High-SNR staining Algorithm for One-Way Wave Equation-Based Modelling and Imaging

staining algorithms based on two-way wave equation migration methods have been applied to improve the signal-to-noise ratio(SNR)of poorly illuminated structures such as those in subsalt zones.In regular staining algorithms,when a source wavefield reaches the stained area that is associated with the target structures,a new wavefield called stained wavefield is excited,and this stained wavefield forward extrapolates synchronously with the real source wavefield.The forward-extrapolated stained and real source wavefields are cross-correlated with the backward-extrapolated receiver wavefield,and we obtain the stained and the real reverse time migration(RTM)images.The staining algorithms for RTM can suppress the noise of non-target regions and obtain high SNR images of the target structures.Whereas RTM methods are limited by the low computational efficiency and SNR,by contrast,one-way wave equation migration(OWEM)methods have the advantages of high efficiency and no interference from multiples.Thus,we developed a new staining method based on the generalised screen propagator(GSP)as a case of OWEM methods for subsalt imaging.Furthermore,a new stained wavefield called stained receiver wavefield is proposed here,forming two new staining strategies for seismic imaging,in which forward-propagated source and backward-propagated receiver wavefields can be conveniently selected to be stained at the stained area.Numerical experiments demonstrated that this staining GSP method is more effective in improving the SNR of subsalt structures compared to conventional GSP migration and RTM methods;moreover,these new staining strategies as applied to the OWEM methods can greatly improve the SNR of weakly illuminated structures in subsalt zones,in comparison with regular staining algorithms for one-way methods.

Comparison ofβ-Amyloid Plaque Labeling Methods:Antibody staining,Gallyas Silver staining,and Thioflavin-S staining

Objective To evaluate senile plaque formation and compare the sensitivity of three differentβ-amyloid(Aβ)labeling methods(antibody staining,Gallyas silver staining,and thioflavin-S staining)to detect Aβdeposition.Methods APPswe/PSEN1dE9 transgenic mice(APP/PS1)of different ages were used to examine spatiotemporal changes in Aβplaque deposition.Antibody staining,Gallyas silver staining,and thioflavin-S staining were used to detect Aβplaque deposition in the same brain region of adjacent slices from model mice,and the results were compared.Results With aging,Aβplaques first appeared in the cortex and then the deposition increased throughout the whole brain.Significantly greater plaque deposition was detected by 6E10 antibody than that analyzed with Gallyas silver staining or thioflavin-S staining(Pstaining and ones with thioflavin-S staining(P=0.0033).Conclusions The APP/PS1 mouse model of Alzheimer’s disease could mimick the progress of Aβplaques occurred in patients with Alzheimer’s disease.Antibody detection of Aβdeposition may be more sensitive than chemical staining methods.

Application of the Reactive Oxygen-Based Cervical Exfoliated Cell staining Solution in Clinical Cervical Cancer Screening

We developed a special methylene blue solution for staining of cervix shedding cells based on catalytic oxidizing chromogenic reaction, which shows a potential for cervical cancer cytology screening. We screened a total of 1 922 women for cervical cancer with the special methylene blue staining method and a conventional Pap smear method using cervix shedding cells. Then, the patients with positive indicators of the Pap smear or this special solution staining method were examined by the electron colposcopy and histopathological examination. staining of cervical exfoliated cells with this reactive oxygen-based special solution showed that the number of positive cases was 140(7.28%). Among them, 21 cases showed dark green(1.09%), and 119 cases showed purple black(6.19%). The results of the Pap smear method showed that the number of positive cases was 123(6.40%), of which ASCUS was 105(5.46%), ASC-H was 5(0.26%), and LSIL was 9(0.47%), and HSIL was 4(0.21%). For cervical exfoliated cell special staining solution for screening cervical intraepithelial neoplasia(CIN-Ⅱ, CIN-Ⅲ) and cervical cancer, sensitivity was 83.33%, specificity was 65.52%, accuracy was 74.29%, missed diagnosis rate was 13.33%, positive coincidence rate was 51.43%, and the negative coincidence rate is 86.67%. Our results proved the value of this method for early screening of cervical cancer through clinical practice in China.

Reduction and Block staining of Human Hair Shafts and Insect Cuticles by Ammonium Thioglycolate to Enhance Transmission Electron Microscopic Observations

We have developed a method where, after glutaraldehyde fixation, human hair shafts and insect cuticles are treated with ammonium thioglycolate (ATG) to improve ultrastructural staining. Conventional transmission electron microscopic (TEM) preparations do not distinguish the A-layer and the exocuticles of hair shafts. However, after ATG treatment, the A-layer appears in higher contrast. ATG treatment has also been used to observe the fibrillar structure in the cortex. In the cuticle of beetles, the epicuticle is stained by ATG. Although the human hair shaft (keratin) and insect cuticle (chitin) are composed of different materials, both can be reduced by the ATG solution. The ammonium in the ATG solution reacts with hair shafts and insect cuticles, causing a reduction of swelled cuticles. Therefore, ATG not only stains, but also reduces human hair shafts and the cuticles of beetles.

Comparative Investigation of Alternative Negative staining Reagents in Bacterial Morphological Study

Negative staining is an effective method that can be used for electron microscopic study to observe fine structural morphology without destruction of bacterial structure. Although uranium acetate is used worldwide as a general dyeing solution, it is extremely difficult to use it by a new purchase at a research institution because it falls under the nuclear regulation substance in Japan. Therefore, we examined alternative reagents for negative staining that could replace uranium acetate through bacterial observation with an electron microscope. Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Streptococcus pyogenes were examined by four stain reagents (phosphotungstic acid (PTA), EMstainer, TI blue, and uranium acetate). Pre cultured bacteria were stained with each stain reagents on a copper grid, washed with PBS, and observed with a transmission electron microscope. In the comparison between bacterial structures, the cell wall structure and bacterial flagella could be observed well in the order of PTA, EMstainer, and uranium acetate. With TI blue staining, flagella could be observed very poorly. In comparison between bacteria, gram negative bacteria such as Escherichia coli and Pseudomonas aeruginosa, could be observed well as compared with gram positive cocci such as Staphylococcus aureus and Streptococcus pyogenes. The uranium acetate looked very coarse in background particles. Since crystals tend to precipitate, TI blue also required filtering, and electron beams were absorbed by the agglomerated crystals, and the frequency of electronic burning occurred high frequency. In this study, there was clear difference in the observation conditions depending on the type of bacteria and the kind of the staining reagents. Especially, it was confirmed that good negative staining features of Pseudomonas aeruginosa by electron microscope were obtained by PTA and EMstainer staining. These alternative reagents are considered to be a candidate for a negative staining.

Introducing a Rapid and Safe Method for Myeloperoxidase staining

Background: Myeloperoxidase staining is used to differentiate leukemias since several decades. Despite implementation of flow cytometric, cytogenetic and molecular techniques for identification of leukemic blasts, histochemical stains such as myeloperoxidase stain are persistently used for better classification of leukemias. The myeloperoxidase staining is a time consuming and hazardous procedure. The present report describes a sensitive, rapid and easy method for assessment of peroxidase activity. Materials and Methods: Bone marrow aspiration slides were stained with Dako product: Code number: K3467 containing DAB chromogen (3,3-diaminobenzidine in chromogen solution) and substrate buffer (Imidasole-HCL buffer, PH 7.5 containing hydrogen peroxide and an anti microbial agent) in a rapid procedure taking only ten minutes time. The staining needs no material preparation steps. Neutrophils in the slide are taken as positive control or another normal smear was costained to be used as control. All cases were followed up with flow cytometry and cytogenetic studies. Result: The reaction product of this stain is brown and granular. Promyelocytes and myelocytes are the most strongly staining cells with positive (primary) granules. Lymphoblasts are negative. The result of classification of leukemias with this technique was in concordance with flow cytometric immunophenotyping. Discussion: Many practical techniques have been described using benzidine as an indicator for myeloperoxidase staining. Benzidine is a carcinogenic material and its usage is severely restricted in laboratory. Formerly we prepared requisite materials for myeloperoxidase staining by hazardous ways (boiling), but we decided to apply ready to use 3,3-diaminobenzidine (DAB), which is used in final step of immunohistochemistry stains. Conclusion: Use of 3,3-diaminobenzidine (DAB) is highly recommended for myeloperoxidase staining, while the result is extraordinary and fully compatible with flow cytometry and the method is safe and rapid.

Effect of exogenous spastin combined with polyethylene glycol on sciatic nerve injury

Polyethylene glycol can connect the distal and proximal ends of an injured nerve at the cellular level through axonal fusion to avoid Wallerian degeneration of the injured distal nerve and promote peripheral nerve regeneration.However,this method can only prevent Wallerian degeneration in 10% of axons because the cytoskeleton is not repaired in a timely fashion.Reconstruction of the cytoskeletal trunk and microtubule network has been suggested to be the key for improving the efficiency of axonal fusion.As a microtubule-severing protein,spastin has been used to enhance cytoskeletal reconstruction.Therefore,we hypothesized that spastin combined with polyethylene glycol can more effectively promote peripheral nerve regeneration.A total of 120 male Sprague-Dawley rats were randomly divided into sham,suture,polyethylene glycol,and polyethylene glycol + spastin groups.In suture group rats,only traditional nerve anastomosis of the end-to-end suture was performed after transection of the sciatic nerve.In polyethylene glycol and polyethylene glycol + spastin groups,50 μL of polyethylene glycol or 25 μL of polyethylene glycol + 25 μL of spastin,respectively,were injected immediately under the epineurium of the distal suture.Sensory fiber regeneration distance,which was used to assess early nerve regeneration at 1 week after surgery,was shortest in the suture group,followed by polyethylene glycol group and greatest in the polyethylene glycol + spastin group.Behavioral assessment of motor function recovery in rats showed that limb function was restored in polyethylene glycol and polyethylene glycol + spastin groups at 8 weeks after surgery.At 1,2,4 and 8 weeks after surgery,sciatic functional index values and percentages of gastrocnemius muscle wet weight were highest in the sham group,followed by polyethylene glycol + spastin and polyethylene glycol groups,and lowest in the suture group.Masson staining was utilized to assess the morphology of muscle tissue.Morphological changes in skeletal muscle were detectable in suture,p

BACKGROUND Hepatocellular carcinoma(HCC) is one of the most common malignant tumors worldwide, and novel methods for early/rapid diagnosis of HCC are needed.Terahertz(THz) spectroscopy is considered to have the potential to distinguish between normal liver tissue and HCC tissue; however, there are few reports on it.We conduct this observational study to explore the feasibility of THz imaging for the diagnosis of HCC.AIM To evaluate the feasibility of THz for discriminating between HCC and normal liver tissues using fresh tissue specimens obtained from HCC patients who had undergone surgery.METHODS Normal liver tissue and HCC tissue were cryosectioned into 50 μm-thick slicesand placed on cover glass. Two adjacent tissue sections were separated subjected to histopathological examination by hematoxylin and eosin staining or THz transmission examination, and THz images were compared with pathologically mapped images. We determined the typical tumor and normal liver tissue regions by pathological examination; the corresponding areas of adjacent sections were examined by THz transmission.RESULTS The transmission rate of HCC tissue was 0.15-0.25, and the transmission rate of typical HCC tissue was about 0.2. THz transmittance in normal liver tissue is slightly higher than 0.4, but there were many influencing factors, including the degree of liver cirrhosis, fat components, ice crystals in frozen sections, and apoptosis.CONCLUSION In conclusion, this study shows that THz imaging can detect HCC tissue. Further research will yield more detailed data of the THz transmission rates of HCC tissue with different degrees of differentiation.

Helicobacter pylori may be an initiating factor in newly diagnosed ulcerative colitis patients: A pilot study

AIM To directly visualize Helicobacter pylori(H. pylori) by the highly sensitive and specific technique of immunohistochemical staining in colonic tissue from patients newly diagnosed with ulcerative colitis(UC).METHODS Colonoscopic biopsies from thirty patients with newly diagnosed UC and thirty controls were stained with Giemsa stain and immunohistochemical stain for detection of H. pylori in the colonic tissue. Results were confirmed by testing H. pylori Ag in the stool then infected patients were randomized to receive either anti H. pylori treatment or placebo.RESULTS Twelve/30(40%) of the UC patients were positive for H. pylori by Giemsa, and 17/30(56.6%) by immunohistochemistry stain. Among the control group 4/30(13.3%) and 6/30(20 %) were positive for H. pylori by Giemsa and immunohistochemistry staining respectively. H. pylori was significantly higher in UC than in controls(P = 0.04 and 0.007). All Giemsa positive patients and controls were positive by immunohistochemical stain. Four cases of the control group positive for H. pylori also showed microscopic features consistent with early UC.CONCLUSION H. pylori can be detected in colonic mucosa of patients with UC and patients with histological superficial ulcerations and mild infiltration consistent with early UC. There seems to be an association between UC and presence of H. pylori in the colonic tissue. Whether this is a causal relationship or not remains to be discovered.
