Background:Accumulation of alpha-synuclein(α-syn)is a main pathological hallmark of Parkinson’s and related diseases,which are collectively known as *** evidence has supported that the same protein can induce remarkably distinct pathological progresses and disease phenotypes,suggesting the existence of strain difference amongα-syn *** studies have shown thatα-syn pathology can propagate from the peripheral nervous system(PNS)to the central nervous system(CNS)in a“prion-like”***,the difference of the propagation potency from the periphery to CNS among differentα-syn strains remains unknown and the effect of different generation processes of these strains on the potency of seeding and propagation remains to be revealed in more ***:Three strains of preformedα-syn fibrils(PFFs)were generated in different buffer conditions which varied in pH and ionic ***α-syn PFFs were intramuscularly(IM)injected into a novel bacterial artificial chromosome(BAC)transgenic mouse line that expresses wild-type humanα-syn,and the efficiency of seeding and propagation of these PFFs from the PNS to the CNS was ***:The three strains ofα-syn PFFs triggered distinct propagation *** fibrils generated in mildly acidic buffer led to the most severeα-syn pathology,degeneration of motor neurons and microgliosis in the spinal ***:The differentα-syn conformers generated in different conditions exhibited strain-specific pathology and propagation patterns from the periphery to the CNS,which further supports the view thatα-syn strains may be responsible for the heterogeneity of pathological features and disease progresses among synucleinopathies.

Single later blastula nuclei from AB strain of zebrafish (Danio rerio) were transplanted into enucleated unfertilized eggs of Long fin strain. Of 1119 cloning embryos, 14 reconstructed embryos developed into fry. DNA fingerprinting systems of the cloned fish were similar to those of the nuclear donor fish, but were distinctly different from those of the nuclear recipient fish. It confirmed that the genetic material originated from nuclear donor cell other than from nuclear recipient egg. The research suggested that the basic technique for nuclear transplantation performed with different strains of zebrafish has made a breakthrough. It should be helpful for the study of some important developmental problems such as gene function, the regulation of gene expression during animal development, the developmental potential of a nucleus and the interactions between the donor nucleus and the recipient cytoplasm, etc.

Objectives:Accumulation of nitrite and biogenic amines(BAs)in fermented meat products is a matter of public health *** study aimed to screen nitrite-degrading and BA-degrading strains from sour porridges and sausages and bacon products in *** and Methods:After screening out 12 strains,the degradation of nitrite,the degradation of BAs,the activities of nitrite-reducing enzymes,and the detection of genes involved in the BAs were assessed by spectrophotometry method with hydrochloric acid naphthalene ethylenediamine,high-performance liquid chromatography,GENMED kit,and polymerase chain reaction,***:Pediococcus pentosaceus labelled M SZ12 and M GC 2,Lactobacillus plantarum labelled M SZ22,and Staphylococcus xylosus labelled Y CC 3 were *** activity of nitrite-reducing enzyme in M SZ22 was 2.663 units/*** degradation rate of total BAs of M SZ22 was 93.24%.The degradation rates of nitrite and BAs of M SZ12 were 86.49%and 37.87%,*** activity of nitrite-reducing enzyme in M SZ12 was up to 1.962 units/mg.M GC 2 showed higher degradation rates of nitrite(89.19%)and Y CC 3 showed higher degradation rates of BAs(36.16%).The genes encoding the multicopper oxidases(suf I/D2EK17)were detected in the four strains,which also did not contain BAs(histidine decarboxylase(hdc),tyrosine decarboxylase(tdc),ornithine decarboxylase(odc),lysine decarboxylase(ldc))formation encoding ***:These four strains(M SZ12,M GC 2,M SZ22,and Y CC 3)are promising candidates to use as starter cultures for nitrite and BAs in fermented sausages.

Objective:To find importance of morphometrie criterion of larval rostellar hook of Echinococcus granulosus(***) and the easy and reliable method for distinguish sheep and camel strains in epidemiologic ***:Larval rostellar hooks(n=1860) of 31 camel and sheep isolates in Iran,which aheady had been characterized by *** carefully processed by computerized imagime analysis system(CIAS) and acquired data about rostellar books were analyzed using software ***:Measurement analysis of rostellar hooks[mean Iength (24.23±3.12lμm]indicated that length of the large hook was a remarkable parameter for strain *** analysis demonstrated that CIAS could be used as a reliable tool to distinguish camel from sheep strains with high sensitivity(95.2%) and specificity(91.5%). Conclusions:CIAS as a specific,sensitive,economic,fast,and reliable means might be used for differentiation of *** *** perimeter and area were measured by digital technology,they were not shown as discriminative criterion as total hook length did.

Objective:To produce comparative data on a group of Trichomonas vaginalis clonal strains with varied drug responses using identical methods and ***:Five clonal strains of Trichomonas vaginalis were isolated from reference strain using agar plate *** variability of growth kinetic and susceptibility of clonal strain to metronidazole,tinidazole, satranidazole and nitazoxanide were observed in 96 well microlilre ***:Among these clonal strains there was a good correlation between rates of growth with the relative susceptibility of the strains to drugs in *** metronidazole,tinidazole and satranidazole susceptibility,different degrees of susceptibility were ***,no difference in nitazoxanide susceptibility was found between the clonal strain tested and a reference strain. Conclusions:This is the first description of biological variability in clonal stock of Trichomonas *** degrees of drug susceptibility were determined among clonal strains tested. Further studies will be necessary to ascertain the importance of this variability in clinical infection.

Characteristics of gut microbiota in representative mice strains:Implications for biological research

Background:Experimental animals are used to study physiological phenomena,pathological mechanisms,and disease *** gut microbiome is known as a potential confounding factor for inconsistent data from preclinical *** many gut microbiome studies have been conducted in recent decades,few have focused on gut microbiota fluctuation among representative mouse ***:A range of frequently used mouse strains were selected from 34 isolation packages representing disease-related animal(DRA),immunity defect animal(IDA),or gene-editing animal(GEA)from the BALB/c and C57BL/6J backgrounds together with normal mice,and their microbial genomic DNA were isolated from mouse feces to sequence for the exploration of gut ***:Mouse background strain,classification,introduced source,introduced year,and reproduction type significantly affected the gut microbiota structure(p<0.001 for all parameters),with background strain contributing the greatest influence(R^(2)=0.237).In normal groups,distinct gut microbiota types existed in different mouse ***-four core operational taxonomic units were obtained from normal mice,and 12 belonged to ***,the gut microbiota in C57BL/6J was more stable than that in BALB/c ***,the gut microbiota in the IDA,GEA,and DRA groups significantly differed from that in normal groups(p<0.001 for all).Compared with the normal group,there was a significantly higher Chao 1 and Shannon index(p<0.001 for all)in the IDA,GEA,and DRA *** changed classes occurred with Firmicutes and *** abundances of Helicobacter,Blautia,Enterobacter,Bacillus,Clostridioides,Paenibacillus,and Clostridiales all significantly decreased in the IDA,GEA,and DRA groups,whereas those of Saccharimonas,Rikenella,and Odoribacter all significantly increased.

Probiotic Viability, Qualitative Characteristics, and Sensory Acceptability of Vegetable Juice Mixture Fermented with Lactobacillusstrains

The aim of the study was to consider the suitability of a mixture of juices from jicama, winter melon, and carrot as a raw medium for producing probiotic juice by Lactobacillus strains (Lactobacillus plantarum CICC22696 and Lactobacillus acidophilus CICC20710), as well as evaluate changes of physicochemical and microbiological characteristics during fermentation and cold storage (4°C, 28 days). Both strains grew well in juice mixtures after 24 h of fermentation at 37°C, reaching nearly 9 and 8 log CFU/mL when inoculated with L. plantarum and L. acidophilus respectively. The viability of L. plantarum was near 8 log CFU/mL at the end of storage whereas viability of L. acidophilus only remained 4.57 log CFU/mL. Degradation of total carotenoids was in the range of 12% - 23% in fermentation periods and 16% - 23% during cold storage depending on the strain used. The values of lightness, redness, and yellowness increased during fermentation. However, this tendency was variable during cold storage when the values of redness and yellowness decreased. Sensory acceptability of the products was enhanced by adding sucrose or multi fruit juice (containing mainly tropical fruit juices). The fermented juice with L. plantarum is efficient to produce a functional probiotic beverage.

Rice yellow mottle virus (RYMV) is a major biotic constraint for rice production in Africa. The resistance-breaking ability of Tanzanian RYMV strains and phylotypes (S4lm (Tz526), S4lv (Tz516), S4ug (Tz508), S5 (Tz429, Tz445), S6c (Tz486) and S6w (Tz539)) were tested by inoculating rice cultivars with RYMV1 resistant alleles (Gigante (rymv1-2), Tog12387 (rymv1-3), Tog5681 (rymv1-3), Tog5438 (rymv1-4), Tog5672 (rymv1-4+rymv2) and Tog5674 (rymv 1-5)) in a screen house. The results revealed multiple resistance-breaking strains and phylotypes on resistant cultivars Gigante, Tog12387, Tog5438 and Tog5681. However, the resistance breakdown was highly variable depending on the strain used, and disease severity ranged from 11% - 75.3%. The virulence potential of RYMV phylotype S4lm (Tz526) was similar to phylotype S6w (Tz539). The impact of strains and phylotypes on yield and its components in rice cultivars revealed highly significant differences (P ≤ 0.001). The lowest percent plant height reduction (2.8%), number of tillers per plant (2.5%), 1000 grain weight (2.7%), spikelet sterility (3.5%) and yield (5%) was recorded in rice cultivar Gigante inoculated with RYMV phylotype S6c (Tz486). Phylotype S6c (Tz486) despite being less virulent compared to other strains, its virus titer in rice cultivar Gigante (1.833) was higher than S5 (Tz429, Tz445) inoculated on Tog5674 (0.171, 0.207) and S6w (Tz539) inoculated on Tog5681 (0.283). The resistant-breaking strain S5 (Tz445) multiplied in resistant rice cultivar Tog5674 without inducing visible symptoms but showed positive reaction to ELISA with low virus titer. The strain S5 overcame wide range of resistant alleles including rymv1-2, rymv1-3, rymv1-4 and rymv1-5 resistance, with exception of rymv1-4 + rymv2. Chromolaena odorata L (Asteraceae) is a bad invasive plant, found in the humid tropics and sub-tropics worldwide. It is used against dysentery, diarrhea, malaria, wound healing, headache and toothache in traditional medicine. In the present study, we investigated the antibacterial activities of different leaves extracts of Chromolaena odorata L. (cyclohexane, dichloromethane, ethyl acetate and butanol) against four clinical diarrheal strains (Klebsiella oxytoca, Salmonella enterica, Shigella sonnei and Vibrio cholera). We demonstrated that C. odorata leaves extracts show an antibacterial activity between 0.156 and 1.25 mg/mL. Bioassay-guided chromatography by bioautography with iodonitrotetrazolium-based colorimetric assay allowed the isolation and identification of two active compounds. After the combination of RP-HPLC, mass spectrometry analysis, 1D and 2D-NMR spectroscopy, we isolated and characterized two active molecules corresponding to 3’,4’,5,6,7-Pentamethoxyflavone (Sinensetin) and4’,5,6,7-Tetramethoxyflavone (Scutellareintetramethyl ether).
