A nano-delivery system expands the insecticidal target of thiamethoxam to include a devastating pest,the fall armyworm

Nano-delivery systems have been applied to deliver various synthetic/botanical pesticides to increase the efficiency of pesticide use and reduce the volumes of pesticides applied.Previous studies have supported the hypothesis that the nanocarriers can help expand the insecticidal target of pesticides to include non-target pests.However,the potential mechanism underlying this interesting phenomenon remains unclear.Herein,a widely applied star polycation(SPc)nanocarrier was synthesized to construct a thiamethoxam(TMX)nano-delivery system.The SPc-based delivery system could promote the translocation of exogenous substances across the membrane of Sf9 cells,increase the cytotoxicity of TMX against Sf9 cells by nearly 20%,and expand the insecticidal target of TMX to include Spodoptera frugiperda(the fall armyworm),with a 27.5%mortality increase at a concentration of 0.25 mg/mL.Moreover,the RNA-seq analysis demonstrated that the SPc could upregulate various transport-related genes,such as Rab,SORT1,CYTH,and PIKfive,for the enhanced cellular uptake of TMX.Furthermore,enhanced cell death in larvae treated with the TMX-SPc complex was observed through changes in the expression levels of death-related genes,such as Casp7,BIRC5,MSK1,and PGAM5.The SPc-based nano-delivery system improved the cellular uptake of TMX and expanded its insecticidal target by adjusting the expression levels of death-related genes.The current study mainly identified the transport and cell death genes related to nanocarrier-based insecticidal target expansion,which is beneficial for understanding the bioactivity enhancement of the nano-delivery system.

The actions of neonicotinoid insecticides on nicotinic acetylcholine subunits Ldα1 and Ldα8 of Leptinotarsa decemlineata and assembled receptors

The insect nicotinic acetylcholine receptor(nAChR)is a pentameric channel protein and also a target for neonicotinoids.There are few reported studies on the molecular interactions of Leptinotarsa decemlineata nAChRs with neonicotinoids.In this study,we analyzed the response of acetylcholine and neonicotinoids(thiamethoxam[TMX],imidacloprid[IMI],and clothianidin[CLO])on hybrid receptors constructed by nAChRα1 andα8 subunits of L.decemlineata(Ldα1 and Ldα8)co-expressed with ratβ2 subunit(rβ2)at different capped RNA(cRNA)ratios in Xenopus oocytes.In addition,we evaluated the expression changes of Ldα1 and Ldα8 after median lethal dose of TMX treatment for 72 h by quantitative polymerase chain reaction(qPCR).The resulting functional nAChRs Ldα1/rβ2 and Ldα1/Ldα8/rβ2 showed different pharmacological characteristics.The neonicotinoids tested showed lower agonist affinity on Ldα1/Ldα8/rβ2 compared to Ldα1/rβ2 at same ratios of subunit cRNAs.The sensitivities of neonicotinoids tested for Ldα1/rβ2 and Ldα1/Ldα8/rβ2 at cRNA ratios of 5:1,1:1 and 5:5:1,1:1:1,respectively,were lower than those for nAChRs at ratios of 1:5 and 1:1:5,respectively,whereas the values of maximum response(Imax)varied.For Ldα1/Ldα8/rβ2,a reduction of Lda8 cRNA resulted in increased sensitivity to IMI and decreased sensitivity to TMX.The expression of Ldα1 and Ldα8 significantly decreased in adults by 82.12%and 47.02%,respectively,while Ldα8 was significantly upregulated by 2.44 times in 4th instar larvae after exposure to TMX.We infer that Ldα1 and Ldα8 together play an important role in the sensitivity of L.decemlineata to neonicotinoids.

Insecticide resistance in Bemisia tabaci from Cyprus

A comprehensive study on the Bemisia tabaci （biotype B） resistance to neonicotinoid insecticides imidacloprid, acetamiprid and thiamethoxam, and pyrethroid bifenthrin was conducted in Cyprus. The resistance level to eight field-collected B. tabaci populations was investigated. The activities of enzymes involved in metabolic detoxification and the frequencies ofpyrethroid and organophosphates target site resistance mutations were determined. Moderate to high levels of resistance were detected for imidacloprid （re- sistance factor [RF] 77-392） and thiamethoxam （RF 50-164） while low resistance levels were observed for acetamiprid （RF 7-12）. Uniform responses by the Cypriot whiteflies could be observed against all neonicotinoid insecticides. No cross-resistance between the neonicotinoids was detected as well as no association with the activity of the P450 microsomal oxidases. Only imidacloprid resistance correlated with carboxylesterase activity. Low to extremely high resistance was observed for insecticide bifenthrin （RF 49 1 243） which was associated with the frequency of the resistant allele in the sodium channel gene but not with the activity of the detoxification enzymes. Finally, the F331W mutation in the acetylcholinesterase enzyme acel gene was fixed in all B. tabaci populations from Cyprus.

Quantification of thiamethoxam in Rhizomes and Leaves of the Hedychium coronarium and Water and Soil by High-Pressure Liquid Chromatography

The linearity, stability, accuracy and inter-day precisions of the assay method were evaluated in water, soil and rhizomes and leaves of Hedychium coronarium of the Zingiberaceae family. Mato Grosso do Sul is a predominantly agricultural state in Brazil and has many rivers and permanent and seasonal lakes, in which the Hedychium coronarium, an aquatic plant, is found the swampy environments. The prepared samples were analyzed quantitatively by High Performance Liquid Chromatography with PDA and UV detection for the presence of thiamethoxam. The thiamethoxam was recovered from these samples at rates ranging from 81.16% - 99.93%. The coefficient of variation in the quantitative analysis of the thiamethoxam was under 5%. The linearity of the method was determined by linear regression. The analysis of the samples spiked with known amounts of analyte demonstrated that the response was proportional to the concentrations of the samples with determination coefficients of r2 = 0.9992 (water and soil) and r2 = 0.9990 (leaves and rhizomes) for the linear range of the analytical calibration curves of the samples. The detection limit was 0.36 μg.L-1 and quantification limit was 1.2 μg.L-1 for thiamethoxam. The method was considered sensitive for quantification of the thiamethoxam in water, soil and rhizomes and leaves of Hedychium coronarium.

Characterisation of Neonicotinoid and Pymetrozine Resistance in Strains of Bemisia tabaci(Hemiptera:Aleyrodidae)from China

Four strains of the Q biotype and one of the B biotype of the whitefly Bemisia tabaci collected from China were characterised for resistance to four neonicotinoid insecticides and pymetrozine.Q biotype strains showed moderate to strong resistance to imidacloprid,thiamethoxam and acetamiprid,but little or no cross-resistance to dinotefuron.Resistance to neonicotinoids was consistently associated with resistance to pymetrozine,despite the latter having a distinct（though unresolved）mode of action.The single B biotype strain proved largely susceptible to all the insecticides investigated.Resistance in the Q biotype strains was associated with over-expression of a cytochrome P450 monooxygenase gene,CYP6CM1,whose substrate specificity presumably accounts for the observed cross-resistance profiles.
