目的:探讨初诊急性髓系白血病(AML)患者外周血中调节性T细胞(Regulatory T Cells, tregs)和白介素-35(IL-35)的表达。方法:收集33例初诊AML患者和40例健康体检者的外周血,采用流式细胞术检测受试者外周血中固有免疫和适应性免疫细胞、tregs的水平,酶联免疫吸附试验(ELISA)方法检测血浆IL-35水平,RT-PCR法检测P35 mRNA和EBI3 mRNA相对表达量,并分析tregs与IL-35的相关性。结果:初诊AML患者外周血中CD4^(+)T细胞、CD8^(+)T细胞、B细胞以及NK细胞比例较对照组显著降低(Ptregs、IL-35水平以及P35 mRNA、EBI3 mRNA表达量较对照组显著升高(Ptregs与IL-35表达水平呈正相关(r=0.668,Ptregs和IL-35可能参与了AML患者抑制性肿瘤免疫微环境的形成,可能成为AML治疗的新靶点。

目的探讨人类免疫缺陷病毒(HIV)感染长期不进展者(LTNPs)接受抗逆转录病毒治疗(ART)人群与HIV进展者接受ART人群的调节性T细胞(tregs)细胞表型与功能上的差异。方法 2011年于新疆伊宁市现场招募HIV/AIDS患者资料库中发现的LTNPs+ART中至2019年12月1日仍存活者,共计38例为病例组。1:1匹配,以HIV进展者+ART为对照组。取LTNPs+ART及HIV进展者+ART的外周血,用流式细胞术(FCM)检测CD4/CD25/CD127/foxp3/CD69/CD39各亚组的Treg细胞比例,使用微量样本多指标流式蛋白定量技术(CBA)、酶联免疫吸附试验(ELISA)试剂盒检测评估Treg细胞因子和趋化因子水平。结果本次研究纳入LTNPs+ART和HIV进展者+ART各38例。LTNPs+ART组Treg细胞亚组表达频率高于HIV进展者+ART组;LTNPs+ART组tregs细胞因子[转化生长因子–β(TGF-β)、白细胞介素–10(IL-10)、白细胞介素–35(IL-35)、白细胞介素–6(IL-6)和干扰素–γ(INF-γ)],和趋化因子C–C–基元配体–17(CCL17)和C–C–基元配体–21(CCL21)水平高于HIV进展者+ART组。结论接受治疗的LTNPs拥有较高比例的tregs,且相对比接受治疗的HIV进展者tregs表现出较好的免疫抑制作用。

目的观察NOD小鼠人源化后,CD4^+和CD8^+调节性T细胞(regulatory T cells,tregs)频率和功能的变化,揭示tregs在人源化NOD小鼠1型糖尿病中的作用及免疫学机制可能的变化。方法流式细胞术分别分析12周龄未发病的人源化NOD小鼠和NOD小鼠脾淋巴细胞和胰腺淋巴结细胞中CD8^+CD122^+T、CD8^+CD28-T、CD8^+CD25^+Foxp3^+T和CD4^+CD25^+Foxp3^+T细胞的频率,并采用3H-Td R掺入法检测脾CD4^+CD25^+T和CD8^+CD25^+T细胞的免疫抑制功能。结果人源化NOD小鼠和NOD小鼠的脾淋巴细胞和胰腺淋巴结细胞中CD4^+CD25^+Foxp3^+T细胞频率无显著性差异(P>0.05),而人源化NOD小鼠脾淋巴细胞和胰腺淋巴结细胞中CD8^+CD122^+T、CD8^+CD28-T、CD8^+CD25^+Foxp3^+T细胞等CD8^+tregs亚群的频率较NOD小鼠都显著降低,但NOD小鼠人源化后,CD4^+CD25^+T和CD8^+CD25^+T细胞的免疫抑制功能并没有显著性差异;同时与人源化NOD小鼠相比,NOD小鼠的胰腺淋巴结细胞中CD8^+T细胞频率更低。结论人源化NOD小鼠脾脏和胰腺淋巴结中CD8^+tregs亚群频率的降低,引起其胰腺淋巴结中CD8^+T细胞频率的升高,导致胰岛β细胞破坏更严重,可能是引起人源化NOD小鼠自发1型糖尿病较NOD小鼠明显提前且加重的因素之一。
