Objectives To examine sequence variation in the ribosomal 18 S gene among taenia multiceps isolates from Hunan Province. Methods The 18 S gene was amplified from individual taenia multiceps isolate, and the 18 S sequences obtained were aligned using the ClustalX 1.81. Results The results showed that the lengths of 18 S sequences were 1 080 bp. The constructed phylogenetic tree revealed that the Hunan isolates and the Taenia multiceps available in Gen Bank were clustered in the same clade. Conclusion It is concluded that 18 S gene can be used as a genetic marker for population genetic studies of cestodes. The present study provided foundation for molecular epidemiology and diagnosis of taenia multiceps.

Methods The ITS gene was amplified from each sparganum isolate by PCR and sequenced, and the ITS sequences obtained were aligned using the Clustal X 2.0. Results The results showed that the lengths of ITS sequences were between 1362 kb and 1382 bp. The similarities of the corresponding sequences between isolates from Hunan province were more than 95.4(ITS-1)and 94.6%(ITS-2), while that between Hunan isolates and other cestode strains were lower than 59.5%(ITS-1)and 56.9%(ITS-2). The constructed phylogenetic tree revealed that the Hunan isolates and Spi-rometra erinaceieuropaei available in Gen Bank were clustered in the same clade. Conclusion It is suggested that ITS gene can be used as a genetic marker for species genetic variation investigation of frog sparganum. The results of the present study provided a useful basis for further studies of molecular epidemiology of frog sparga-num,and diagnosis of the resultant disease.
