The bactericidal spectrum and virucidal effects of silver nanoparticles against the pathogens in sericulture

Silver nanoparticles have mainly been studied for their antimicrobial potential against bacteria, but have also proven to be active against several types of viruses, even against pebrine as well. Methods: Petri dishes and transmission electronic microscope were applied to observe silver nanoparticles against bacteria strains and nuclear polyhedral viruses (NPV) and cytoplasmic polyhedral viruses (CPV). For biological test, 108 CFU/mL NPV with strong pathogenicity, and the NPV samples treated with 1:100 ratio of “Sumerian Silver” for 8 hrs were sprayed on the mulberry leaves, respectively, for silkworm rearing. Conclusion: Silver nanoparticle solution showed a strong bactericidal effect against both G+ and G﹣bacterial pathogenic strains from Bombyx mori and mulberry. Under transmission electronic microscope (TEM), Bacillus sp. Samples showed light particles in the cells when treated with silver nanoparticles, in contrast, untreated samples showed homogeneity contents. Further, NPV particles showed no significant differences between treated and untreated samples, but CPV showed strong effects that almost all CPV were collapsed. For biological test, “Sumerian Silver” treated NPV showed no diseased silkworm but nearly all silkworms were dead with no treatment. It seems like that silver nanoparticles were proved to be more effective against CPV than that of NPV. The significant differences between two antivirus or virucidal mechanism should be greatly aroused the scientific interest.

With many years of benign development,Dongtai City has established a complete technical system for the sericulture industry,an advanced industrialized operation mode,a mature operation mechanism of trade,industry and agriculture,and a perfect social service *** production level of sericulture in Dongtai City is constantly improved. However,there are still problems such as small scale of individual household,slow technological progress,low level of industrial mechanization and labor saving,and hidden dangers in safe production of sericulture. It is recommended to fully build mulberry gardens and lay a solid foundation for the sericulture industry; implement compound operations to increase the level of income growth; deepen three major innovations,break through the bottlenecks; expand industrial functions,promote integrated development,and accelerate the high quality development of the sericulture industry in Dongtai City.

Mulberry(Morns spp.)is the sole plant consumed by the domesticated ***,the genome of domesticated mulberry has not yet been sequenced,and the ploidy level of this species remains ***,we report a high-quality,chromosome-level domesticated mulberry(Morus alba)*** of genomic data and karyotype analyses confirmed that *** is a diploid with 28 chromosomes(2/7=2x=28).Population genomic analysis based on resequencing of 134 mulberry accessions classified domesticated mulberry into three geographical groups,namely,Taihu Basin of southeastern China(Hu mulberry),northern and southwestern China,and *** mulberry had the lowest nucleotide diversity among these accessions and demonstrated obvious signatures of selection associated with environmental *** phylogenetic analysis supports a previous proposal that multiple domesticated mulberry accessions previously classified as different species actually belong to one *** study expands our understanding of genome evolution of the genus Morus and population structure of domesticated mulberry,which would facilitate mulberry breeding and improvement.

Detection of Contamination and Analysis of Vertical Transmission of BmNPV in Eggs and Moths of Bombyx mori

This study reports the molecular detection of Bombyx mori nucleopolyhedrovirus (BmNPV) in silkworm strains of the Universidade Estadual de Maringá Brazilian Germplasm Bank (UBGB). DNA extraction was carried out by using six Bombyx mori female moths of each strain, followed by PCR amplification. A pair of primers was designed based on a specific sequence of the baculovirus genome related to the BmNPV ORF 14. Another pair of primers was used to amplify the silkworm Actin A3 gene segment, which was used as positive control. Twenty gene pools were analyzed, and fifteen revealed a fragment of 443 base pairs (bp), which indicated the presence of the BmNPV. The frequency of contaminated moths was as following: 100% for silkworm strains M18-2, M12-2 and J1;83% for C25, C75 and C24 strains;66% for KR01;50% for M11-A;33% for AS3, B106, M8 and M11 and 16% for C211, E8 and Hindu strains. These are promising results for the identification of contaminated B. mori moths by BmNPV, which may prevent virus proliferation in subsequent generations. We also analyzed DNA samples extracted from B. mori eggs, but the results were not conclusive regarding the detection of the fragments of the expected size (443 bp). The difficulty in detecting BmNPV contamination in B. mori eggs may be due to the low concentration of virus in samples.
