We have demonstrated for the first time that (ⅰ) mouse Sertoli cells predominantly secrete tPA under the action of FSH and cAMP-generating agents, whereas Leydig cells mainly produce uPA; (ⅱ) Sertoli cells are also capable of secreting PAI-1, as well as FSH, growth factors and GnRH increase PAI-1 gene expression; (ⅲ) the increases in tPA and PAI-1 activities by different hormones in the conditioned media of Sertoli cells correspond to the increases in the levels of tPA and PAI-1 mRNA in the cultured cells, suggesting that the synthesis of the activator-inhibitor in mouse Sertoli cells is regulated at a transcriptional level and the tPA secreted by Sertoli cells may be involved in the spermatogenesis.

The cellular localization and specific expression of TR3 mRNA in rat testis were investigated by the method of immunohistochemistry and in situ hybridization. It was demonstrated that orphan receptor TR3 was expressed in a significant amount in rat testis and TR3 protein and mRNA were specifically localized in germ cells, suggesting that TR3 may have an important function in regulating rat germ cell development.

在本实验中,光镜和电镜观察发现,在一例正常大鼠的局部睾丸的间质组织中,存在着不同发育时期的生殖细胞,同时还见支持细胞。上述各类细胞的超微结构同精细管内的无明显差别;但在位置上,它们却是分散于间质组织内,或者同间质细胞紧密相靠排列,或者分布于血管周围。本事实表明,在大鼠睾丸的间质组织中,可能也存在着生殖细胞赖以生存、发育和分化的环境条件。本文对这一现象的意义作了讨论。

The cloning and expression characterization of the centrosome protein genes family (centrin genes) in rat testis

Centrins are members of the centrosome protein family, which is highly conserved during revolution. The homologous genes of centrin in many organisms had been cloned, but the sequences of the rat centrin genes were not reported yet in GenBank. We cloned the cDNA fragments of centrin-1, -2 and -3 from the rat testis by RT-PCR, and analyzed the homology of the deduced amino acid sequences. The expression characterization of centrin genes in rat spermatogenesis.was carried out by semi-quantitative RT-PCR. The results show that the homology of the corresponding centrin proteins in human, mouse and rat is high. The expression of centrin-1 is testis-specific, spermatogenic cell-specific and developmental stage-related. Centrin-1 begins to be transcribed when the meiosis occurs, and its mRNA level reaches the peak in round spermatids. Centrin-2 and centrin-3 are highly expressed in spermatogonia and their mRNA level decreases markedly when meiosis occurs. These results suggest that centrin-1 may play roles in meiosis and spermiogenesis, and centrin-2 and centrin-3 may be related to mitosis.
