Macrophages(Mφ) are involved in spiral artery remodeling(SAR) in normal pregnancy.However,the underlying mechanism of Mcp involved in preeclampsia(PE) remains unknown.Here,we found that trophoblasts-derived galectin-9(gal-9) was highly expressed in placenta and plasma of PE patients as well as in the PE mouse models.RmGal-9 administered caused development of PE features,whereas gal-9 blockade was protective.We found that rhGal-9 treatment enhanced the adhesion of decidual Mcp to extravillous trophoblast(EVT) cells via activating FAK/Src pathway.Moreover,gal-9 induced the expansion of CD11 c Mcp and enhanced their activated phenotype via interacting with CD44.IL-1(3 and TNF-α produced by CD11 c Mcp after gal-9 stimulation remarkably increased the apoptosis of EVT cells,which is adverse to SAR.In addition,gal-9/CD44 interaction promoted the ubiquitination of RIPK1 and induced autophagy via the AMPK-mTOR pathway.The degradation of ubiquitinated PKR1 mediated by the autophagy manner,led to the decreased angiogenic factor IL-8 production by Mcp.Accordingly,the tube formation and SAR were impaired due to the less IL-8.Therefore,our results revealed a new mechanism by which Mφ mediated abnormal SAR,and gal-9/CD44 is suggested to be a potential therapeutic target for PE.

Adrenomedullin and endocrine control of immune cells during pregnancy

The immunology of pregnancy is complex and incompletely understood. Aberrant immune activity in the decidua and in the placenta is believed to play a role in diseases of pregnancy, such as infertility, miscarriage, fetal growth restriction and preeclampsia. Here, we briefly review the endocrine control of uterine natural killer cell populations and their functions by the peptide hormone adrenomedullin. Studies in genetic animal models have revealed the critical importance of adrenomedullin dosage at the maternal-fetal interface, with cells from both the maternal and fetal compartments contributing to essential aspects underlying appropriate uterine receptivity, implantation and vascular remodeling of spiral arteries. These basic insights into the crosstalk between the endocrine and immune systems within the maternal- fetal interface may ultimately translate to a better understanding of the functions and consequences of dysregulated adrenomedullin levels in clinically complicated pregnancies.
