A method of rapidly detecting three kinds of microcystins( MCs) in environmental water samples by using disk SPE- ultra high performance liquid chromatography- tandem mass spectrometry( UPLC- MS / MS) was established. Firstly,environmental water samples were extracted by disk SPE column( C_(18)),and three kinds of MCs were separated by Waters BEH C_(18) chromatographic column with acetonitrile- 0. 2% formic acid solution as the mobile phase. After the gradient elution separation,the external standard method was used for quantitative and qualitative analysis under MRM of UPLC- MS / MS. The results showed that the three kinds of MCs in the range of 0. 05- 10 μg / L showed good linear relation,and the correlation coefficients were higher than 0. 999 4,while the method detection limit was 0. 04 ng / L. Under 0. 1,1,and 5 μg / L standard addition for the same environmental sample,the average recovery was 82. 8%- 108. 8%,and the relative standard deviation of determination results was2. 1%- 10. 1%( n = 6). This method is rapid,sensitive and accurate,so it can be effectively applied in the monitoring of MCs in environmental water samples.

A method based on UV-Vis spectrometry（UV） and vertex component analysis（VCA） was proposed to characterize the processing batch for preparation of prepared radix scutellariae using water steamingIn the proposed method, UV spectra of the radix scutellariae that were collected at different intervals of the processing procedure were measured, then the measured UV spectra of mixtures were resolved by VCA, and the estimated vertex components（VCs） and their variation of relative concentration were used to characterize the processing batchThe results show that estimated VCs have chemical significance with spectral characteristics of source signal when the source spectra are overlappedThe variation of the relative VCs’ concentrations can explain the physical and chemical changes of active components during the processing batchThe endpoint of the processing batch was determined as 35 min, which is more accurate and overcomes the shortcomings that different endpoints may be made by sensory analysis of different operatorsthe endpoint of the process batch that was determined by inspection of the variation of VCsUV-VCA method provides an alternative approach for characterization of the traditional Chinese medicines processing procedure.

Stereoselective separation and determination of quizalofopethyl in tobacco by ultra-performance convergence chromatography with tandem mass spectrometry

As one of the most commonly used medicinal plants,ginseng has been an attractive model herb for study.A wide range of analytical methods has been used to characterize its ***,less effort has been devoted to the rare ginseng saponins,especially their isomers and sugar *** this study,we used segmental monitoring and diagnostic ion filtering to characterize ginseng saponins by rapid liquid chromatography with time-of-flight mass spectrometry(LC-TOF-MS).By using selected diagnostic ions,specific groups of ginseng saponins were readily extracted from the complicated matrix.20(R) and 20(S) stereo-saponins were differentiated using the peak abundance ratio of[M-H20+H]+ to[M-2H20+H]+.The fragmentation behavior of ginsenosides was first reported in negative ion mode by MS/MS with high-energy collision-induced dissociation, producing rules to determine sugar numbers,positions and *** the rules,we identified and compared the nontarget ginseng saponin profiling of raw and steamed American ginseng roots and *** characterized 70 saponins in the *** strategy can be extended to screen and characterize other rare ginseng saponins and their metabolites

The purpose of this study was to develop and validate a novel and sensitive method for simultaneous enantiomeric analysis of quizalofop-ethyl in tobacco using ultra-performance converge nee chromatography with tandem mass spectrometry(UPC-MS/MS).Sample preparation prior to UPC^2-MS/MS analysis followed a QuEChERS(quick,easy,cheap,effective,rugged and safe)*** UPC^2-MS/MS method used an Acquity UPC2 Trefoil CEL2 column with a mobile phase of CO2 and *** injection volume was 2 μL and run-time was 6 min with the mobile phase running at 2.0 mL/min flow *** temperature,auto back pressure regulator pressure(ABPR),and modifier solvent were optimized for separation *** the optimal conditio ns,the recoveries for all the ena ntiomers were 81.3%-94.8% with relative standard deviations(RSD)less than 5.0% at 0.1,1.0 and 2.0 mg/kg levels,*** coefficients of determination(R^2≥0.993 7)were achieved for all the studied enantiomers in the tobacco over the concentration range of 10-250 ng/*** limits of detection(LODs)varied from 0.02 mg/kg to 0.04 mg/kg,and the limits of quantification(LOQs)were ≤ 0.13 mg/*** results confirm that the proposed method is green,convenient and reliable for the enantioselective determination of the enantiomers of quizalofop-ethyl in tobacco.

The purpose of the study was to establish an analytical method to simultaneously determine multiple sterols in tobacco leaves rapidly and *** this gas chromatography-triple quadruple tandem mass spectrometry(GC-MS/MS)based method,various conditions for sterols extraction were assessed and the instrumental operation parameters were optimized with 5α-cholane as an internal *** this method,eight plant sterols,namely lanosterol,campesterol,chenodeoxycholic acid,cholesterol,β-sitosterol,stigmasterol,dihydrotachysterol and ergosterol,were separated in less than 30 *** linear correlation coefficients were over 0.998 9,the low detection limits were in the range of 0.010 3-0.141 4 μg/*** recoveries were from 87.30% to 115.60%,with low standard *** conclusion,this method demonstrates good repeatability,accuracy,and high sensitivity,and is best suited for rapid analysis of multiple sterols in tobacco leaves.

Objective To investigate the changes in mass spectrometryof proteins in patients with 1-bromopropane(1-BP) poisoning after treatment and their biologicalfunctions. Methods From May 2016 to December 2017,3 male patients aged 31-47 years with 1-BP poisoning inBao'an District of Shenzhen,China,were enrolled in thisstudy. The whole blood sample (2 ml) was collected beforeand after treatment. Label-free mass spectrometrybasedproteomics was used for protein identification andquantification. The differentially expressed proteins aftertreatment were analyzed. Bioinformatics tools were usedto analyze the functions of the identified proteins and thebiological processes involved in. Results Proteomic analysisshowed that there were 47 proteins that were differentiallyexpressed more than 2-fold ( P < 0. 05) aftertreatment in the patients with 1-BP poisoning;27 wereup-regulated and 20 were down-regulated in the serum oftreated patients.

[Objectives] To study the chemical components and relative content of Achyranthes bidentata leaves and provide a scientific basis for further development and utilization of A. bidentata leaves.[Methods] The chemical components of A. bidentata leaves were rapidly analyzed using the ultra high performance liquid chromatography-time of flight-high resolution mass spectrometry (UHPLC-TOF-MS).[Results] Thirty eight chemical compounds were identified in samples of A. bidentata leaves collected from Wen County of Henan Province, in which seven chemical compounds had the relative content higher than 5%, linoleic acid reached 25.7% and inokosterone A reached 13.8%.[Conclusions] A. bidentata leaves contain many kinds of chemical compounds. This study is expected to provide a certain basis for further extraction of linoleic acid and inokosterone A.
