Several nitrate and ammonia transporter genes in root of wheat has been cloned in previous studies,but little is known about the seasonal expression patterns of these transporter genes up to *** present study,seasonal expression of four nitrate transporter genes (NRT2.1,NRT2.2,NRT2.3) and two ammonia transporter genes(AMT1.1 and AMT1.2 ) were studied in roots of two wheat cuhivars with higher(XY107) and lower(XY6) abilities in N *** were grown in about 2.5 L pots in greenhouse from Dec.25,2012 to May.10 *** nitrogen(N) treatments,low N(nil N application) and high N(0.3 g·mg) treatments were established,and there were four repeats for each *** roots were sampled at tillering(F3),jointing(F7),heading(F10) and filling(F11) stages, and the expression of the transporter genes were quantified using real-time PCR method. The results indicated that the two wheat cultivars significantly differd in their abilities in N uptake in either low N(LN) or high N treatment(HN).The cultivar XY107 could continuously uptake N during the whole growth stage,but XY6 almost stoped uptaking N since F7 or F10 *** expression of all the transporter genes showed significantly seasonal dynamics,but different between the two wheat cultivars and N *** XY107,all the six transporter genes had increased expression level from F3 to F7 or from FIO to F11 stage in both N treatments;however,in XY6,most of the transporter genes had a increase expression from F3 to F7 stage,but a stable or decrease expression since F7 *** LN,the expression of the transporter genes in XY107 was lower than that in XY6 before F10 stage,but higher at F11 stage;however,in HN,the genes expression in XY107 was generally higher than that in XY6 during the whole growth *** conclude that the expression of N transporter genes showed significant seasonal dynamics in wheat roots,and wheat cultivar with higher N uptake ability could keep higher N transporter genes expression in roots

Current study systematically investigated the interaction of two alkaloids, anisodine and monocrotaline, with organic cation transporter OCT1, 2, 3, MATE1 and MATE2-K by using in vitro stably transfected HEK293 cells. Both anisodine and monocrotaline inhibited the OCTs and MATE transporters. The lowest IC50 was 12.9 μmol·L-1 of anisodine on OCT1 and the highest was 1.8 mmol·L-1 of monocrotaline on OCT2. Anisodine was a substrate of OCT2(Km = 13.3 ± 2.6 μmol·L-1 and Vmax = 286.8 ± 53.6 pmol/mg protein/min). Monocrotaline was determined to be a substrate of both OCT1(Km = 109.1 ± 17.8 μmol·L^-1, Vmax = 576.5 ± 87.5 pmol/mg protein/min) and OCT2(Km = 64.7 ± 14.8 μmol·L^-1, Vmax = 180.7 ± 22.0 pmol/mg protein/min), other than OCT3 and MATE transporters. The results indicated that OCT2 may be important for renal elimination of anisodine and OCT1 was responsible for monocrotaline uptake into liver. However neither MATE1 nor MATE2-K could facilitate transcellular transport of anisodine and monocrotaline. Accumulation of these drugs in the organs with high OCT1 expression(liver) and OCT2 expression(kidney) may be expected.

The present study aimed to evaluate the effects of manganese methionine hydroxyl analog chelated(Mn-MHAC)as a manganese(Mn)source on growth performance and trace element deposition in broilers.A total of 432 Arbor Acres commercial female broilers were fed a basal corn-soybean diet containing Mn at 25.64 mg/kg diet for 10 *** were then randomly assigned to 6 groups,including a control group(the basal diet),a Mn sulfate group(the basal diet supplemented with Mn at 100 mg/kg diet),and 4 Mn-MHAC groups(the basal diet supplemented with Mn-MHAC at 25,50,75 and 100 mg Mn/kg diet,respectively).The results showed that compared with the control group,groups supplemented with Mn-MHAC had a positive effect on BW(quadratic,P=0.017)and ADG(quadratic,P=0.017).Moreover,the Mn-MHAC(50 mg Mn/kg diet)group had significantly greater BW and ADG(Ptransporter 1(DMT1)(P=0.024),ferro-portin 1(FPN1)(P=0.049),and Cu transporter-1(CTR1)(P<0.001)in the duodenum,as well as CTR1 in the jejunum and ileum(P=0.040 and 0.011,respectively)were higher in the Mn-supplemented group than in the control ***,the relative mRNA level of DMT1 in the jejunum and ileum of broilers in the Mn-MHAC group(50 mg Mn/kg diet)did not differ from those in the control group,but was lower than those in the Mn sulfate group(P<0.05).In conclusion,Mn-MHAC dietary supplementation improved the growth performance and trace element deposition in *** this study,we recommend the opti-mum Mn-MHAC level to meet the Mn requirement of broile

Background: Milk protein is crucial for milk quality in sows and health of newborn piglets. Plasma amino acids(AA)in sows are important precursors for milk protein synthesis in the mammary gland. In order to study the regulation of AA transported in sow mammary glands and possible underlying mechanisms, we measured the expression of genes coding for milk proteins, AA transporter expressions, and plasma AA concentrations in sows at three different physiological stages(D-17, D1 and D17 of lactation), and then further investigated the regulation of AA transport across the cell membrane by adaptive mechanisms using pig mammary epithelial cells(PMEC) as an in vitro *** were cultured in DMEM:F12 with 4 amino acid concentrations(0 × AA complex, 1 × AA complex, 5 × AA complex,and 25 × AA complex). Classes of AA complexes evaluated in this study included neutral AAs(L-Ala + L-Ser + L-Cys), acidic AAs(L-Asp, L-Glu) and neutral + basic AAs(L-Ala + L-Ser + L-Cys + L-Lys).Results: Our results indicated that m RNA expression of genes coding for milk protein(αs1-casein, αs2-casein,β-casein and κ-casein) increased significantly with the advance of physiological stage(P transporters including excitatory AA transporter 3(EAAT3), alanine/serine/cysteine/threonine transporter(ASCT1) and sodium-coupled neutral AA transporter 1(SNAT2) were greater in lactating sow mammary glands compared with sow mammary glands in late pregnancy(P < 0.05). The m RNA expressions of SLC38 A2, SLC1 A1,SLC6 A14 increased significantly in the cell mediums supplemented with 5 × and 25 × of AA complexes compared with those cells cultured in DMEM/F12 cell medium(P < 0.05). Cancer cells reprogram their gene expression to promote growth,survival,proliferation,and *** unique expression of certain uptake transporters in cancers and their innate function to concentrate small molecular substrates in cells make them ideal targets for selective delivering imaging and therapeutic agents into cancer *** this review,we focus on several solute carrier(SLC)transporters known to be involved in transporting clinically used radiopharmaceutical agents into cancer cells,including the sodium/iodine symporter(NIS),norepinephrine transporter(NET),glucose transporter 1(GLUT1),and monocarboxylate transporters(MCTs).The molecular and functional characteristics of these transporters are reviewed with special emphasis on their specific expressions in cancers and interaction with imaging or theranostic agents[e.g.,Ⅰ-123,Ⅰ-131,123Ⅰ-iobenguane(mIBG),18F-fluorodeoxyglucose(18F-FDG)and 13C pyruvate].Current clinical applications and research areas of these transporters in cancer diagnosis and treatment are ***,we offer our views on emerging opportunities and challenges in targeting transporters for cancer imaging and *** analyzing the few clinically successful examples,we hope much interest can be garnered in cancer research towards uptake transporters and their potential applications in cancer diagnosis and treatment.

transporters comprise the largest family of membrane proteins in human organism, including members of solute carrier transporter and ATP-binding cassette transporter families. They play pivotal roles in the absorption, distribution and excretion of xenobiotic and endogenous molecules. transporters are widely expressed in various human tissues and are routinely evaluated during the process of drug development and approval. Over the past decade, increasing evidence shows that drug transporters are important in both normal physiology and disease. Currently, transporters are utilized as therapeutic targets to treat numerous diseases such as diabetes, major depression, hypertension and constipation. Despite the steady growth of the field of transporter biology, more than half of the members in transporter superfamily have little information available about their endogenous substrate（s） or physiological functions. This review outlines current research methods in transporter studies, and summarizes the drug-transporter interactions including drug-drug and drug-endogenous substrate interactions. In the end, we also discuss the therapeutic perspective of transporters based on their physiological and pathophysiological roles.
