Genome-wide identification,characterization and functional prediction of the SRS gene family in sesame(Sesamum indicum L.)

Sesame(Sesamum indicum L.)is an ancient oilseed crop of the Pedaliaceae family with high oil content and potential health benefits.shi RELATED SEQUENCE(SRS)proteins are the transcription factors(TFs)specific to plants that contain RING-like zinc finger domain and are associated with the regulation of several physiological and biochemical processes.They also play vital roles in plant growth and development such as root formation,leaf development,floral development,hormone biosynthesis,signal transduction,and biotic and abiotic stress responses.Nevertheless,the SRS gene family was not reported in sesame yet.In this study,identification,molecular characterization,phylogenetic relationship,cis-acting regulatory elements,protein-protein interaction,syntenic relationship,duplication events and expression pattern of SRS genes were analyzed in S.indicum.We identified total six SiSRS genes on seven different linkage groups in the S.indicum genome by comparing with the other species,including the model plant Arabidopsis thaliana.The SiSRS genes showed variation in their structure like2–5 exons and 1–4 introns.Like other species,SiSRS proteins also contained‘RING-like zinc finger'and‘LRP1'domains.Then,the SiSRS genes were clustered into subclasses via phylogenetic analysis with proteins of S.indicum,A.thaliana,and some other plant species.The cis-acting regulatory elements analysis revealed that the promoter region of SiSRS4(SIN_1011561)showed the highest 13 and 16 elements for light-and phytohormone-responses whereas,SiSRS1(SIN_1015187)showed the highest 15 elements for stress-response.The ABREs,or ABA-responsive elements,were found in a maximum of 8 copies in the SiSRS3(SIN 1009100).Moreover,the available RNA-seq based expression of SiSRS genes revealed variation in expression patterns between stress-treated and non-treated samples,especially in drought and salinity conditions in.S.indicum.Two SiSRS genes like SiSRS1(SIN_1015187)and SiSRS5(SIN_1021065),also exhibited variable expression patterns between control vs PEG-tre

Neutron activation analysis of sources of raw material of Emperor Qin shi Huang's Terracotta Warriors and Horses

There have been selected 83 samples of terracotta warriors and horses of Emperor Qin shi Huang's Mausoleum, 20 samples of clays taken from around Qin's Mausoleum and 2 samples of Yaozhou porcelain bodies. All these samples have been measured by instrument neutron activation analysis (INAA) and as many as 32 kinds of element contents of each sample are measured. The following conclusion has been reached when fuzzy cluster analysis is conducted to element contents of all these samples: (i) The samples are roughly classified into five categories: namely, samples from pits No. 1 and No. 2; samples from pit No. 3; loam layers; the mixture of loam and loess; and Yaozhou porcelain bodies. (ii) The terracottawarriors and horses in pits No. 1, No. 2 and No. 3 are relatively independent from one another. The clays from which they were made are not entirely identical.We have found that samples in pit No. 3 are very closely related and their claysources are comparatively concentrated. Samples in pits No. 1 and No. 2 are less related and their clay sources are comparatively scattered. (iii) The clays from which the terracotta warriors and horses were made are closely related to theloam layer near Qin's Mausoleum, particularly to the loam layer of Zaoyuan village and Gaoxing village, but they are not so related to loess layers there, nor to the loam layers of Anhoubao, even less related to Yaozhou porcelain bodies. Arational deduction thus drawn is that the raw material of clays from which the terracotta warriors and horses were made might probably be taken from loam layers around Zaoyuan and Gaoxing, or loam layers near Qin's Mausoleum whose properties are identical with those of loam layers of Zaoyuan and Gaoxing, rather than loess layers around the above places. Since the raw material of the terracotta warriors and horses was taken from loam near Qin's Mausoleum, it could be deductedthat the kiln sites might be located in around Qin's Mausoleum.

A recent study suggests that the deep dolomites in well Xike-1 located more than 1000 m deep have a very low porosity,and no permeability for most of the samples.Given that the largest oil and gas fields in the South China Sea are all composed of biohermal dolomites while the deep dolomites of Xike-1 serve as the caprock formations rather than the reservoirs,this represents the first discovery and is of great theoretical importance.In this paper,core photos of the tight dolomites are presented.Hand specimens description and systematic porosity-permeability reveal a very low porosity,and no permeability for most of the samples.PLM,SEM and XRPD analysis discovered that the dolostones in the deep tight dolomites are quite large in size and euhedral or subhedral in shape.Alizarin red-stained sections are mostly pink in color and have a full coverage,which are considered as evidence for multiple calcite intrusions after the formation of ankerite dolomites.Biological relict textures are developed.The order degree is medium to low.The dolomites could be the product of penecontemporaneous dolomitization.

目的建立人急性单核细胞白血病(AMLM5b)细胞系并研究其生物学特性。方法从1例AMLM5b患者白血病复发时的骨髓标本分离出单个核细胞,用液体培养法进行培养。采用瑞特染色、电子显微镜、细胞化学染色、流式细胞仪、R显带核型分析、逆转录聚合酶链反应(RTPCR)、荧光原位杂交(FISH)、半固体甲基纤维素集落培养、裸小鼠致瘤实验、荧光定量PCR、DNA荧光染色法及支原体肉汤培养法、短串联重复序列(STR)PCR、p53基因的PCR扩增产物测序、多色FISH(MFISH)和3HTdR掺入实验等方法对shi1细胞的生物学特性进行了鉴定。结果建立了1个可持续增殖的人单核细胞白血病细胞系shi1;形态学和免疫表型呈现典型的单核系特征;核型分析显示shi1细胞系有和患者复发时骨髓细胞完全相同的异常46,XY,t(6;11)(q27;q23),del(17)(p11);RTPCR检出MLLAF6融合基因的转录本;FISH检测结果显示存在6号和11号染色体之间易位、MLL基因的重排和p53基因的缺失;PCR产物测序结果显示1个p53等位基因6号外显子发生点突变ATC→ACC集落培养显示shi1细胞具有较强的集落形成能力;皮下接种4只裸小鼠均形成实体肿瘤;荧光定量PCR提示无EB病毒感染;DNA荧光染色法和支原体肉汤培养法未检出支原体;MFISH证实传代至2003年3月的shi1细胞除有t(6;11)、del(17)(p11)外,?

Specific Adsorption of Trivalent La, Ce and Y by Soils and Ferro-manganese Oxides and Its Mechanism

In this paper,the adsorption-desorption variations of trivalent La,Ce,Y and mixed rare earths are discussed.The curves of pH-rare earth element adsorption were very well fitted to the equation:1nD=a+b pH.The selectivity of RE(rare earth element) ions by the samples decreased in the following order:Ce>RE>La>Y,but the sequences were:La>Ce>Y on kaolinite and Y>La on amorphous iron oxide.Since the trivalent RE ions existed in the form of RE(OH)^2+ in the solutions from pH<5.45 to 7.0,the ratio of H^+ displaced to RE^3+ adsorbed in micromole was proposed to be about 2.The specific adsorption mechanism for RE was proposed to be that the RE ions complexed with oxide surface and the ion-surface complex of Ce^3+ promoted oxidization on Mn hydroxide.

Converting Instance Checking to Subsumption: A Rethink for Object Queries over Practical Ontologies

Efficiently querying Description Logic (DL) ontologies is becoming a vital task in various data-intensive DL applications. Considered as a basic service for answering object queries over DL ontologies, instance checking can be realized by using the most specific concept (MSC) method, which converts instance checking into subsumption problems. This method, however, loses its simplicity and efficiency when applied to large and complex ontologies, as it tends to generate very large MSCs that could lead to intractable reasoning. In this paper, we propose a revision to this MSC method for DL shi , allowing it to generate much simpler and smaller concepts that are specific enough to answer a given query. With independence between computed MSCs, scalability for query answering can also be achieved by distributing and parallelizing the computations. An empirical evaluation shows the efficacy of our revised MSC method and the significant efficiency achieved when using it for answering object queries.

目的　探讨组蛋白去乙酰化酶抑制剂三丁酸甘油酯 (TB)体外诱导白血病细胞株shi 1生长抑制、分化和凋亡作用 ,并对其作用机制进行初步研究。方法　应用细胞计数、锥虫蓝拒染法观察TB对细胞的生长抑制作用 ,通过细胞形态学观察、NBT还原实验、细胞表面抗原CD11b和CD14的检测、Annexin标记、DNA凝胶电泳等方法分析TB对shi 1细胞的诱导分化及凋亡作用。通过West ernblot方法及逆转录聚合酶链反应检测TB作用前后细胞组蛋白H3乙酰化水平以及 p2 1WAF1/CIP1表达量的改变。结果　①TB可以抑制shi 1细胞的增殖及活力 ,呈剂量时间依赖关系。②TB 0 .1mmol/L可诱导shi 1细胞发生部分分化 ,NBT阳性率升高 ,细胞表面CD11b、CD14表达上调。③shi 1经TB 0 .5～ 1.0mmol/L作用 4 8h ,出现典型的凋亡形态学改变。细胞AnnexinⅤ结合力明显上升 ,并可见典型的DNA梯形条带。TB作用后shi 1细胞组蛋白H3乙酰化水平升高以及 p2 1WAF1/CIP1表达上调。结论　TB能抑制shi 1细胞的增殖并影响细胞活力 ,诱导shi 1细胞分化和凋亡 ,其机制与TB引起组蛋白乙酰化水平升高、继而上调 p2 1WAF1表达有关。

Political History of Books:A Case Study of Precedents of Ritual Protocol for Jin's Imperial Ministers and Annotations to the Report on Jin's Official Posts

This article takes Fu Chang's Precedents of Ritual Protocol for Jin's Imperial Ministers and Xun Chuo's Annotations to the Report on Jin's Official Posts as a means to observe the interactions between books and political processes during the Western and Eastern Jin dynasties. Both Fu Chang and Xun Chuo were employed as high-ranking officials, with a background of being from the Central Plain, by the regime of the Later Zhao founded by shi Le. Their purpose for writing the two books was to provide guidance for the regime's institutional establishment. The collapse of shi Le's regime led to the flow of people and their possessions to the south. During these movements, the two books along with other materials and records were brought to Jiankang and then contributed to the institutional and cultural development in the middle and late Eastern Jin Dynasty. As historical facts and references for political reformation, these books revealed the duality of institutional writings. The collection and dispersion of books after the Disaster of Yongjia, as well as the social and cultural changes, should be placed in a wider political process for further examination.
