Inhalation of taraxasterol loaded mixed micelles for the treatment of idiopathic pulmonary fibrosis

Idiopathic pulmonary fibrosis(IPF)is a chronic and fatal lung disease characterized by pulmonary inflam-mation,oxidative stress,and excessive extracellular matrix(ECM)*** anti-fibrotic drugs for IPF treatment in the clinic lack selectivity and demonstrate unsatisfactory efficacy,highlighting the urgent necessity for a novel therapeutic ***(TA),which has biological activities against lung injury induced by various factors,is a potential anti-IPF drug due to its anti-inflammatory,antiox-idant and lung-protective ***,the protective effect of TA on IPF has not been confirmed,and its clinical application is limited due to its poor aqueous *** this study,we demonstrated that TA could inhibit epithelial-mesenchymal transition(EMT)and migration of A549 cells by inhibiting the transforming growth factor-β1(TGF-β1)/Smad signaling *** improve the aqueous solubility and pulmonary administration performance of TA,we prepared TA loaded methoxy poly(ethylene glycol)-poly(d,l-lactide)(mPEG-PLA)/d-α-tocopheryl polyethylene glycol succinate(TPGS)mixed polymeric mi-celles(TA-PM).Then a MicroSprayer^(R) Aerosolizer was used to deliver TA-PM once every two days for three weeks to evaluate their therapeutic effects on bleomycin(BLM)-induced IPF *** results demonstrated that inhaled TA-PM significantly inhibited BLM-induced inflammation,oxidative stress and fibrosis in lung ***,TA-PM exhibited high pulmonary deposition and retention by pul-monary administration,along with a favorable safety ***,this study emphasizes the potential of inhaled TA-PM as a promising treatment for IPF,providing a new opportunity for their clinical appli-cation.

taraxasterol(TS)is a naturally occurring pentacyclic triterpenoid extracted from the traditional Chinese herb Taraxacum *** studies have highlighted its significant roles in exhibiting anti-inflammatory,anti-oxidant,and liver protective *** the present study,the anti-cancer potential of TS against cervical cancer was investigated,employing network pharmacology techniques,molecular docking,and in vitro experimental *** exhibits its anticancer properties by modulating multiple targets,pathways,and biological *** vitro experiments demonstrated the potent inhibitory effects of TS on cancer cell growth and migration,while no significant impact on apoptosis was *** primary objective was to elucidate the anti-cancer potential of TS,which is a crucial lead compound in the treatment of cervical *** findings may serve as a basis for the development of novel anticancer therapeutics and medicine-based interventions for cervical cancer.

Anti-inflammatory effects of taraxasterol on animal models

Purpose: Traditional Chinese medicine Taraxacum officinale has been widely used to treat inflammatory diseases. taraxasterol is one of main active components isolated from Taraxacum officinale. Recently, we have demonstrated that taraxasterol has the in vitro anti-inflammatory effects. This study aims to determine the anti-inflammatory effects of taraxasterol against animal models. Methods: Anti-inflammatory effects were assessed in four animal models by using dimethylbenzene-induced mouse ear edema, carrageenan-induced rat paw edema, acetic acid-induced mouse vascular permeability and cotton pellet-induced rat granuloma tests. Results: Our results demonstrated that taraxasterol dose-dependently attenuated dimethylbenzene-induced mouse ear edema and carrageenan-induced rat paw edema, decreased acetic acid-induced mouse vascular permeability and inhibited cotton pellet-induced rat granuloma ***: Our finding indicates that taraxasterol has obvious anti-inflammatory activity on in vivo animal models. It will provide experimental evidences for the traditional use of Taraxacum officinale and taraxasterol in inflammatory diseases.

Purpose: taraxasterol was isolated from the traditional Chinese medicinal herb Taraxacum which has been frequently used as a remedy for inflammatory diseases. In the present study, we determined the in vivo anti-arthritic effect of taraxasterol on arthritis induced by Freund’s complete adjuvant(FCA) in ***: Rats were immunized with FCA by intradermal injection into the right hind metatarsal footpad, and were orally treated daily with taraxasterol at 2, 4 and 8 mg/kg from day 2 to 28 after immunization. Paw swelling, arthritis index, body weight, spleen index and thymus index were evaluated. The levels of TNF-α, IL-1β, PGE2, OPG and RANKL in sera were measured using ELISA. Histopathological changes in joint tissues were examined using hematoxylin and eosin(H&E).Results: taraxasterol significantly suppressed paw swelling and arthritis index, attenuated body weight loss, decreased the spleen index and thymus index induced by FCA. Furthermore, taraxasterol significantly inhibited the overproduction of serum TNF-α, IL-1β, PGE2 and RANKL, and increased serum OPG production in FCA-induced rats. Histopathological examination indicated that taraxasterol attenuated synovial hyperplasia, bone and cartilage damage, and inflammatory cell ***: These results suggest that taraxasterol has the potential protective effect against FCA-induced arthritis in rats.
