any of other markers. CA125 had the highest accuracy, specificity and sensitivity with peritoneal metastasis in gastric cancer patients. CEA and AFP were more frequently positive with hepatic metastases in gastric cancer patients. The response rate ofDecreasing, Stable and Increasing group based on CEA, CA19-9, CA125, CA72-4 and CA242 levels achieved the significance in statistic. Positive cases of CA125, CA242, CA19-9 and CA72-4 showed poor prognosis, and CA125 and CA242 indicated significant differences in 3 yearsurvival rates. These results indicated that different tumor markers in gastric cancer indicated different metastasis sites. CEA, CA242, CA19-9, CA125 and CA72-4 are predictive biomarkers in evaluating the effectiveness of chemotherapy. The elevated CA125, CA242, CA72-4 and CA19-9 levels at diagnosed had association with shorter overall survivals, especially CA125 and CA242.
