Bacterial blight, caused by Xanthomonas oryzae pv. oryzae(Xoo), is one of the most destructive diseases of rice(Oryza sativa L.) worldwide. The type III secretion system(T3SS) of Xoo, encoded by the hrp(hypersensitive response and pathogenicity) genes, plays critical roles in conferring pathogenicity in host rice and triggering a hypersensitive response(HR) in non-host plants. To investigate the major genes conferring the pathogenicity and avirulence of Xoo, we previously constructed a random Tn5-insertion mutant library of Xoo strain PXO99A. We report here the isolation and characterization of a Tn5-insertion mutant PXM69. Tn5-insertion mutants were screened on indica rice JG30, which is highly susceptible to PXO99A, by leaf-cutting inoculation.Four mutants with reduced virulence were obtained after two rounds of screening. Among them, the mutant PXM69 had completely lost virulence to the rice host and ability to elicit HR in non-host tobacco. Southern blotting analysis showed a single copy of a Tn5-insertion in the genome of PXM69. PCR walking and sequencing analysis revealed that the Tn5 transposon was inserted at nucleotide position 70,192–70,201 in the genome of PXO99A, disrupting the type III hrc(hrp-conserved) gene hrcQ, the first gene in the D operon of the hrp cluster in Xoo. To confirm the relationship between the Tn5-insertion and the avirulence phenotype of PXM69, we used the marker exchange mutagenesis to create a PXO99Amutant, ΔhrcQ::KAN, in which the hrcQ was disrupted by a kanamycin-encoding gene cassette at the same site as that of the Tn5-insertion. ΔhrcQ::KAN showed the same phenotype as mutant PXM69. Reintroduction of the wild-type hrcQ gene partially complemented the pathogenic function of PXM69. RT-PCR and cellulase secretion assays showed that the Tn5-disruption of hrcQ did not affect transcription of downstream genes in the D operon and function of the type II secretion system. Our results provide new insights into the pathogenic functions of clustered hrp genes in Xoo.

The stable isotopic δ18O and δ13C composition of foraminiferal shell calcite varies as a function of many factors including temperature and salinity. In order to understand and interpret the variations in the isotopic composition of foraminiferal shell calcite, research has been recently focused on the role of the "vital effects’. Our examination of the lamella structure of several recent planktonic foraminifera indicates that the secretion of sequential lamellae results in multiple lamillae on earlier chambers and a single lamella on the final chamber. We used a very simple procedure to separate the individual whole test of foraminifera into several chambers and measured the isotopic composition of each growth stage chamber. The results indicate that the stable isotopic composition (carbon and oxy -gen), particukrly that of the last two chambers, of the foraminiferal test varies as a function of the individual growing process.

Oviduct is not merely conduit which join the uterus and ovaries,but the venue of fertilization,sperm capacitation and sperm storage.Sperm storage observed in many animal species ranges from few hours to several days.Turtle are known to store sperm for period upto year or more.We examined the ultrastructure of oviduct epithelium and glandular secretions in Pelodiscusis sinensis.The storage of spermatozoa was restricted to isthmus,uterus and vagina.Epithelium of oviduct was lined with ciliated,secretory and third type of cells.The third type of cells was observed with abundant mitochondria,vacuoles and narrow nuclei.Ultrastructuraly the two basic cells were found which spanning the entire epithelia.The predominant were secretory cells while the others were ciliated.The prominent characteristic of ciliated cells was the presence of secretory vacuole released in the lumen in apocrine fashion.Secretory cells have typical biphasic granules with dark and lucent appearance.In addition,gland cells show prominent feature including the presence of different shape,size and electron density granules.The synthesis of these granules are described firstly present study,Apperantly mitochondria appear to play a role in dense granule formation.These granules are displaced toward the apical cytoplasm when the material of the less dense portion is added more concentric layers for final maturation with the assistance of the rough endoplasmic reticulum and microfilaments.The composite granules are released into the lumen by exocytosis when the less dense portion merges with the luminal content.The substance leaching out of from epithelium as well as glands may contribute to keeping the sperm quiescent and alive during storage in the female reproductive tract.

Objective:Physcion,an active anthraquinone constituent isolated from the rhubarb,a traditional Chinese herbal medicine which is widely used in clinical treatment,has cardiovascular protective properties.However,it remains unclear whether the cardiovascular protective actions of physcion are related to an activation of cardiac natriuretic hormone secretion.The purpose of the present study was to explore the effect of physcion on the secretion of atrial natriuretic peptide（ANP）,a first member of the family of cardiac natriuretic hormones,and its mechanisms involved.Methods:Experiments were performed in perfused beating rabbit left atria.The changes in ANP were measured by a specific radioimmunoassay.The changes in atrial mechanical dynamics,atrial stroke volume and pulse pressure,were chased during the experiments.Results:Physcion increased ANP secretion concomitantly with a decrease in atrial pulse pressure and stroke volume in a concentration-dependent manner.Physcion-induced changes in ANP secretion and atrial dynamics were attenuated by inhibition of L-type Ca2+channels with nifedipine.Atropine,methoctramine and tertiapin-Q had no significant effect on the Physcion-induced changes in ANP secretion and mechanical dynamics.Conclusion:These findings indicate that physcion increases ANP secretion via inhibition of L-type Ca2+channels.The results also provide a rationale for the use of emodin in the treatment of impairment of the regulation of the cardiovascular homeostasis.

Brain-derived neurotrophic factor(BDNF) attracts increasing attention from both research and clinical fields because of its important functions in the central nervous system. An adequate amount of BDNF is critical to develop and maintain normal neuronal circuits in the brain. Given that loss of BDNF function has beenreported in the brains of patients with neurodegenerative or psychiatric diseases, understanding basic properties of BDNF and associated intracellular processes is imperative. In this review, we revisit the gene structure, transcription, translation, transport and secretion mechanisms of BDNF. We also introduce implications of BDNF in several brain-related diseases including Alzheimer's disease, Huntington's disease, depression and schizophrenia.

＜正＞Objective Brain-derived neurotrophic factor（BDNF） and its receptor,tropomyosin-related kinase receptor B（TrkB）,play a critical role in activity-dependent plasticity processes such as long-term potentiation,learning, and memory.It has been shown that BDNF exerts different or even opposite effects on behavior depending on the neural circuit.However,the detailed role of BDNF in memory process on the basis of its location has not been fully understood.Here,we aim to investigate the regional specific involvement of BDNF/TrkB in hippocampal-independent conditioned taste aversion（CTA） memory processes.Methods Real time PCR and ELISA were respectively used to detect the BDNF mRNA and protein changes at various time points after CTA learning.TrkB phosphorylation levels normalized to total TrkB were measured by immunoprecipitation and immunoblotting.Microinjection was used to detect role of endogenous BDNF on CTA memory formation.Results We found region-specific changes in BDNF expression during CTA learning.CTA conditioning induced increased BDNF levels in the central nuclei of amygdala （CeA） and insular cortex,but not in the basolateral amygdala（BLA） and ventromedial prefrontal cortex.Interestingly, we found that the enhanced TrkB phosphorylation occurred at the time point before the increased BDNF expression, suggesting rapid induction of activity-dependent BDNF secretion by CTA learning.Moreover,targeted infusion of BDNF antibodies or BDNF antisense oligonucleotides revealed that activity-dependent BDNF secretion and synthesis in the CeA,but not the BLA,was respectively involved in the short- and long-term memory formation of CTA.Finally, we found that infusion of exogenous BDNF into the CeA could enhance CTA learning.Conclusion These data suggest that region-specific BDNF release and synthesis temporally regulate different CTA memory phases through activation of TrkB receptors.
